Recent studies in an animal model of autoimmune disease, experimental allergic encephalomyelitis, have provided clues about possible mechanisms governing the production and control of human demyelinating disease, particularly multiple sclerosis. The findings are reviewed, and their relevance to concepts about the events involved in the induction and loss of immunologic tolerance is emphasized.
