Levels of Interleukin 2 (IL2), IL 2 receptors (IL2R) and free radical scavengers were measured in 25 Graves' disease patients prior to and following an 8 week course of Carbimazole therapy in an attempt to understand the mechanism by which the drug acts on the immune system.In untreated Graves' patients IL2R levels were elevated and IL2 levels reduced. Free radical scavengers were also reduced in these patients. Following treatment IL2R levels fell and IL2 levels rose. Levels of the free radical scavengers also rose. It is not clear whether it is the fall in IL2R levels or the rise in the level of free radical scavengers (implying a reduction in free radical activity) which is responsible for the rise in IL2.

In order to clarify the role of natural cytotoxicity (NC) in the damage of epidermal keratinocytes (EK) in pemphigus vulgaris (PV) we used the direct51Cr-release assay in studying the cytotoxic activity of large grandular lymphocytes (LGL), obtained from 34 acute PV patients and 19 healthy donors against EK of PV patients, donors, intact newborn BALB/c mice and mice with experimental PV, shedding and non-shedding grass snakes. We also investigated the effect of pemphigus antibodies and shedding snake serum protein (SSSP) upon EK sensititivity to cytotoxic effects of NC effectors. In other experiments, studied were the amount of serine proteinases secreted by donor LGL in the presence of membrane antigen of antibody-or SSSP-transformed EK, and direct effect of pemphigus antibodies and SSSP upon the release of radioactive label and endoproteinases from EK. It was found that EK, which had interacted with pemphigus antibodies or SSSP, became a sensitive target for NC effectors. Membrane antigen of antibody-and SSSP-transformed human, murine and snake EK induces the secretion of endproteinases by NC effector cells. The conclusion was made that the effect of both pemphigus antibodies and SSSP on EK resulted in the emergences of a new antigen on EK cellular membrane which is recognized by NC effectors and plays the role of the ligand in receptor/ligand interaction between LGL and their targets.

We investigated the pathogenesis of mesangial proliferative lupus nephritis in NZB/W mice under conditions that allowed us to examine removal of immune complexes from the circulation, uptake by the mononuclear phagocyte system, and localization in kidney tissue. These studies were performed at a time when variables such as the quantity of endogenous immune complexes, complement concentration, and carrier state of blood cells (platelets) were controlled. NZB/W mice and C57BL/6 (control) mice showed comparable kinetics for removal of a subsaturating dose of immune complexes (2.5 mg bovine serum albumin-antibovine serum albumin) from the circulation; additionally, the liver uptake and kidney localization of these immune complexes were comparable between NZB/W and control mice. The localization of immune complexes in the glomerular mesangium of NZB/W mice could not be attributed to enhanced production of endogenous immune complexes, to decreased removal of immune complexes from the circulation, to impaired uptake by the liver, or to complement concentration and carrier state of blood cells. It appears, by exclusion, that mesangial deposits of immunoreactants in early lupus nephritis may result from interaction of antibodies with antigens in mesangia.

The distribution of T cells, B cells and murine senile amyloid protein in Peyer's Patches was examined in senescence accelerated (SAM-P/1) and control (SAM-R/I) mice ranging in age from two to ten months. An immunohistochemical detection of lymphocyte surface antigens of T cells and B cells. A murine senile amyloid protein in Peyer's patches was detected by immunohistochemical staining with the specific antiserum, Congo red staining and electron microscopy. The T cell population increased and B cell population decreased slightly with age in SAM-P/1 mice, but little change was observed with age in SAM-R/i mice. Murine senile amyloid protein positive spots were seen surrounding the vessels of the thymus-dependenl area at 6 months of age, and were observed throughout the Peyer's patches at 10 months of age, but were not observed by 10 months of age in SAM-R/1 mice.

The effects of two novel immunosuppressants, FK506 and 15-deoxyspergualin (15-DSG), on experimental autoimmune uveoretinitis (EAU) were evaluated in the rat. Rats were immunized with retinal soluble antigen (S-antigen) and treated with FK506 or 15-DSG, and the disease induction as well as the immune responses to the antigen were examined. The results were compared with animals treated with cyclosporine (CsA) which has been widely used to treat refractory uveitis in humans. A dose-response study showed that FK506 suppressed EAU induction at doses 10–30 times lower than CsA when given on days 0–14 postimmunization, while 15-DSG suppressed the disease development at doses very similar to CsA. As with CsA, FK506 suppressed EAU induction when given only in the induction phase (days 0–5 postimmunization) or in the effector phase (days 7–12), but at doses much lower than CsA. On the other hand, 15-DSG suppressed EAU only at toxic doses with these schedules. The antigen specific mitotic response of lymphocytes was markedly suppressed by the three agents, while the antigen specific antibodies in sera were suppressed by 15-DSG and FK506 but not by CsA at doses effective in suppressing the disease induction. More significant findings were uniquely prolonged immunosuppressive effects of FK506: EAU induction as well as the immune responses to S-antigen were suppressed long after the cessation of drug treatment.

In order to evaluate two new immunosuppressive agents, FR900506 (FK 506) and 15-Deoxyspergualine (15-DSG), the kinetics of infiltrating cells in the eyes of Lewis rats with experimental autoimmune uveoretinitis (EAU) were studied. Rats were immunized with retinal S-antigen and treated with different doses of either FK 506 or 15-DSG. The inflammatory ocular tissues obtained at various intervals during the process of EAU were examined using an immunoperoxidase technique. The results were compared with those eyes developing EAU without treatment or with suboptimal doses or a suboptimal dose of Cyclosporin (CsA). BothFK 506 and 15-DSG, like CsA, delayed the cellular kinetics during the course of EAU. However. FK 506 had the greatest effect on the kinetics of T lymphocyte subsets by causing the greatest increase in the recruitment time of the T suppressor/cytotoxic population. FK506 treatment resulted not only in the highest inhibition of expression of IL-2 receptors on T cells, but also in the prevention of the expression of MHC class II antigens on ocular resident cells. Treatment with 15-DSG resulted in general immunosuppression on various infiltrating inflammatory cells.

The autoimmune response to mouse accessory glands (MAG) was investigated in male BALB/c mice immunized with different doses of chemically modified mouse accessory glands (MMAG) and complete Freund's adjuvant (CFA). This autoimmune response was studied at several time intervals using the skin test with MAG. It was found that 5 mg of MMAG induced on the day 15 an autoimmune response detected by specific skin test at 20min., 3h and 24 h. The results of the immediate type hypersensitivity (ITH) were higher than those with the other skin tests. In order to study the type of immunoglobulin involved, the ITH was also analyzed by passive cutaneous anaphylaxis (PCA) at different time intervals with treated and untreated sera at 56°C. The findings suggest the presence of reaginic antibodies, IgE being the major antibody as detected by enzime linked immunosorbent asay (ELISA).The MAG was subsequently fractionated using Sephandex G-100 and the fractions thus obtained (FI. FII and Fill) were used to challenge mice immunized with MMAG. It was found that FI was the only fraction which revealed an ITH similar to that revealed by MAGThe effect of infection withTrypanosoma cruzion the autoimmune response to MAG was analyzed with different mouse groups intraperitoneally treated with 2×103blood trypomastigotes/animal at several time intervals: namely, on days−5, 0, +5 and + 10 with respect to the immunization with MMAG. The autoimmune response to MAG showed suppression when the animals received the parasites on the same day as the autoantigen.

Using a combination of avidin-biotin microELISA and solid phase radioimmunoassay, we examined sera from 23 patients with systemic lupus erythematosus (SLE), two patients with established sensitivity to ingested shrimp, and 15 healthy normal subjects. In addition to IgG antibodies, varying amounts of IgE antibodies specific for native DNA (nDNA), denatured or single-stranded DNA (dnDNA), RNA, and tRNA were demonstrable in the sera of SLE patients, but not in the sera of normal subjects. A comparison of the specificity of nucleic acid-specific IgE antibodies present in the sera of shrimp-sensitive patients with those present in the sera of seven SLE patients revealed that the IgE antibodies in the sera of shrimp-sensitive patients specifically recognized shrimp tRNA but not yeast tRNA, calf thymus RNA, or calf thymus DNA, while those present in the sera of patients with SLE recognized all these nucleic acid antigens. The IgE antibodies directed against nDNA, dnDNA, RNA, and tRNA may mediate mast cell and basophil degranulation and thus contribute both to immediate-type hypersensitivity phenomena including hives seen in patients with SLE and to the localization of IgE-nucleic acid complexes in target tissues.

In a previous work we characterized a major epitope of thyroid peroxidase (TPO). which was recognised by 66% of 157 patients with autoimmune thyroid disease (AITD) and 9 out of 50 patients with non-thyroi-dal autoimmune disease (NTAID) 6 of whom had antibodies to the gastric parietal cell antigen (PCA).In the present study we have affinity purified C2 antibodies and demonstrated that they bind to the native TPO enzyme in a radioimmunoassay (RIA).We have measured antibodies to the C2 peptide and a second TPO peptide, C21, in enzyme-linked immunosorbent assays (ELISA) in 30 patients with NTAID all of whom have antibodies to the gastric PCA, having first determined the incidence of antibodies to C21 in 98 patients with AITD who do not have antibodies to the gastric PCA. 58% of patients with AITD have antibodies to C21, a peptide of TPO which overlaps C2 by 21 amino acids in the region containing an 11 residue fragment which is very similar to a fragment of the H+K+ATPase, which has recently been shown to be a major component of the gastric PCA.In patients having NTAID and antibodies to the gastric PCA, 60% are positive for C2 Ab and 100% for C21 Ab, which is suggestive of an epitope shared by TPO and H+K+ATPase, corresponding to TPO 659→669 and H+ K+ATPase 177–187.We conclude that C2 antibodies are heterogenous and comprise activities which bind to the intact enzyme and activities binding to an epitope which may be common to TPO and H+K+ATPase.
