摘要:
We performed bronchoalveolar lavage (BAL) in MRL-lpr/lpr (MRL/I) and MRL - +/+ (MRL/n) mice and evaluated various cellular and humoral components of the bronchoalveolar lavage fluid (BALF) to clarify the pathogenic mechanism of pulmonary fibrosis in MRL/l mouse. The numbers of macrophages, neutrophils and lymphocytes, N-Acetyl-β-glucosaminidase (β-NAG), and fibronectin increased in the BALF from MRL/l mice than that from MRL/n mice, but no significant differences were observed in total protein,β-glucuronidase, acid phosphatase, or phospholipid level. Increased fibronectin level in the BALF from MRL/l mice may be related with pathogenesis of pulmonary fibrosis.

The effect of interleukin 1 (IL-I)-like factor(s), produced by cells isolated from the synovial fluids of rheumatoid arthritis (RA) patients, on anin vitromurine model of spontaneous autoimmunity, i.e., the development of plaque-forming cells (PFC) to bromelain-treated mouse red blood cells (Br-MRBC) in mouse peritoneal cell (PC) cultures, has been investigated.It has been found that IL-I-containing culture supernatants from cells isolated from joint fluids of RA patients, as well as recombinant IL-I. determine a marked increase in anti-Br-MRBC PFC development. Moreover, factor(s) of 10-20 KD molecular weight, with IL-I -like biological activity, capable of increasing the anti-Br-MRBC PFC development in mouse PC cultures, have been demonstrated in joint fluids from RA patients.The finding that synovial inflammatory cells produce factors that activate autoreactive B cells further supports the role of autoimmunity in the pathogenesis of rheumatoid arthritis, as self-perpetuing disorder.

T cell lines and clones were derived by coculturing peripheral blood mononuclear cells from young children with newly diagnosed insulin dependent diabetes mellitus (IDDM) with sonicates of HLA-DR haploidentical human islet cells. These cells proliferated in response to human islet cell sonicates but failed to do so when stimulated with sonicates of human exocrine pancreas or thyroid gland. Preparations of islet cells obtained by repeated freezing and thawing also stimulated proliferation of the lines but purified membrane or protein preparations of the islet failed to induce proliferation, suggesting that determinants recognized by T cells were lost on further purification. This method of deriving T cell lines and clones appears to be significantly easier and quicker than non-antigen cloning using anti CD3.

We examined the ability of recombinant IL-2 to reconstitute the autologous mixed lymphocyte reaction (AMLR) defect in peripheral blood mononuclear cells (PBM) from patients with rheumatoid arthritis (RA). Our results revealed an ability to fully reconstitute RA AMLRs with pharmacologic concentrations (100 units/ml), but not physiologic concentrations (10 units/m;) of IL-2. Full reconstitution of RA AMLRs was achieved whether IL-2 was added as the initiation of culture or at 48 or 72 hours prior to termination of the cultures. Impaired IL-2 production was noted throughout the time course of the RA AMLRs. Neither an inhibitor of IL-I nor IL-2 was detected in AMLR culture supernatants. Moreover. IL-1 in pharmacologic concentrations up to 50 units/ml failed to reconstitute impaired AMLR reactivity. In 2 patients whose AMLRs failed to reconstitute fully with 100 units/ml IL-2, addition of 10 units/ml IL-I in combination with IL-2 fully reconstituted the AMLR defect. The results may suggest that defective IL-2 generation alone cannot fully account for impaired AMLR reactivity in RA patients.

We have used restriction fragment length polymorphism (RFLP) analysis to investigate the immunoglobulin constant heavy chain (IgCH) loci and the associated locus D14S1, of Caucasoids from South East England and South-West, West Germany. Haplotypes were determined using probes to the Ig heavy chain switch loci Sμand Sα1, the IgCγ3 and lgCγ2 loci as well as the D14Sl locus which is 3′of the IgCH loci. The 6.3:1.7 kilobase (kb) Bst EII Cγ3-Cγ2 haplotype was the most prevalent in the population from South-East England (frequency 0.364), whilst the 2.33.7kb Cγ3-Cγ2 haplotype was the major haplotype in the German population (frequency 0.400). With one exception, the major haplotypes of these two populations differed from the ones previously published for a Caucasoid population from California. This suggests that there may be a major ancestral IgCH haplotype which has been maintained in the population, whilst other haplotypes tend to be specific for a particular group of Caucasoids.

Pristane-induced arthritis was investigated in DBA/1, DBA/2, and BALB/c mice, and F1 hybrid mice generated from inter-crosses between these strains. The incidence of disease in FI hybrid mice was significantly lower than the susceptible parental strains (DBA/I and BALB/c). and resistance to arthritis was observed in both DBA/2 mice and the (DBA/2×BALB/c) FI hybrid mice. Several cellular immune abnormalities were observed in pristane-injected DBA/1 mice. Con A mitogen responses were depressed following pristane injection, and a functional suppressor cell population was detected. Delayed type hypersensitivity responses to type II collagen were observed in pristane injected mice. The intraperitoneal injection of pristane appears to alter immune regulation and induce autoimmune responses to connective tissue components.

Diabetes mellitus, a disorder of hyperglycemia recognized for centuries by its classical symptoms of polyuria and polydipsia, affects about 5% of the population in Western nations. Among many diabetic syndromes now recognized, insulin dependent diabetes mellitus (IDDM, Type I) and non-insulin dependent diabetes mellitus (NIDDM, Type II) are by far the most common1. IDDM accounts for≊10% of all cases of diabetes and characteristically appears in children, whereas NIDDM is primarily a disorder of obese adults. A growing consensus supports the hypothesis that IDDM is a disorder of autoimmunity2-4. Evidence supporting this view falls into several broad categories.

摘要:
The class II molecules of the human major histocompatibility complex bind intracellularly processed peptides and present them to T-helper cells. They therefore have a critical role in the initiation of the immune response. A salient feature of the class II molecules is their polymorphism. It has been shown that some autoimmune diseases are associated with certain class II alleles. This article reviews the basic structural features of class II molecules, and the genes encoding them as well as mechanisms governing the development of their extraordinary polymorphism.

The demographic imperatives of the 1990s and the coming decades point to a growing need for research on aging. Investigations of the mechanisms underlying age-related decrease in host defenses have been the focus of many clinical studies on immune function of the elderly1-3. Moreover, it is now estimated that 5% of the elderly population suffer from some form of autoimmune disease contributing to increased morbidity and, at times. to increased mortality in the aged population4-10and that 10-15% of seemingly healthy individuals over the age of 60 have a significant level of autoantibodies in their serum11,12. The incidence of autoimmunity tends to increase in the aged population as compared with race- and sex-matched young adults4,11,12. Some immunological explanations of this phenomenon may include (i) a repeated or prolonged insult by endogenous auto-antigen, or (ii) molecular mimicry with environmental antigens, or (iii) decrease in the generation of suppressor cells.
