HLA‐D clusters associated with DR2 and the definition of HLA‐D“AZH”: a new DR2 related HLA‐D specificity in Israel |

In order to investigate the HLA‐D clusters associated with DR2 in Israeli Jews, 40 DR2 positive unrelated individuals were studied with a panel of DR2 associated homozygous typing cells (HTC's) which detect the lymphocyte defined specificities HLA‐Dw2, Dw12, Dw9 and D‐WJR. The results confirmed the existence of two distinct HLA‐D clusters associated with the same serologically defined DR2. Of 40 individuals 22.5% (9/40) were Dw2 and 50% (20/40) were Dw12 carriers. Yet, no HLA‐D specificity could be assigned to the remaining 11 DR2 positive individuals. In the present study we have defined a unique DR2‐associated Dw specificity, HLA‐D“AZH”. The donor of the HTC was of Moroccan origin and an offspring of a first cousin marriage. This cell was not typeable with the known DR2‐associated homozygous typing cells nor with other HTC's which define the well established HLA‐Dw1 to Dw11 specificities. In the scope of the cellular part of the 9th International Histocompatibility Workshop, the offered homozygous typing cells (HTCs) of several specificities have been screened in a pre‐Workshop. In Leiden and Seattle all HTCs typing for “HLA‐Dw6” have been tested. This implied that in both laboratories mixed lymphocyte culture (MLC) matrices were performed between the Dw6 HTCs and that all HTCs were tested as stimulator cells against a panel of heterozygous responder cells. The results clearly demonstrated that “HLA‐Dw6” as defined by the different participating laboratories can be split into two major groups, Dw6.a and Dw6.b. To determine the genetic relations between HLA and deficiencies of steroidogenic enzymes other than 21‐hydroxylase, we investigated HLA and congenital hypoaldosteronism in two families with corticosterone methyl‐oxidase type 2 (CMO2) and one family with type 1 (CMO1) deficiency, respectively. Apart from a first documentation of HLA in CMO1 deficiency, our results, combined with those reported previously, excluded close linkage of HLA and CMO2 deficiency. To investigate whether an association could be found between HLA and lepromatous leprosy a population study was performed in Tachira, Venezuela. This was done in the same endemic area in which recently both non‐random parental HLA‐haplotype and preferential segregation of the HLA specificity LB‐E12 (MB1, DC1, MT1) was demonstrated in lepromatous leprosy patients from multicase families. In this study 32 lepromatous patients and 32 healthy controls were typed for HLA‐A, ‐B, ‐C, ‐DR and the specificities MB and MT. The frequency of LB‐E12 (MB1, DC1, MT1) showed a significant increase in lepromatous leprosy patients (p=0.04). Antigen HLA‐A, B sharing couples have been previously observed in abnormal pregnancies of unknown etiology. We have determined the HLA‐A, B, C and DR antigens of 20 couples with recurrent spontaneous abortions (RSAs), 32 control couples and 100 normal controls. The results showed that in couples with more than two idiopathic repeated abortions, there is no significant increase in the HLA antigen sharing couples. But we were able to observe, in the affected couples, a significant increase in HLA‐DR antigen sharing as regards the control couples. HLA‐A, B, C and DR typing was performed in healthy IgA deficient donors and in IgA deficient patients with recurrent respiratory tract infections. In healthy donors, a strong association with HLA B8 and DR3 was observed. In the patient group however, no statistically significant association with these antigens could be found. In contrast, an increased frequency of HLA B40 was noted.The preference for HLA B8/DR3 in healthy donors could not be attributed to higher levels of serum immunoglobulins since no major differences in the concentrations of IgM, IgG or the IgG subclasses were seen between the two group

Since major histocompatibility complex (MHC) antigens of class II play an important role in organ transplantation, we attempted to demonstrate their expression on endothelial cells which are abundant in transplants with vascular blood supply. Our analysis was performed with monomorphic and polymorphic monoclonal antibodies employing a microscopic immunofluorescence assay and flow cytometry on unstimulated endothelial cells isolated from umbilical cord veins. No evidence was found for the presence of HLA‐DR antigens and determinants associated with MT, MB and SB. MHC class I antigens exhibited reduced expression. These findings were confirmed in respect to alloantigens by the use of conventional cytotoxic tissue typing antisera. MB and SB antigens were present on most cord B lymphocytes, but could only be demonstrated on a subpopulation of monocytes exhibiting a lower antigen density at the cell surface. Cellular cytotoxicity generated in vitro has been studied with canine T and B lymphoblasts as target cells. B lymphocytes were isolated by resetting with protein A labeled sheep red blood cells and stimulated with sepharose bound protein A. Such highly purified stimulated B lymphoblasts appeared to be useful target cells for cellular cytotoxicity. Cold target inhibition studies and limiting dilution analysis revealed that B lymphocytes did not carry targets for cytotoxic T lymphocytes (CTL) which were not present on PHA stimulated T lymphoblasts.

We reported the data of HLA‐DR frequencies in a new series of 40 unrelated patients suffering from giant cells arteritis (Horton's disease).As previously reported by us, a large increase of HLA‐DR4 antigen frequency is noted in patients compared with 146 healthy controls. Moreover, gathering together these 40 patients with the 48 other patients of our first published data, increase of the DR4 frequency is largely confirmed with a PC<0.
