AbstractTwenty‐four patients with cancer and concomitant infections with either herpes virus hominis or varicella zoster virus were treated with polyinosinic·polycytidylic acid (rIn·rCn) to determine: (1) the reliability of rIn·rCnto induce interferon production, and (2) the toxicity of the drug. Seven additional patients with herpes zoster were observed as controls. Two lots of rIn·rCnwere used; Lot 1 was consistently effective in stimulating serum interferon at doses of 9 and 12 mg/kg, whereas Lot 2 was effective at doses of 3–12 mg/kg. There was no correlation between rIn·rCndoses within these ranges and the resultant interferon levels. Generally, peak serum interferon occurred within the first day.Toxicity to rIn·rCnconsisted of fever in 21/24 patients, mild elevation of liver enzymes in 8/24 patients, and laboratory abnormalities of coagulation in 9 patients. The coagulation abnormalities appeared linearly related to the dose of rIn·rCnused. All these abnormalities were reversible, and none were considered severe or lifethreatening.Sincer rIn·rCnwas effective in stimulating interferon and since toxicity was considered acceptable, a randomized double‐blind study was initiated to determine whether rIn·rCnis effective in the treatment of

AbstractThe structural proteins in purified preparations of variola, monkeypox, and vaccinia viruses were separated and compared by using a high resolution SDS‐polyacrylamide gel electrophoresis system. About 30 proteins were resolved for each virus by autoradiography of longitudinally‐sliced gel rods. Although the autoradioelectropherograms of each virus were similar, it was possible to differentiate them by their unique protein pattern in the 30,000 to 40,000 molecular weight region of the gels. A single virion glycoprotein (mol. wt. = 38 × 103) and a virion phosphoprotein (mol. wt. = 12 × 103) were associated with each of the virus preparations.Cross‐absorbed monospecific immune sera against variola, monkeypox, and vaccinia virus‐infected cells were used in immunodiffusion tests to precipitate radiolabeled, homologous, soluble antigen proteins. AbstractWhile the involvement of measles‐like virus with subacute sclerosing panencephalitis (SSPE) has been shown in many laboratories, the possible mechanisms by which a normal measles infection may lead to the chronic condition are unclear. The work reported here demonstrates that the growth and maintenance of Vero cells infected with the MV 30 strain of measles, in the presence of multivalent antibody, results in a redistribution of the viral antigens within the cells and an alteration in the properties of this attenuated virus which persists even after removal of the antiserum from the culture medium. It is suggested that these in vitro findings could be parallel to the situation that occurs in vivo with measles encephalitis in hamsters and human SSP

AbstractAlthough several studies have been done to analyze the peptides of purified 22‐nm HbsAg particles, no information has been published about the peptides of the core of the Dane particle which bears the other hepatitis B viral antigen, HbcAg. Dane particles and Dane particle cores (produced by NP‐40 treatment of Dane particles) were purified by equilibrium centrifugation in CsCl density gradients. Two populations of Dane particles were observed at densities 1.27 and 1.24 g/ml, respectively. The higher buoyant density Dane particles yielded exclusively cores of buoyant density 1.38 g/ml in CsCl, and the lower buoyant density Dane particles yielded two kinds of cores with buoyant densities of 1.38 and 1.325 g/ml, respectively. Only the higher density Dane particles and cores manifested endogenously primed DNA polymerase activity. The peptides of density 1.38 g/ml Dane particle cores purified by equilibrium CsCl density gradient centrifugation and HBcAg particles from HBV infected chimpanzee liver purified in the same way were analyzed by SDS‐polyacrylamide gel electrophoresis. Both kinds of particles were found to consistently contain 3 Coomassie blue staining peptides with approximate molecular weights of 19,000, 70,000 and 80,000 daltons (designated P‐19, P‐70 and P‐80 respectively). In addition, the HBcAg particles from infected liver regularly yielded a protein component with molecular weight greater than 200,000 daltons. This component was occasionally present in electrophoresis runs of core peptides from only one of two patients. Its irregular appearance after gel electrophoresis suggests it may have been an aggregate not completely dissociated under the conditions used. The lower density core component consistently contained P‐19, P‐70, and P‐80, and infrequently additional minor peptides of uncertain origin. AbstractExperimental infection of two chimpanzees with the Phoenix Antigen strain of HAV resulted in the cyclic excretion of virus particles on days 9–11, 14–15, and 20–21 postinoculation. Isopycnic banding in CsCl of stool suspensions prepared from 9–11; 14–15;and 17, 19, 21 dav stool pools revealed multiple buoyant densities for the associated HAV particles. Hollow HAV particles found in the 9–11 day pool banded primarily at a buoyant density of 1.30 g/cm3. HAV in the 14–15 day stool banded bimodally in a CsCl gradient, with antigen peaks at buoyant densities of 1.29 and 1.33 g/cm3. AbstractThe present investigation describes the biological and biochemical properties of a persistent SSPE virus infection. Persistently infected cells were derived by cocultivation of infected brain cells and uninfected Vero cells, and cultures were maintained by normal subculturing methods.No infectious virus was ever released from these cultures, and all attempts to induce infectious virus release were unsuccessful. Biological assays showed that infected cells contained nucleocapsid and salt‐dependent hemagglutinin antigens, whereas the normal hemagglutinin appeared not to be present. Electron microscopic examination demonstrated the presence of both intranuclear and cytoplasmic nucleocapsids together with the release of virus particles (defective?) from the cell membrane.Biochemical analysis demonstrated that approximately 90% of the intracellular genomic RNA was defective or subgenomic although a small quantity of infectious genomes was present.It is proposed that the large quantities of defective genomes in the infected cells are the major factor in the maintenance of this persistent infectio
