SUMMARYBasement membrane components and collagen biosynthesis were studied in suction blisters in human skin. The basement membrane components were characterized by immunofluorescence using specific antibodies to type IV collagen, laminin and fibronectin, and collagen biosynthesis was studied by assaying galactosylhydroxylysyl glucosyltransferase.In suction blisters, the separation of epidermis and dermis occurred above the lamina lucida, indicating that the basement membrane, composed of lamina lucida and lamina dcnsa, forms a mechanically strong entity. During the regeneration phase of blisters, type IV collagen and laminin were not observed in the old epidermal blister roof. This indicates that keratinocytes when separated from the underlying basement membrane or connective tissue do not synthesize laminin or type IV collagen.GalactosylhydroxylysyJ glucosyltransferase activity could be demonstrated in blister fluid and was about the same as in serum when expressed on the basis of protein in fresh blisters. It increased by 2–3 fold during the repair of blisters, indicating that there was local production of this enzyme. Further studies revealed that pure epidermis contained galactosylhydroxylysyl glucosyltransferase and hydroxyproline and this suggests that epidermis may synthesize some collagen type which, according to these studies, is not type IV (basement membrane) collage

SUMMARYPurified antibodies against type IV collagen and laminin were used to localize basement membranes by indirect immunofluorescence in various anatomical regions of normal and diseased human skin. The two proteins showed extensive codistribution. A continuous linear staining was found along the epidermal‐dermal junction and around hair follicles, sebaceous gland acini and small capillaries. The same proteins also surrounded individual cells such as those found in vessels, hair erector muscles and subcutaneous tissue.Blister formation in bullous pemphigoid left type IV collagen and laminin on the floor of the blister, while the bullous pemphigoid antigen as detected by human autoantibodies was found on both sides of the blister. In solid basal cell carcinoma a strong staining was found around all tumour islands as well as focally within the cell clusters. SUMMARYClinically normal forearm skin often subjects was abraded and biopsied at 24 h and at 5 and 10 day intervals. SUMMARYThe presence in clinically normal human skin of the proteinase inhibitor antithrombin III (ATIII) has been studied by indirect immunofluorescence (IF) microscopy. The IF staining reaction for ATIII in skin tissue sections was most prominent in vessel wall structures. The simultaneous use of fluorescent staining techniques for nuclear DNA revealed that ATIII antigen tends to be concentrated at the level of the vascular cndothelial basement membrane, in contrast to the typical intra‐endothelial distribution of Factor VIII related antigen.Although our findings indicate that ATIII is normally associated with dermal vessel walls, it remains to be seen whether its presence there is of importance, e.g. in modulating inflammatory skin response

SUMMARYTopical sodium laurate produced little or no immediate change in the amount of rat epidermal acid phosphatase but the levels of this enzyme progressively increased to reach a maximum at about 3 days. This coincided with acanthosis and thickening of the stratum granulosum, indicating epidermal repair. The increase depended on protein synthesis as it could be blocked by treatment with cycloheximide. This inhibitory effect was associated histologically with loss of the stratum granulosum, supporting the belief that the expression of acid phosphatase is closely linked with this layer during normal keratinization.

SUMMARYMetabolic activity and structure of the stroma adjacent to basal cell carcinomas are closely related to cytological aspects and proliferative activity of the neoplastic cells.Basal cell carcinomas without clear evidence for regression compress or infiltrate an apparently normal dermis where cellular replication is low. Conversely, regressing zones of basal cell carcinomas are surrounded by a remodelled stroma where fibroblasts and endothelial cells show signs of proliferation. When regression affects the palisaded array of cells, clefts occur between the basal cell carcinoma and the adjacent stroma.Progression of most types of basal carcinomas may proceed by successive extension and partial regression. More than one type of stroma reaction may be present in a single basal cell carcinoma and the overall pattern may be regarded as the result of successive phases of growth and regression of the neoplasm.

SUMMARYSingle cells isolated from the stratum granulosum and observed by phase microscopy were seen to contract upon exposure to adenosine triphosphate (ATP) in the presence of calcium and magnesium. These cells, mechanically isolated from trypsin‐treated epidermis of guinea‐pig, man and cattle, had been briefly exposed to glycerol or Triton® X‐100. The treatment with glycerol itself, but not Triton® X‐100, also induced a reversible contraction of the granular cell. SUMMARYThe glycocalyx of epidermal keratinocytes from psoriatic patients has been investigated by means of lectins.
