For genetic diagnosis of conditions in which the affected gene cannot be identified, we must fall back on the technique of linkage—but with a difference. Work with restriction enzymes has produced a near-revolution in clinical genetics; it has enormously increased the number of traits accessible to evaluation by linkage with identifiable DNA polymorphisms.
