SUMMARYThe blistering response provoked by application of 50% ammonium hydroxide solution was measured in two groups, young adults (aged 18–30 years) and older adults (aged 65–75 years). The initial response, characterized by the appearance of tiny follicular vesicles, occurred more quickly in older adults. This age‐associated reduction in initial response time (IRT) probably reflects either enhanced diffusion through appendageal shunts or loss of stratum corneum integrity. In contrast, the time required to produce a full blister was greatly prolonged in the aged. This age‐associated increase in blister formation time (BFT) probably reflects a diminished microvasculature which cannot supply fluid to fill the blister. It was also noted that the older adults displayed a decreased sensitivity to the mild discomfort experienced during this procedure. SUMMARYWe present Stereological procedures for morphometric evaluation of intercellular junctions appearing in ultrathin sections of epithelia. A model representing desmosomes and hemidesmo somes as flat, circular discs differentiated within the plasma membrane of epithelial cells forms a basis for estimating number per unit volume of tissue or per unit area of cell surface. Methods for estimating relative surfaces are also provided. Consideration is given to problems of tissue sampling, varying feature size and finite section thickness. The procedures are improvements over earlier estimates of number per unit area of section or per unit length of membrane. Worked examples of calculations are included and methods are illustrated further by reference to quantitative studies of hamster cheek pouch epithelium during experimental carcinogenesis.

SUMMARYTopical application of difluoromethyl ornithine, an irreversible inhibitor of ornithine decar‐boxylase, on UV‐irradiated mouse skin, reduced epidermal putrescine and spermidine contents and greatly enhanced the epidermal uptake of subsequently applied methylglyoxal bis(guanyl‐hydrazone). High epidermal concentrations of methylglyoxal bis (guanylhydrazone) could effectively be lowered by a single application of an ointment containing spermidine. SUMMARYThe relationship of the rate of epidermal proliferation to ultraviolet (UV)‐induced cell death, namely sunburn cell (SC) formation, was investigatedin vivousing the guinea‐pig model. The number of SCs was correlated with (1) cellophane tape stripping which stimulates the proliferation rate and (2) treatment with antimitotic and antimetabolic agents which suppress the proliferation rate. The tape stripping performed 14 h before UV irradiation significantly increased the number of SCs counted 24 h after UV irradiation from a mean of 17.4 to 62.6 per section, and the tritiated thymidine (TdR‐3H)‐labelling index (LI) increased from a mean of 8.1 to 27.4%. SC counts and LI were markedly decreased by the intradermal injection of colchicine, methotrexate, hydroxyurea or 5‐fluorouracil given once immediately after the tape stripping. The autoradiographic study showed that 23.2% of SCs were labelled when TdR‐3H was given intradermally 1 h before UV exposure, while no SCs were labelled when TdR‐3H was given immediately after UV exposure. The labelled SC count (23.2%) which presumably represents S phase cells at the time of UV exposure, was higher than the LI (mean 8.1%). These results suggest that proliferating cells, possibly S phase cells, may be responsible for

SUMMARYA study of the abnormal sunlight response seen in nine patients taking benoxaprofen showed that the action spectrum was between 310 and 325 nm, which is similar to the drug's absorption spectrum.In vitrostudies using three separate methods indicated that benoxaprofen has a powerful phototoxic action which may be directed mainly against cellular membranes.

SUMMARYDistinctive abnormalities in the immunofluorescence/immunoperoxidase staining pattern of dermal elastic tissue were observed with antibodies to serum amyloid P component (SAP) in solar elastosis, lupus erythematosus, porphyria and pseudoxanthoma elasticum, and resembled those seen with conventional elastic tissue stains. Electron microscopy of elastotic skin revealed the presence of vacuotated disintegrating elastic fibres. Islands of amorphous microfibrillar material were surrounded by a rim of specific staining with anti‐SAP, indicating an elastic tissue origin; there was no evidence for the involvement of collagen fibres in the formation of elastotic material. SUMMARYThe chemical nature of the dermal amyloid deposits in primary localized cutaneous amyloidosis (PLCA) was investigated using histochemical and immunohistochemical techniques. Deposits of macular, papular and nodular PLCA showed potassium permanganate resistance of their affinity for Congo red dye, unlike amyloid A protein (AA) deposits, and did not bind antibodies to AA, prealbumin or fibronectin. Despite the frequent presence of immunoglobulins in deposits of macular and papular PLCA, there is no evidence that the amyloid fibrils are composed of protein AL.
