摘要:
A new approach which presumably recognizes MLC determinants (or at least structures closely linked to MLC and different from the FOUR, A J and LA series determinants) by serology is described.As the test can be performed within 10 h it could be used to match cadaveric kidney donors for these MLC determinants. Perhaps even more important is the fact that the antibodies detected by this technique could be used to study the immunochemistry of a class of determinants differing from the well known SD determinants.

HL–A antigens of 70 leprosy patients and 40 normal healthy individuals were determined by the standard microlymphocytotoxicity test. Both lepromatous and non‐lepromatous leprosy patients were tested for the presence of 11 HL–A antigens, and the frequency of each specificity was compared with that in a normal population of the same ethnic group. Although the statistical significance of HL–A8 specificity was found to be marginal in lepromatous leprosy patients, when using ordinary 2x2 statistics, there did seem to be a decreased frequency of HL–A9 among the non‐lepromatous type. HL–A typing of cadaver kidney cell suspensions by fluorochromasia cytotoxicity was successful in 76 out of 124 cases. In the 44 cases with confirmed phenotypes, 24 had identical results for lymphocytes and kidney cells. Twenty kidney cells had HL–A antigens not detected on that donor's lymphocytes, most commonly HL–A7 and HL–A8. In eight of these 20 cases, the additional kidney antigens brought the total to more than two per segregant series, in disagreement with an earlier report from this laboratory. The discrepancy was traced to a‘change in the method of complement preparation. The complement was successfully deprived of the resulting non‐specific cytotoxicity for kidney cells by absorption with human red blood cells. Prior to absorption, the complement had rendered kidney cells susceptible to the lytic effect of anti‐A and B red cell antibodies. Using the absorbed complement, a patient who had hyperacutely rejected two cadaver kidneys provided sera with an antibody reacting with, and absorbed by, the kidney cells but not the lymphocytes

The purpose of this investigation was to see whether or not the hypothetical disease susceptibility locus to ankylosing spondylitis is linked to the MLC determinant 27a. Firstly, we discovered a strong MLC determinant which we called 27a because of its association with the second series specificity W27. Later, we found that this determinant was the same as that which Jorgensen et al. (1973) found to be associated with the second series specificity W5. These MLC determinants may, therefore, be in linkage disequilibrium with more than one HL–A specificity (in this case, of the second series) as previously described by Dupont et al. (1973). However, we found no association between 27a and ankylosing spondylitis.On the other hand, we confirmed the association of ankylosing spondylitis with W27 and also found an increase of HL–A2 of the first series which, however, was not statistically significant. In view of the high incidence of HL–A2 in the general population, more information is required to definitely establish such an association. We found no special association with specificities A J and Hu (Sa 532), third series antigens, but confirmed their linkage disequilibrium with W27.Disease predisposition loci seem, therefore, to be associated with either type of MHS marker, multiple sclerosis and MLC determinant 7 a, or ankylosing spondylitis and the second series antigen W27. A high association of HL–A27 specificity with ankylosing spondylitis was found in Japanese patients in spite of a very low frequency of this specificity in a normal Japanese population. These findings coincide well with those in Caucasian patients, and indicate the strong relationship between the susceptibility to ankylosing spondylitis and HL–A27 specificity beyond racial differences.No particular HL–A patterns were noted in patients with the ossification of the posterior longitudinal ligament of the cervical vertebrae.
