SynopsisThe physico‐chemical and biological properties of an amphoteric/anionic system and its behaviour against a proteinic support have been thoroughly investigated. A considerable inhibition of adsorption of SLS (sodium lauryl sulphate) on human callus caused by the presence of definite amounts of AABet (alkyl‐amido‐betaines) in the treatment bath is observed. These physico‐chemical results are in agreement with those obtained by somein vivobiological tests. A mechanism of the process via the formation of mixed micelles is postulated emphasizing the stability of such systems as a function of pH, the influence of the chain length of the amphoteric surfactant and the molar relative ratio SLS/AABet. SynopsisL'activité d'un produit appliqué localement sur la peau peut, dans certains cas, se manifester par une augmentation du métabolisme cellulaire et se traduire par une augmentation de la consommation d'oxygène.Les auteurs ont utilisé deux méthodes faisant appel soit aux cellules épidermiques de rat soit aux cellules hépatiques de souris et applicables aux principes actifs et au produit fini.Les résultats portant sur extraits placentaires humains lyophilisés, liquides amniotiques humain et bovin, placenta Filatov sont exposés et les auteurs ont essayé de relier l'activitéà la présence dans chacun des extraits de facteurs pouvant être considérés comme favorables ou défavorables: taux de phosphatases alcalines, nature et pourcentage des acides aminés, présence de conservateur, de propylène glycol, de glycérol, influence du pH.Enfin, l'évaluation de l'efficacité d'un produit fini a été réalisée par application d'une préparation contenant 5% d'extrait placentaire humain lyophilisé sur 2 lots de 10 rats quotidiennement pendant 2 à 4 mois. SynopsisDans ce travail sont rapportées des données pharmacocinétiques concernant l'absorption percutanée de l'acide γ‐linolénique14C (A γ‐L) incorporéà 4% dans une base émulsionée.L'autoradiographie de coupes de peau de rats sans poils montre que A γ‐L demeure dans les structures superficielles de l'épiderme et les glandes sébacées.Sur la peau humainein vitropar la technique de ‘stripping’ et après séparation de l'épiderme et du derme, il est constaté que A γ‐L migre peu à peu dans la partie profonde du tégument (à 8 heures, 18% A γ‐L sont dans l'épiderme malpighien).En cellule de diffusionin vitrochez le rat sans poils âgé de 18 mois, l'absorption est réduite (Kp: 4.6 × 10−6cm h−1, 6 heures), elle est beaucoup plus importante chez le rat carencé en acides gras essentiels (Kp: 31 × 10−6cm h−1, 6 heures). Pour la peau humaine la constante de perméabilité atteint après 24 heures 4.67 × 10−6cm h−1. (0.5% de la quantité déposée sont alors absorbés).Chez le rat sans poils âgé de 18 mois, la radioactivité présente dans le plasma et les tissus après application sur la peau reste à l'état de traces. L'acide A γ‐L demeure concentré dans le tissu sous‐cutané (9 μg/g au temps 8 heures) et le muscle situé sous la zone d'application.Ces données pharmacocinétiques apportent une justification à l'utilisation par voie percutanée de A γ‐L en thérapeutique ou en cosmétologie.Percutaneous absorption and distribution of14C‐γ‐linolenic acid in hairless rats and manSummaryDeficiency of γ‐linolenic acid (γ‐LA), an essential fatty acid (EFA) in animals, provokes cutaneous disorders such as scaly lesions of the skin, acanthosis, loss of elasticity of the skin and irregularities in cutaneous barrier function. These disorders disappear with restitution of EFA. Although the deficiency does not exist in man, EFA could be of interest in cosmetology and dermatology, particularly in the treatment of dryness of the skin and some other cutaneous disorders observed in skin ageing. The efficiency of this treatment depends on an adequate cutaneous absorption of the acids.Experiments were performed with 4%14C‐γ‐LA incorporated in an O/W emulsion and applied to the skin.Detection of γ‐LA was only in the superficial epidermal structures and in the sebaceous glands, as shown by autoradiography obtained with hairless rat skin sections.On human skinin vitro, by using the stripping technique, very slow diffusion into cutaneous structures was observed during the first 2 hours. Eighteen percent of skin radioactivity was found in Malpighian layers after 8 hours.Permeability constants were measured in vitro with diffusion cells. In hairless rats 18 months old, Kp was 4.6 × 1−6cm h’. This value was 31 × 10−6cm h‐−1in rats of the same age deficient in EFA. With human skin, the permeability constant after 24 hours was in the same range as that in normal rats (4.67 × 1C6 cm h−1). During this time, 0.5% of γ‐LA was absorbed.In anaesthetized hairless rats, 18 months old, the radioactivity in plasma, liver, kidney, muscular and fatty tissues remained negligeable up to 8 hours after cutaneous administration of γ‐LA.
