Antibodies to native chick and bovine Type II collagen were measured by radio immunoassay, in 83 rheumatoid arthritis (RA) patients and 14 normal controls. Anti‐chick Type II collagen and anti‐bovine Type II collagen antibodies were found in 48% and 43% of RA patients, respectively. A strong correlation of antibodies to chick and antibodies to bovine collagen was described, suggesting cross‐reactivity between different collagen species. There was an association between the presence of anti‐native Type II collagen antibody and the expression of HLA‐DR2. 摘要:
An anti‐HLA‐A2 and ‐A28 monoclonal antibody, XV.17, has been prepared by immunizing a Balb/c mouse with PBL. This XV.17 monoclonal antibody is a cytotoxic IgM. Its reactivity was tested by lymphocytotoxicity test and indirect immunofluorescence technique, in parallel with an alloantiserum ORA having the same anti‐HLA‐A2, ‐A28 reactivity pattern, against different panels. Family studies were undertaken. Absorptions‐elutions and cytofluorometry experiments were performed to study the cross‐reaction. HLA‐A, B, C typing was performed on 37 unrelated healthy blood donors with selective deficiency (<0.05 g/l) or total lack (<0.00002 g/l) of serum IgA. A significant increase in HLA‐B8 and A1 antigen frequency was found in the 15 individuals with lack of serum IgA. This was not observed among those 22, who had only deficiency of serum IgA (0.00002–0.05 g/l). The HLA antigen frequencies were compared to those of 900 randomly selected healthy blood donors.Increased frequency of HLA‐A1 and B8 in association with total lack of serum IgA, but not with deficiency has not been demonstrated before. In a previous publication we demonstrated that polymorphic HLA antigens could be detected on fresh and dried peripheral blood lymphocytes using the enzyme‐linked immunosorbent assay (ELISA) with HLA alloantisera (Bishara et al. 1983). In the present study we investigated whether the ELISA technique can be used in determination of gene‐dose effect for antigens of the HLA‐A and B loci. Lymphocytes from HLA‐A1 and HLA‐B14 homozygous individuals are shown to bind significantly more anti‐HLA alloantibodies than their heterozygous siblings for the same HLA antigens. We describe three human proliferating T cell colonies, derived from mixed leukocyte culture with a non‐diabetic individual (DR3+4) as the source of responding cells and an insulin‐dependent diabetic patient (also DR3+4) as the source of stimulating cells. One colony detects HLA‐Dw10 or a closely related antigen, and two detect an antigen that we call BO1 (Boston 1). BO1 is found so far on cells of all persons with DR5, about half of those with DRw6, and a particular subset of those with DR3. Among DR3‐positive subjects, BO1 is positively correlated with HLA‐B18 and BfF1, and negatively correlated with HLA‐B8. These findings suggest that BO1 occurs in linkage disequilibrium with DR5, DRw6, and the haplotype B18, BfF1, DR3, the latter being common in southern Europe and reported previously to be a marker for insulin‐dependent diabetes. In limited testing (21 subjects), BO1 was completely included in the supertypic specificity MT2. BO1 is a Class II HLA antigen, as demonstrated by blocking with monoclonal antibodies, but is distinct from all known antigens of the DR, MB(DC), MT, and SB series. In this study, we analysed 13 samples of the French population ‐ 4147 non‐related individuals living in Bordeaux, Brest, Caen, Dijon, Limoges, Lyon, Marseille, Nancy, Paris, Poitiers, Rennes, Strasbourg and Toulouse ‐ all of whom were typed for 10 alleles of the HLA‐A locus and 16 alleles of the HLA‐B locus. The results showed a strong heterogeneity (χ2= 675.13 for 324 df, p<105). A diagram has been drawn up, showing the matrix of genetic distances obtained thanks to the B2of Balkrishnan&Sanghvi (1968). This diagram enables us to envisage the hypothesis of 6 homogeneous clusters. The HLA‐A and HLA‐B phenotypes of 329 unrelated Haitian adults were determined and gene and haplotype frequencies were estimated. A summary of the data is presented together with a brief comparison with American Black, American White, and African Black populations.
