MRL mice homozygous for the recessive1prgene develop an accelerated autoimmune syndrome and massive lymphadenopathy. Because the function of the expanded lymph node population is unclear, we have studied the subunits of the T cell receptor for antigen (TcR). DNA and RNA were prepared from MRL/Mp-1pr/1pr (1pr)and congenic MRL/Mp- +/+ (+/+) mice by standard techniques and studied by Southern blot, northern blot, and dot blot analysis using the cDNAs TT11, specific for the TcRαchain; 86T5, specific for the TcRβchain; and T3δ; specific for the subunit of the T3 molecule. Surface protein was immunoprecipitated with antisera 8177, which recognizes TcR framework determinants, and resolved by diagonal SDS-PAGE. FACS analysis was performed with a monoclonal antibody to murine T3, and with the KJ16-133 and F23.1 monoclonal antibodies, which recognize determinants encoded by the Vβ8 subfamily ofβchain variable region genes.When compared with +/+ controls, surface TcR density as detected by immunofluorescence using all three antibodies was significantly diminished on1prspleen and lymph node cells, as well as on1prlymph node cells which had been depleted of L3T4+and Ly2+cells by negative selection. There appeared, however, to be selective expression of the genes encoding the epitopes binding F23.1. Southern blot analysis of DNA showed polyclonal rearrangements of the TcRβchain genes. There were increasedα,β, and T3δRNA transcripts in the double negative lymph node cells. The paradoxical decrease in TcR surface expression in the setting of large quantities of full length transcript is yet to be explained.

In a large, representative sample of newly-diagnosed IDDM patients, using a highly sensitive assay to detect islet cell cytoplasmic antibodies (ICA), no marked differences were found between ICA + and ICA - patients on various clinical, genetic, immunologic, and epidemiologic characteristics. In particular, there was no evidence for associations between ICA status at diagnosis and either sex, race, family history of IDDM, HLA-DR phenotype, antibody titers to Coxsackie B viruses, immunoglobulin levels, C-peptide and glycosylated hemoglobin concentrations, or insulin requirements. The most significant relationship was between the presence of ICA and a young age at diagnosis; however, the large overlap between the distributions of the ages at onset for ICA + and ICA - groups on this variable suggests that this association is of limited importance. These data suggest that the presence or absence of ICA at diagnosis may not be useful in defining possible subtypes of IDDM.

The source of IgG in the cerebrospinal fluid (CSF) in guinea pigs with chronic relapsing experimental allergic encephalomyelitis (CR-EAE) was investigated using quotient analysis of total IgG and albumin concentrations and by computing CSF-plasma ratios of specific IgG concentrations. Increased blood-CSF barrier (B-CSFB) permeability was shown by elevated albumin quotients in both relapse and remission phases of CR-EAE and intrathecal production of IgG was indicated by raised ratios of IgG to albumin in the CSF. Intrathecal IgG synthesis was greatest in guinea pigs which had little B-CSFB damage.When enzyme-linked immunosorbent assays (ELISA) for whole cord, myelin basic protein (MBP) orMycobacterium tuberculosiswere performed with CSF and plasma adjusted to the same concentration of total IgG, the CSF/plasma ratios of ELISA results for specific antibodies were less then unity and ratios for whole cord and MBP were lower than those forM. tuberculosis.There was thus no evidence for a selective increase in the CSF of antibody specific either for the neuroantigens tested or for adjuvant components. The CSF-plasma ratios for each specific antibody were inversely correlated with the extent of total IgG intrathecal synthesis, suggesting that much of the antibody production within the CNS is the result of polyclonal B cell activation.

The nature of the immunosuppressive effect of antithyroid drugs has been a subject of controversy. It has been claimed that these agents exert a direct effect on the immune system, although we and others have suggested that the drugs affect the thyroid cells primarily with consequent reduced thyrocyte-immunocyte signalling. This may occur from reduced thyroid hormone production and/or reduced antigen presentation by the thyrocytes to local T lymphocytes.Using a cytotoxicity assay system, with chromium-51 labelling, monoclonal antibodies against thyroperoxidase (TPO) and HLA-DR, and complement, we have measured the expression of TPO and HLA-DR on cultured normal human thyroid cells; we have also measured thyroglobulin (Tg) release by radioimmunoassay into the medium of the cultured cells. The thyroid cells were stimulated with TSH or thyrotropin binding inhibitory immunoglobulin (TBII) for 48 hours before measuring for TPO induction, and with interferon gamma (IFN-γ) (with or without TSH or TBII) for thyrocyte HLA-DR expression.A dosage of 1.6 milliunits per ml of TSH resulted in a significant increase in TPO expression on thyrocytes when compared with control unstimulated thyroid cells (p<0.001). The concentrations of Tg released into the medium with TSH or TBII were also significantly higher than those of the control thyrocytes. IFN-γat 200 units per ml induced HLA-DR expression, but did not induce thyrocyte TPO expression, or Tg release. Addition of the antithyroid drug, methimazole (MMI), at different concentrations, in addition to the other stimulators, IFN-γ, TSH, or TBII, did not result in any inhibition of TPO, Tg release, or HLA-DR expression on the thyroid cells. It would thus appear that the pathways for stimulation for the expression of TPO and HLA-DR appear to be different. Finally, MMI does not cause its immunosuppressive effect by any reduction of thyroid antigen expression or release.

Bullous pemphigoid (BP) and cicatricial pemphigoid (CP) are subepidermal bullous autoimmune diseases which have distinct clinical features but identical immunological status. In order to determine whether these diseases could be dissociated on the basis of qualitative differences in serum antibodies to basement membrane zone (BMZ) antigens, the reactivity of sera from 7 CP and 29 BP patients with proteins extracted from normal human epidermal sheets (containing most of the lamina lucida components) was analysed using immunoblotting and compared to that of 10 normal sera.20 out of the 29 BP sera contained antibodies recognizing one or several protein(s) of 240, 200, 180 and 165kD molecular weight (MW). Antibodies in 4 out 7CP sera specifically reacted with one or two polypeptides of 240 and 120kDMW. These data confirm the heterogeneity of BP antigens and show the presence in CP of a novel 120kDMW polypeptide which is found only in CP but not in BP.Taken together these findings demonstrate that in BP and CP, autoantibodies are directed to both common and specific BMZ antgiens, their physiopathological significance need to be understood.

The nature of the minute amounts of cryoprecipitable proteins present in all normal sera has been investigated in particular relation to the physicochemical and functional properties of the precipitated immunoglobulins.Cryoprecipitated material from 48 normal donor sera were first analyzed for their protein, IgG and IgM content. SDS-PAGE analysis revealed many bands in a majority of samples but in about one third a very limited number of bands was observed. IgM, IgG and albumin were identified and a lower molecular weight component (16 KD) was always present. Restricted clonality was demonstrated by IEF in 25-30% of the samples. In 50% of cryoprecipitates IgG was present in higher molecular weight fractions than 7S on sucrose density gradient at neutral pH. Ultracentrifugation at acid pH lead to a dissociation of these IgG fractions in most of the samples. A positive reaction for IgM rheumatoid factor was seen in 63% of the samples and there was a specific enrichment of RF activity in cryoprecipitates as compared to the corresponding serum. These data suggest that the cryoprecipitation of immunoglobulins in normal serum reflects specific interactions between immunoglobulin molecules rather than merely a non-specific preipita-tion of cold-insoluble molecules.

The HEp2 cell cultures appeared highly sensitive in detecting the antinuclear antibodies (ANAb) in systemic sclerosis, principally anticentromere antibodies of the CREST syndrome. The immunoblotting used with either complex cellular extracts from HeLa and rabbit thymus or purified nuclear components (high mobility group (HMG) proteins and histones) is able to identify precisely the ANAb targets and to contribute to diagnosis. With nuclear extracts of HeLa cells, the sera from 75.8% of CREST syndrome subjects stained 18 and 22 kD proteins. Corresponding antibodies were also detected in 72.7% of these patients, on HEp2 centromeres by indirect immunofluorescence. With the same extracts, 33.3% of sera from diffuse sclerosis/acrosclerosis patients contain antibodies staining 86, 73, 32 and 30kD. These sera also stain 77, 66 and 63kD from thymus extracts. Corresponding antibodies will be the anti-SCL-70 antibodies defined by double immunodiffusion. The anti-HMG antibodies were infrequent in systemic sclerosis, rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE) and consequently without interest for diagnosis. The anti-whole histones antibodies which are less frequent in diffuse sclerosis/acrosclerosis (35.7%) than in SLE (41.3%) recognize especially HI and H2A in the first diseases, HI and H2B in SLE and HI and H3 in RA.
