We have used a lysostripping technique to investigate the molecular relationships of the determinants recognized by human alloantisera and murine monoclonal antibodies reactive with HLA‐D region products. Cytotoxicity mediated by sera that define the specificities DR1, DR2, DR3, DR4, DR7, DRw11, DRw12, DRw13, DRw14, DRw52 and DRw53 was inhibited by anti‐DR but not by anti‐DQ Mabs. In contrast, sera that define DQw1, DQw2 and DQw3 were inhibited by anti‐DQ but not anti‐DR monoclonal antibodies. We found some sera used to define DR specificities that were not inhibited by monoclonal anti‐DR. Cytotoxicity mediated by monoclonal antibodies reactive with the recently described specificities TA10 and MCI was inhibited by anti‐DQ and anti‐DR monoclonal antibodies, respectively. We have studied the HLA associations of MS patients in Arab patients from two regions. Those patients from the Mediterranean area showed an association of MS with HLA‐DR2 (p<0.001) and ‐DQw1 (p<0.05), whilst in those from the Gulf there was an association with ‐DRw53 (p<0.02). In the Mediterranean patients ‐DRw52 was negatively associated with MS (p<0.05).In the control populations from the two regions there was a considerable variation in the incidence of HLA antigens. Thus, Gulf Arabs had an increased incidence of ‐DR1 (p<0.05), ‐DR2 (p<0.02), ‐DR6 (p<0.05) and decreased incidence of ‐DR5 (p<0.05), ‐DRw52 (p<0.02) and ‐DQw3 (p<0.01) compared to the Mediterranean control population. To simplify the screening procedure for murine monoclonal antibodies specific for polymorphic HLA determinants, spleen cells from a mouse immunized with the human cell line BJAB‐B95.8.6 were fused with NSI mouse myeloma cells, and hybridoma supernatants were screened for their reactivity on BJAB‐B95.8.6 and two gamma ray‐induced HLA‐loss mutants of this line.The use of these HLA‐loss mutants allowed the rapid identification of two new al‐lospecific MOABs designated TÜ160 and TÜ161. Serological as well as biochemical studies revealed TÜ160 to be specific for HLA‐A2, and TÜ161 for HLA‐B13 molecules, respectively. Large batches of Cytotoxic T Lymphocytes (CTLs) specific for antigenic components of Purified Protein Derivative of tuberculin (PPD) were generated from 20 donors. Peripheral Blood Mononuclear cells (PBM) were stimulated with PPD, expanded with Interleukin‐2 (IL‐2) and finally aliquotted and cryopreserved. It was found that CTLs could be cryopreserved without loss of activity, and compared to generating CTLs in primary culture alone, IL‐2 expanded CTLs were stronger and discriminated better.To study their HLA restriction specificity, the CTLs were tested against a panel of 50 target cell donors using PPD pulsed monocytes as antigen presenting target cells. The HLA‐restriction specificity of a set of Cytotoxic T‐Lymphocytes (CTLs) derived from 20 different individuals and with specificity for antigenic components in Purified Protein Derivative of tuberculin (PPD) were examined against a panel of 50 unrelated target cell donors. PPD pulsed monocytes were used as antigen presenting target cells. CTLs were generated by Interleukin‐2 (IL‐2) expansion ofin vitroPPD activated Peripheral Blood Mononuclear Cells (PBM). The results confirm our previous finding that PPD‐specific CTLs are restricted by HLA‐class II — and not by class I antigens.The 20 CTLs used together provide reliable cellular typing reagents for the antigens HLA‐DR2, ‐3, ‐4 and ‐7 and less so for the antigens HLA‐DR1 and ‐5. In contrast, sharing between CTL‐ and target cell donors of HLA‐DRw6 and ‐w8 correlates poorly to PPD‐specific cytotoxicity.A few consistent exceptional patterns were observed. In one case lack of lysis in spite of HLA‐DR antigen sharing could be explained by a split of HLA‐DR2 into a normal and a short variant. Positive reactions in combinations, where no HLA‐DR antigens are shared, were only few and evenly distributed among CTLs.
