A method for preparing radiolabelled human Ia antigens and a utilization of the labelled antigens in defining the la specificity of antigens and antisera are presented.The standard preparation procedure involves solubilization with Renex 30 (a non‐ionic detergent), purification by gel filtration and lectin‐affinity chromatography, and radioiodination, and yields an Ia preparation with immunological purity of 48–62%.The basic procedure for defining the Ia specificity of antisera is a direct binding assay: it allows to react a given antiserum with a panel of labelled la preparations of different specificities and measures the extent of binding by the double antibody technique. The major or putative specificity can be deduced from the reaction pattern. The specificity can be further evaluated by a sequential binding test or a binding inhibition test.The Ia tissue typing is performed by the binding inhibition assay: it measures the ability of test tissues to inhibit the binding of a reference labelled Ia preparation with a reference Ia antiserum. Two patients with type I Glanzmann's thrombasthenia and 20 kindred of these patients belonging to 2 families of the Manouches gipsy tribe have been studied. Quantitative measurements of platelet membrane glycoproteins GP IIb and GP IIIa have made it possible to classify the patients into normal, thrombasthenic or carriers of the thrombasthenic abnormality. We have examined several red cell alloantigens and antigens of the major histocompatibility complex. These studies have shown that: 1) type I Glanzmann's thrombasthenia (GP IIb and IIIa abnormality) segregates independently of Ss and Fy systems and the A, B, C, Bf, C2 and C4 loci of the HLA complex; 2) a rare hemolytically inactive C4 variant segregates in these families but is not associated with the GP IIb and IIIa abnormality.

Positive associations between the antigens D(R)3and D(R)4and negative associations of D(R)2have been reported with insulin dependent or type I diabetes mellitus. It has been suggested that susceptibility factor(s) associated with the D(R)3and D(R)4haplotypes and a resistance factor associated with the D(R)2haplotype may be involved in the pathogenesis of the disease. We propose a hypothesis herein which attempts to unify these findings based on our present understanding suggesting (a) the existence of multiple antigenic determinants associated with any one D(R) haplotype and (b) the sharing of “single” D region encoded determinants between what we now refer to as different D(R) haplotypes. Aiming at the production of anti HLA‐DR test sera, eight healthy human volunteers were immunized by repeated intradermal injections of lymphocytes which were selected to be incompatible for one HLA‐DR antigen, and matched as well as possible for HLA‐A,‐B,‐C antigens. One out of 3 recipients immunized exclusively against HLA‐DR produced lyrnphocytotoxic HLA‐DR antibodies. The remaining 5 recipients were immunized against 1 or more HLA‐A,‐B,‐C antigens in addition to one HLA‐DR antigen. After 3 immunizations, 3 of these reacted with strong HLA‐A or ‐B antibody production; however, only one showed a parallel anti HLA‐DR antibody response detectable by complement dependent lymphocytotoxicity.Testing of the recipient sera in the antibody dependent cell‐mediated cytotoxicity (ADCC) assay revealed that 6 of the 8 recipients did react early to the immunizations with HLA specific antibody production. CHEV, a new Epstein‐Barr virus negative Burkitt's lymphoma derived cell line has been studied. Karyotype analyses demonstrated the t (8; 14) characteristic translocation. The LD50 of total body irradiation (TBI) for the bone marrow (BM) syndrome and the gastrointestinal (GI) syndrome was determined in dogs as 3.7 Gy, and 8.5 Gy respectively. Five Gy TBI was adequate conditioning for BM cells of littermate donors identical for the major histocompatibility complex (MHC). The maximum tolerated TBI (about 7.5 Gy) caused more side effects than 5.0 Gy TBI and was insufficient for engraftment of realistic numbers of BM cells of MHC mismatched donors. In autologous and MHC matched transplants, the rate of hemopoietic recovery correlated with the number of BM cells given. Approximately 2 × 107autologous and 1 × 108MHC identical BM cells.kg−1were needed for radiation protection. Platelet recovery was significantly more rapid in allogeneic combinations in comparison to autologous transplants. Low numbers of autologous cryopreserved bone marrow cells were as effective as fresh bone marrow cells in rescuing animals after lethal TBI. Other factors that influence BM cell engraftment were confirmed (prior sensitization of the recipient, donor selection) or identified (purification of BM cells on density gradient and selective gastrointestinal decontamination of the recipient). Consistent engraftment of gradient separated, MHC identical, BM cells was found after conditioning with two fractions of 6.0 Gy TBI, separated by 72 h. One MHC haplotype mismatched marrow did engraft after two TBI fractions of 6.0 Gy. Engraftment no longer occurred with gradient purified bone marrow cells from this type of donor. Late effects of TBI were early greying in all animals, and secondary uterinc inertia in female dogs after 7.5 Gy TBI. Fertility in males or females was not changed by radiation. An increased incidence of pancreas fibrosis was noted in dogs receiving two fractions of 6.0 Gy
