Fibroblasts from a carrier of an X/1 translocation, 46, XY, t(X;l)(q28;q31), were fused with Chinese hamster cells. The resulting hybrids were analyzed for human No. 1 and X-chromosome markers. The data indicate that the loci for PGM1, PGD, PPH, and GuK1 are situated either in the long arm proximal to a break point in band lq31 or in the short arm. The loci for Pep-C, FH, and GuK1 are located distal to the break point. HPRT and G6PD are probably situated distal to a break point in band q28 of the X chromosome; α-Gal A is situated proximal to the break point, either on the long or short arm of the chromosome

Homomorphic, chromosomally abnormal roosters were mated to normal hens. The 23 hens produced 67 embryos, including two triploids and a haploid-triploid mosaic at about 26 hours of incubation. Both of the triploid embryos were conceived within a 5-day period. The presence of a single genome of paternal origin with marker chromosomes in each triploid led to the conclusion that these embryos were derived from diploid, ZW-type ova fertilized by haploid, Z-type spermatozoa. The inheritance pattern of the mosaic embryo was clearly due to a spermatozoal origin for the haploid cell line; and one genome of the three in the triploid cell line was paternal. The sex chromosomes were Z/ZZZ, with one Z of each cell line being a translocation product of paternal derivation.

Indian Muntiacus muntjac G-banded chromosomes were used for computerized analysis for standardized karyotype generation. Individual chromosomes on high-contrast photographic negatives were scanned densitometrically. Alignment of each chromosome for analysis was achieved by locating predominant peaks as well as the centromere. This provided better alignment than the use of the chromosome-end locations. The standardized set was obtained by determining the root-mean-square average density along 10–20 homologous chromosomes. The resulting standard karyotype differs from those published earlier. Prophase chromosomes exhibited greater detail than more condensed metaphase chromosomes. Although Indian muntjac chromosomes were used as a model, the method of analysis should be readily adaptable for examining chromosomes of any origin. We have developed a simple, reproducible microtechnique for obtaining metaphase chromosomes from peripheral blood of live mice. The method has been successful with mice of several different genetic backgrounds and has been repeated in three other laboratories.

A technique is described for visualizing late-replicating regions by a Hoechst 33258-Giemsa-staining procedure combining the techniques of Latt (1973) and of Perry and Wolff (1974). The advantages are two-fold: distinct bands are obtained and many possible mistakes and interpretation difficulties with autoradiography are avoided. The time sequence of late-replication patterns (excepting C-group chromosomes) has been established in human fibroblasts of adults, and these results have been compared with three of four different cell types from amniotic fluid. No significant differences in late-replication patterns and time sequence of the different cells could be discovered. As expected, the replication patterns are in good concordance with the patterns of G- and Q-bands. Some exceptions are described.

Hyla versicolor (2n = 48) is a tetraploid counterpart to H. chrysoscelis (2n = 24). Cytochemical measurements revealed that no cytological parameter of the two species conformed to the expected 2:1 ratio. Every cytological factor of the tetraploid appeared to have been regulated toward the diploid level. H. versicolor-to-H. chrysoscelis mean ratios and their standard errors were: DNA, 1.90 ± 0.03; nuclear histone, 1.70 ± 0.18; nuclear RNA, 1.63 ± 0.19; total nuclear protein, 1.46 ± 0.05; nuclear sizes, 1.42 ±
