摘要:
The potential advantages of automated microscopy for reading tissue typing results are accuracy and speed. A technique which depends on fluorescence emission of both Carboxy fluorescein (CF) in live cells and Propidium Iodine (PI) in dead cells has been developed. Factors which influence the data have been investigated and appropriate mathematical corrections proposed. A practical guide is given in an appendix.

摘要:
A complement fixing monoclonal antibody (moab) was prepared which reacts with a polymorphic determinant on HLA class II molecules. The moab IIB3 recognises all DQwl (DO, MB1, LB‐E12) positive cells as well as some DR4, DR7, DRw8 and DRw9 positive cells. The moab reacts mainly with B‐cells and not or with only a minority of the monocytes. Segregation of the determinant with HLA‐DR could be shown. The determinant is strongly expressed on DR2, DR4 and DRw6 positive cell lines but only weakly on DR1 lines.In contrast to a monoclonal antibody against a monomorphic determinant on class II molecules IIB3 did not give a distinct inhibition of the MLC nor did it inhibit the generation of CTLs in MLC as has been described for the DQwl like moab BT 3/4 (Corte et al. 1982). A weak association between subacute (de Quervain's) thyroiditis and HLA‐DRw8 (relative risk = 2.19, X2= 3.86) was observed. However, the association was not statistically significant if p value was corrected. Since it was confirmed that the association of subacute thyroiditis with HLA‐B35 was primary, and since this association was extremely strong, the two models (dominant and recessive) for the effect of an HLA‐linked gene which has a major effect on the pathogenesis of subacute thyroiditis were tested by the method described by Thomson&Bodmer using the frequency of HLA‐B35 in the patients. A cytotoxic murine monoclonal antibody, designated P5.1, was tested against 613 unrelated donors and found to react with 401 who were positive for HLA‐A2 (sensitivity = 100%) and with 8 of 82 positive for HLA‐A28. The latter split of A28 corresponds to the “A28* variant” that in the Ninth International Histocompatibility Workshop (9WS) was designated Aw69(28*). The epitope recognized by antibody P5.1 is distinct from the alloantisera‐defined determinants that characterize HLA‐A2 and A28. Immunoprecipitation of specific antigens with selected monoclonal antibodies and isoelectric focusing gel electrophoresis demonstrated that A2, Aw68(28) and Aw69(28*) are distinct polypeptides. The expression of HLA class I a chains by clones derived from a human x mouse T‐cell line containing human chromosomes 6 and 15 was analysed by immunofluorescence. Evidence is presented showing that the expression of HLA class I a chains is associated with HLA‐β2m. This is based on the phenotypic and quantitative fluorescence flow cytometric analysis of a series of cloned hybrids. The HLA type DR3 was present in 11 of 26 patients with Chronic Idiopathic Thrombocytopenic Purpura (ITP), a significantly increased frequency (p<0.05). Levels of platelet associated immunoglobulin M(PAIgM) were significantly higher in the B8 and/or DR3 positive group of chronic ITP patients than in the B8 DR3 negative patients despite similar clinical outcomes. Other immunologic parameters assessed, including serum immunoglobulin levels, rate of catabolism of transfused IgG, and antibody coated autologous red cell clearances were similar for both groups. These results suggest that there is an immunobiologic abnormality associated with the B8 DR3 allotypes which may result in a predisposition not only to chronic ITP, but also to a significant increase in PAIgM. These results are in accord with studies linking autoantibody with B8 DR3.

摘要:
Oral submucous fibrosis (OSF) is a disease of the mouth and oropharynx characterised by progressive deposition of collagen leading to severe limitation of movement of the jaw in advanced cases. It is almost completely confined to inhabitants of, or migrants from India who chew‘betel nut'. The histopathological and clinical features suggest that autoimmune mechanisms may be involved. Because all chronic autoimmune diseases show distubance in the frequencies of HLA antigens, we have HLA typed 50 OSF patients and a similar number of healthy subjects of the same ethnic origin. Raised frequencies of A10 and DR3 were observed. The results support the concept that OSF is a chronic autoimmune disease, initiated by constituents of betel nut, and occurring in genetically susceptible individuals. The first two cases of post transfusion purpura (PTP) due to recognized antibodies against the platelet specific antigen Zwb(P1A1) are described. In both cases, the antibodies were detectable in the platelet enzyme immuno‐assay (ELISA), while the platelet specific alloantibodies could be ascertained neither in the platelet suspension immunofluorescence test (PSIFT) nor in the platelet fixation test (CFT) due to coexisting potent multispecific HLA‐antibodies. The two sera reacted in parallel (coefficient of correlation, r = 0.87; P<10−5) and the reactions of the two sera were negatively associated with the Zwa‐antigen (P = 0.0002 and 0.003, respectively). Lymphocytotoxic antibodies to the red cell blood groups A and B were purified from human sera by elution from synthetic immunoadsorbents (Synsorbs from Chembiomed). The lymphocytotoxic activity of the eluted antibodies was better preserved when elution was carried out at pH 11, whereas it was less stable when elution was performed at pH 3.
