Ultraviolet Microscopy Distinguishes the Insoluble Polysaccharides from the Non-Polysaccharides in Periodic Acid-Schiff-Colored Biological Tissues

摘要:

In bright field microscopy of plant material, the cytoplasm, nucleoli and insoluble polysaccharides are colored magenta by the periodic acid-Schiff reaction. Blue-violet light causes an obvious distinction between the polysaccharide and non-polysaccharide tissue components by producing a yellow fluorescence of the cytoplasm and nucleoli, while the polysaccharides appear red.
