SUMMARYPolyacrylamide gel electrophoretic investigations were carried out on solubilized proteins from psoriatic and normal stratum corneum obtained by adhesive tape stripping. The proteins in the scales adhering to the tape were solubilized by incubating the tape in 1% sodium dodecyl sulphate (SDS) solution. The electrophoretic behaviour of these solubilized proteins on SDS‐polyacrylamide gel was compared with the α‐fibrous proteins (keratin) of callus. The proteins isolated from callus of normal human heel showed six main bands which were similar to those of the keratin isolated by the 8 M urea‐mercaptoethanol method. The lesional skin of forty‐five psoriatic patients consistently showed nine main bands on polyacrylamide gels, but only two main bands were observed in the non‐lesional, non‐heel skin. Six of these nine bands had mobilities and relative intensities almost identical with those of α‐keratin extracted by the mercaptoethanol method, but the other three bands had greater mobilities on the gels. These results suggest that this technique may have considerable potential for studying changes in a‐keratin in patients with psoriasis and other disorders

SUMMARYBuccal mucosa biopsies from patients (seven with and seven without psoriasis) were studied using autoradiography and SDS polyacrylamide gel electrophoresis. The flash labelling indices were similar in psoriatic and non‐psoriatic patients and the SDS‐gel patterns showed variable amounts of 70,000 mol. wt (70K) protein which did not correlate with the presence or absence of psoriasis or with individual labelling indices. SUMMARYFive dark‐skinned individuals presented with widespread well‐demarcated hypopigmented lesions, biopsy of which revealed the histopathological features of mycosis fungoides. Ultrastructural studies showed focal invasion of the epidermis by mycosis cells with degenerative changes in adjacent melanocytes and keratinocytes. The majority of melanocytes exhibited swelling of cytoplasmic organelles and disordered melanogenesis with production of spherical incompletely melanized melanosomes. In addition disintegrating melanocytes were occasionally seen. SUMMARYThe reproducibility of the pulsed ultrasound technique for the determination of skin thickness was investigated, using two independent observers. No systematic difference was found and a high correlation was obtained. Studies were also undertaken to validate the pulsed ultrasound technique as a measure of true skin thickness. Skin thickness determinedin vitrowas found to be greater than whenin vivodeterminations were made by either the pulsed ultrasound or a xeroradiographic technique, probably due to the release ofin vivotension within the dermis after excision. Skin thickness was found to increase linearly with age up to the age of 20 years and to decrease linearly with age subsequently. Differences in skin thickness between the sexes and in different sites of the body were demonstrated.

SUMMARYA modified photometric technique has shown considerable differences between the refatting kinetics of the scalp and forehead. The refatting on the forehead occurs rapidly, probably due to the large follicular reservoir. The follicular reservoir of the scalp, which bears a large terminal hair, is much smaller and so the refatting time is longer. Thus, sebum excretion and sebum production rates probably correlate better on the scalp than on the forehead, and we suggest that the scalp may be a better site than the forehead for investigating sebaceous gland physiology.
