Immune tolerance has an antigen-directed specificity analogous to that of immune responsiveness. Studies have shown the direction of response—immunization or totelization—is often dependent on the physical form and the dose of antigen, and that the two major lymphocyte classes, B cells and T cells, have different tolerance requirements. The critical relationships of these findings to autoimmunity are explored.
