In the present work we studied the influence that an infection withToxoplasma gondiiin thymus proximity produces on the suppression of autoimmune response to autoantigen of rat male accessory glands (RAG). The suppression was achieved injecting syngeneic animals with low doses of autoantigen of RAG previous to the immunization with chemically modified rat male accessory glands (MRAG). Rats were infected in thymus proximity with 3×103trophozoites ofT. gondiibefore or after to be suppressed. Controls were rats only infected or only suppressed. The delayed hypersensitivity response against MRAG, (DTH test), was significantly potentiated in rats only infected and in the animals suppressed before the infection (p<0.001). The suppression was not inhibited in the animals suppressed after infection. The suppression of humoral response against MRAG studied by ELISA and passive hemagglutination test was prevented in rats infected before as well as in rats infected after the induction of suppression (p<0.001). Decrease of CD4+ CD8+ and Ox18+ (class I MHC antigen) and increase of CD4+ CD8–and CD4–CD8–thymocytes was observed in the rats where the DTH response was potentiated. These results indicate that the infection withT. gondiiin thymus proximity was able to inhibit the suppression of response to autoantigen of RAG producing selective impairment in thymic suppressor influence.

Lymphocyte subsets of the peripheral blood in 43 patients with myasthenia gravis (MG) were determined by two-color flow cytometry using a number of monoclonal antibodies.In the MG patients without thymectomy (Tx) and prednisolone (PSL) treatment, lymphocyte counts, B-cells, CD4+cells and their subsets were normal, but numbers of T-cells, CD8+cells and CD8+CD 11−subsets were significantly decreased. Furthermore, proportions of activated cells in T-cells, CD 16+Leu7–and CD16+Leu7+NK subsets were significantly high in the patients. The changes in T-cells, CD8+cells and activated T-cells were less marked in the MG patients than Sjögren's syndrome (SS) used as a disease control. Contrary to MG patients, lymphocyte counts, CD4+cells and their subsets were decreased, and the proportions of B-cells were high in SS patients. These results suggest altered immunologic conditions, immunologically active and deficient conditions, in both diseases, although the alterations were more prominent in SS than MG.PSL treatments and Tx significantly altered the lymphocyte profiles: PSL decreased lymphocytes, B-cells, T-cells, CD4+cells and their subsets, while the proportions of CD8+cells were increased. The changes were compatible with the known immunosuppressive effects of PSL. After Tx, lymphocytes and B-cells decreased, but the proportions of T-cells, CD8+cells and their subsets, and NK cells subsets returned toward normal. CD4+CD8+cells were not increased in MG patients. and the cells did not decrease after Tx. Some of these observations might be relevant to clinical effects of Tx, although the mechanism responsible for these changes is still unknown.

Sixty six infertile men with cytotoxic sperm antibodies had fewer and lower percentages of OKT4–positive helper/inducer T lymphocytesp= 0.004 and 0.003, respectively), than the fertile (n= 14) and infertile (n= 12) antibody-negative controls. Ratio of OKT4/OKT8 was decreased (p<0.01). while numbers of OKDR and BD-Leu 12 positive predominantly B lymphocytes were increased (p<0.000l) in infertile men with sperm antibodies. T lymphocytes positive for OKT3, OKT11 and OKT4 and the OKT4/OKT8 ratio were decreased in infertile women with sperm antibodies, while B lymphocytes positive for OKDR and BD-Leu12 were increased in infertile women with or without sperm antibodies (p<0.0001, versus fertile controls). Lymphocyte responses to phytohemagglutinin (PHA), concanavalin A (ConA) and pokeweed mitogen (PWM), however, were similar in 20 fertile and 136 infertile couples. Lymphocytes from infertile men with sperm antibodies had an enhanced stimulatory response to autologous sperm and seminal plasma. It is concluded that sperm antigens from autoimmune infertile men stimulate heightened immune responses to sperm antigens and altered distribution of white cell populations in both partners.

To examine whether the presence of thyrogastric autoantibodies is associated with an increased susceptibility towards developing type 1 diabetes we have tested for thyroid (microsomal and thyroglobulin) and gastric-parietal cell antibodies in 86 pairs of identical twins, 47 discordant and 39 concordant for type 1 diabetes. Autoantibodies were detected in both twins of a pair in 35 and in neither twin in 45 pairs. In only 6 pairs (3 discordant) was there a discrepancy in the antibody results between co-twins. The frequency of antibodies was similar in twin pairs discordant, (21/44,48%), and concordant, (14/36, 39%), for diabetes. Thyrogastric antibodies were not more frequent in pairs that were female, diagnosed above the age of 20, or had HLA DR3 as opposed to DR4. We conclude that thyrogastric autoantibodies are common in both type 1 diabetic patients and their non-diabetic identical twins. Their presence appears to be genetically determined but does not increase the susceptibility of developing diabetes. The presence of autoantibodies does not appear to indicate a separate aetiological type of diabetes.

Antibodies to various nuclear proteins are frequently found in sera of patients affected by connective tissue diseases and other autoimmune diseases.We investigated the specificity of circulating autoantibodies to poly(ADP-ribose)polymerase (pADPRP) in different autoimmune and connective tissue diseases: Systemic Lupus Erythematosus (SLE), Rheumatoid Arthritis (RA), Progressive Systemic Sclerosis (PSS), Sjogren's Syndrome (SS), Undifferen-tiated Connective Tissue Disease (UCTD), Cryptogenic Fibrosing Alveolitis (CFA) and Sarcoidosis.pADPRP was purified from calf thymus. Antibody specificity was detected by ELISA, western blot and enzyme activity precipitation.Positive values (mean O.D. values + 3 S.D. of 36 normal controls) were obtained in 7/15 SLE patients, 1/18 RA patients, 1/30 PSS, 3/14 SS, 0/5 UCTD, 5/21 CFA and 4/25 Sarcoidosis. The positive sera also recognized the pADPRP protein when tested by western blot. Furthermore the enzyme activity was inhibited after immunoprecipitation by some highly positive sera.

The frequency of autoantibodies to thyroid microsomes (MAb), thyroid peroxidase (TPOAb) and thyro-globulin (TgAb) was studied in 736 women during and after pregnancy. The aim was to study the relationship between TPO Ab and post-partum thyroid disease, and to compare their behaviour with MAb. Seventy-five (10%) were either MAb or TgAB positive, of whom 36 were sampled serially at 18, 30 and 36 weeks of pregnancy and 1, 6 and 12 months post partum. Twenty of the antibody negative mothers were selected at random for controls. Twelve of the 36 antibody positive mothers developed post-partum thyroid dysfunction (group I), 24 did not (group 2) compared with none of the 20 controls (group 3). Six months post partum. TPO Ab titres rose overall in both groups I and 2, but only in those with measurable TPO Ab during the first tremester. A TPO Ab was less frequently positive than MAb, but the difference was not statistically significant. TPO Ab are not superior to MAb for predicting post partum thyroid disease in pregnant women.
