Serologic identification of anti‐PLAIand other platelet specific antibodies is difficult because other platelet reactive antibodies (e.g. anti‐HLA) are commonly found in the samples. We have developed and evaluated a method using Western blotting to identify platelet specific antibodies. Since anti‐PLAIis the most frequently encountered antibody, we used this antibody to validate the method. The PLAIantigen was found on a 95,000 Dalton protein, confirmed as glycoprotein IIIa by two‐dimensional electrophoresis. The usefulness of Western blotting in resolving difficult platelet serologic problems was compared to a radiolabeled antiglobulin test using intact platelets. Our results showed that all sera with anti‐PLAIidentified by serology were confirmed by Western blotting. In addition, one anti‐PLAIwas detectable only by the proposed method. Western blotting was also as useful as a serologic method to PLAItype platelets. HLA antibodies were not demonstrable by this technique. In summary, Western blotting is a highly specific and sensitive way of identifying anti‐PLAIin a variety of clinical situations. Class II MHC antigen (HLA‐DR) is a normal constituent of the ductular lining cells of the sweat gland. In development, HLA‐DR is first expressed on the developing bud of the growing gland as it extends down from the epidermis. The cells immediately behind the bud lose this staining. The antigen subsequently appears on the ductular lining cells and extends from the acrosyringium down the full length of the duct. The coils are negative. Class II molecular variation among a panel of ten HLA‐DRw11(5) homozygous cell lines (HCL) was investigated by analysis of restriction fragment length polymorphisms in genomic DNA. Hybridization of Bgl II, Hind III, and Taq I digested cellular DNA with DQ α and DQ β cDNA probes identified a clustering of characteristic polymorphisms. Considerable diversity was observed between the HLA‐DRw‐11(5), DQw3 positive haplotypes studied, as well as compared to a DRw11(5), DQw1 positive haplotype. In contrast to the observed DQ genomic variability, hybridization with a DR β probe revealed relatively limited diversity. This paper describes a new cell isolation and HLA typing technique, which permits cell separation and HLA class I or class II typing to be performed in 70 min. Magnetic monodisperse microspheres (Dynabeads TM) were coated with monoclonal antibodies (MAbs) specific for the CD8 T cell antigen or for HLA class II monomorphic epitopes. They could then be used to obtain HLA class I or class II positive cells directly from ACD blood in approximately 15 min by the use of magnetic separation. The cells (attached to the microspheres) were subsequently used in microcytotoxic HLA typing (total incubation time of 55 min) using acridin orange/ethidiumbromide to stain viable (yellow) and dead (red) cells. It was found that this immunomagnetic (IM) HLA typing technique was specific, has a sensitivity superior to that observed for conventional microcytotoxicity assays and gave low background staining. IM HLA‐ABC typing of 50 healthy donors and 10 patients and IM HLA‐DR typing of 25 healthy donors and 30 patients gave results corresponding well with that obtained independently by conventional HLA typing (concordancy rates 92–100%). Furthermore, the IM HLA typing technique permitted reliable HLA class II typing of blood cells from six patients where conventional HLA class II typing was impossible. A pregnancy‐induced alloantibody has allowed definition of a class II HLA antigen, AE, which has a frequency of about 10% in Danes, is completely included in DR4, and highly significantly (p = 10‐9) associated with the cellularly defined Dw14 determinant which is completely included in AE. AE is absent on Dw4 and Dw13 cells, but was also found on one Dw15 HTC and in two of four Dw10‐positive cells. It is in positive linkage disequilibrium with B40 but not with B15 or B44. It is hypothesized that the AE specificity is due to the co‐occurrence of an arginine in position 71 and an alanine in position 86 of the first domain of the DRB1chain.
