摘要:
The kinetics of a pharmaceutical formulation can be determined by modelling the relation between concentration of drug in the plasma and the time elapsed since administration. The resulting concentration-time curve reflects the absorption, distribution and elimination of the drug. The simplest model that represents this,kind of data is the one compartment open model. In many situations, this serves as a convenient model for estimating the bioavailability parameters area under the concentration-time curve, time to peak concentration, and peak concentration.

A Monte Carlo study was conducted to investigate the estimated standard errors of the estimate and the 95 percent confidence interval estimates associated with the trimmed Spearman-Karber (SK) estimators of the ED50 and the logistic model maximum likelihood estimator (MLE). The simulated binary response bioassay experiments had widely spaced doses with 5, 10, or 20 subjects per dose. For data following a logistic tolerance distribution, the trimmed SK confidence intervals were nearly as accurate as the logistic MLE intervals. For heavy-tailed tolerance distributions, the trimmed SK confidence intervals were more accurate than those based on the logistic MLE.

A simulation experiment compares the accuracy and precision of three alternate estimation techniques for the parameters of the STARMA model. Maximum likelihood estimation, in most ways the "best" estimation procedure, involves a large amount of computational effort so that two approximate techniques, exact least squares and conditional maximum likelihood, are often proposed for series of moderate lengths. This simulation experiment compares the accuracy of these three estimation procedures for simulated series of various lengths, and discusses the appropriateness of the three procedures as a function of the length of the observed series.

This paper discusses shrinkage estimation in nonparametric Bayesian survival analysis using censored data. The shrinkage estimators proposed are based on estimating the parameter measure of a prior Dirichlet process in a nonparametric Bayesian survival curve estimator which is the posterior mean of this process. The shrinkage is toward a prior family of exponential survival curves. The estimators are then compared by simulation with the wholly nonparametric estimator of Kaplan-Meier and the maximum likelihood estimator for the exponential family.
