摘要:
Elongated, tapered inclusions, present in the cytoplasm of macrophages in mouse bone marrow, were studied by electron microscopy. The bone marrow of adult mice that were injected with the hemolytic agent phenylhydrazine, displayed a statistically significant increase in the number of inclusions compared with bone marrow from control animals. Ultrastructural analysis demonstrated that ferritin, a known product of red cell destruction, was present in these mclusions. It is suggested that the inclusions are derived from the degradation of phagocytosed red cells.

The postnatal development of myocardial cells in the bilateral atria and ventricles of rats at days 1, 10, 20, 30 and 60 was studied by means of an ultrastructural quantitative method. No clear differences between the right and left myocardium were found throughout the developmental period or in the adult stage. The myocardial cells increased in thickness more rapidly up to day 20 than during the subsequent period. Myofibrils occupied about 50–60% of the cytoplasm of almost all kinds of cardiac muscle cells except those of the newborn atrium. The fraction of mitochondria increased after day 10, whereas that of glycogen conversely decreased. Although the presence of specific granules characterized the atrial myocardial cells even in newborns (day 1), no differences in other cellular components or fiber thickness between the atrium and the ventricle were seen at this stage. The distinction between atrial and ventricular muscle cells, in terms of fiber thickness, mitochondrial and glycogen content, began to appear at day 10 and became increasingly prominent with age until virtually all muscle fibers reached an almost mature state at day 2

摘要:
The bone marrow of resected bones was studied by electron microscopy before and during cultivation on the 9th, 13th and 15th day of explantation. The absence of collagenous fibers in the inoculum and its appearance in individual sites in 9-day cultures simultaneously with growth of fibroblasts, in numerous sites in 13-day cultures and in almost all sites in 15-day cultures point to the appearance and growth of coUagenous fibers after a certain cultivation time. This is also confirmed by the appearance, in the latest cultures, of coUagenous fibers with transverse striation, which indicates that they mature during cultivation. The results obtained may serve as evidence of growth of collagen-producing cells (true fibroblasts) in the above-mentioned cultures.

Tannic acid was found to fix and stain glycocalyx heavily. After removal of the major component of surface glycopeptides by trypsin, the surface coat was stained vaguely, and after the treatment with collagenase, the surface coat was moderately stained. It is concluded that tannic acid stained non-specific surface glycopeptides.

Spermiogenesis and the stages of the seminiferous epithelium in the testis of the giant rat were studied in domesticated animals. 19 steps of spermiogenesis on the basis of the development of the acrosomic system and flagellum were observed, and the cycle of the seminiferous epithelium can be divided into 14 stages. Although the developing spermatids and spermatozoa of the giant rat differed structurally from those of the albino rat, the general trend of development and the cellular content of the seminiferous epithelium were similar in both species of animals.

Different oxidants regulate the staining ability, the hue, and the staining target in bone, cartilage and connective tissue in vitro in connection with alizarin red S (ARS). The use of oxidants should promote the staining ability on guinea pig bone, also slightly on rabbit bone, but not on rat bone. Quite selective staining of bone, cartilage and connective tissue was achieved in this study via ARS oxidation. In addition to oxidizing, complex formation, especially when regulating the staining target, may be considerable. In these twelve experiments with oxidizing agents, the most promising results as concerns selectivity were obtained with phosphomolybdic acid, ferric chloride, stannic chloride and chromium trioxide. As agents activating the staining ability, potassium persulfate seems best. Based on the oxidizing influence of potassium persulfate and according to the potentiometric and precipitation analyses, the coordinative bonding of Ca2+ to the ARS molecule seems obvious. When the β-phenolic group of ARS is partly oxidized and, on the other hand, partly ionized, indicating the titration range, Ca2+ should be bound to it. Oxidized α-quinonic oxygen begins to form a coordinative bond with the same Ca2+ and thus the chelate is formed. On the slightly alkaline side, where the α-phenolic group has its ionization ranges, the oxidant causes a clear broadening and increase in the complex formation effect, which seems to be an indication of reactions of the calcium ions with two oxidized and partly ionized ARS molecul

The aim of this study is to present a detailed and thorough desciption of the acetylcholinesterase enzyme (AChE) content of the limbic structures of the rabbit, now commonly used in our laboratory for investigating the electrographic-behavioral correlations of the cholinergic limbic system. We use ‘chemitrodes’ for intracerebral microinjections of cholinomimetic drugs; a detailed histochemical study (using Koelle’s method) of the distribution of AChE enzyme in the limbic system of the rabbit was deemed necessary. Such areas as the septum and the amygdala are particularly poorly described in the rabbit and an attempt was made to clarify the confusion that still exists in the literature on the comparative anatomy and exact localization of these structures. On the whole, it could be inferred from this study that AChE-bearing neurons are generally better developed in phylogenetically older parts of the brain. Practically all the relays in Papez’s ‘cortical circuits of emotion’ have a fairly high AChE activity. The fine-structural localization of non-specific esterase was studied in the alveolar tissues of hamster lung by incubating sections of glutaraldehyde-fixed tissues in a medium containing lead ions and thiolacetic acid. The nature of the reactive enzymes was determined by exposing some sections to different inhibitors. The results showed that nonspecific esterase activity was strongest in the inclusion bodies and vacuoles of alveolar macrophages. Weaker reactions were detected in the inclusions of type II pneumonocytes, the specific granules of eosinophilic and polymorphonuclear leucocytes, and the dense bodies of type I pneumonocytes and septal fibrocytes. It is suggested that the esterase within the alveolar macrophages and type II pneumonocytes may be responsible for the catabolism of surface-active lipids.

Rat pancreatic tissues were fixed with our modification of Dalton’s fixative and the islets from various pancreatic regions were examined by electron microscopy. In addition, the effect of the alloxan-diabetic state on the pancreatic polypeptide cell was examined at the ultrastructural level. The results are as follows. (1) The islets of the rat contain four types of glandular cells: A, B, D and PP cells. (2) The PP cell is characterized by approximately 170-mµ-sized secretory granules with a rather ill-defined core. The ultrastructure of this type cell is described in some detail. (3) In the pancreatic portion which hooks around posterior to the superior mesenteric vessels, PP cells are abundant and some of them are markedly degranulated. Their cell organelles, such as the nucleolus, granular endoplasmic reticulum and Golgi apparatus are well developed. (4) In alloxan-diabetic rats, B and PP cells show disappearance and numerical increase, respectively. In the PP cells, hypertrophy of the Golgi apparatus and appearance of extremely electron-translucent secretory granules are not

The pH-metric titration curve of alizarin red S (ARS) showed that ARS ionizes in three stages: firstly the –SO3Na group, secondly the β-hydroxyl group, and finally on the alkaline side the α-hydroxyl group. The titration of ARS solution with calcium ions indicates that complex and chelate formation between ARS and calcium are distinct. The in vitro staining experiments were carried out at different pH levels. Completely ionized ARS had the best affinity for growing bone surfaces. The next most effective staining occurred just prior to neutralization. The least effective staining by ARS was found after the neutralization point had been reached.
