DR4 related serological heterogeneity differences between type I diabetic and normal individuals previously observed appear to be due to differences in the DQ subregion. An Australasian sera study has identified two subsets of DQw3, the DQ allele in linkage disequilibrium with DR4, one of which (DQw3S) is present in 95.2% of diabetic DR4 compared with 58.1% of normal DR4 individuals (p<0.0005). The DQw3 subset showing a negative association with diabetes, DQw3L, appears to be closely related to or identical to TA10 a DQ subregion product previously described and studied in the 9th International Workshop. Analysis of the Workshop data showed a strong association of TA10‐ in type I diabetic DR4 individuals (93.5% v 69.1% in DR4+ normals) (p<0.0005).B44 appears to show an association with the presence of TA10in DR4 positive individuals. Two‐dimensional gel electrophoresis of material immunoprecipitated from radiolabeled cells by anti‐HLA class II monoclonal antibodies has been a useful technique but the patterns observed have varied considerably between laboratories. In this study we investigated the parameters that vary between different laboratories. We found that heterogeneity due to post synthetic modification of β and α chains of DR and DQ can be avoided by labeling cells for 30 min with35S‐methionine. A mixture of ampholytes pH range 3.5–10: 5–7 of 1:4 run for 3000 V/h gave the best resolution of β and α chains. With this technique we could clearly reveal differences in the DR β–2 chains of those DR2 haplotypes, Dw2, Dw12 and LD'tb24′. The DR β1 position varied between

Sixty unrelated Greek patients with haemophilia (46 with haemophilia A and 14 with haemophilia B) were typed for HLA‐A, B and DR antigens. A highly significant increase in the frequency of HLA‐DR5 was observed in both groups of patients (58.6% vs 30.0%, χ2= 10.47, pc<0.03, RR = 3.31 for haemophilia A and 78.5% vs 30.0%, χ2= 12.32, pc<0.007, RR = 8.5 for haemophilia B). An increased frequency of HLA‐B13 was also observed in patients with haemophilia A (15.2% vs 5.7%, χ2= 5.74, pc<0.4, RR = 2.9). Thirty of the 60 patients (50.0%) were positive for LAV/HTLVmantibodies. HLA‐DR5 was equally distributed in patients with and without these antibodies (63.3% and 63.3%, respectively). The presence of DR5 did not correlate with the severity of haemophilia A or B. These results may suggest 1) an influence of gene(s) on chromosome 6 in haemophilia A and haemophilia B and 2) no effect of HLA antigens in the susceptibility to LAV/ HTLVmInfection among haemophilia

The characterization of a monoclonal antibody with specificity for a monomorphic determinant on rhesus macaque class II antigens is described. Lymphocytes from highly selected donors were primed for 10 days and subsequently bulk‐expanded in IL 2 (TCGF) containing cultures. Two well‐discriminatory PLT (CDP = Copenhagen DP) reagents against each of the DPw1‐w6 specificities and one against each of the two “new” specificities, CDP4sand CDPHEI, were selected for further studies. Three combinations made in two recombinant families and four of ten HLA‐A, B, and DR compatible combinations discriminated well in contrast to seven of 46 DR compatible, but HLA‐A or B incompatible combinations. All reagents gave highly reproducible results, and high correlations (r‐values between 0.73–1.00) for DP assignments were obtained with CDP and GNN reagents. No triplets were found for the DPw1 ‐ w6 and CDP HEI specificities. The “new” specificity CDP HEI defined in an HLA‐DR/GLO recombinant family gave a coefficient of correlation with GNN 8 of 0.91. Another “new” specificity, CDP4sconstitutes a subgroup (“split”) of DPw4. The gene frequencies ofDPw1 ‐ w6estimated in 102 unrelated randomly selected Danes agreed with those reported for other Caucasoid populations. The gene frequenciesCDP HEIandCDP4swere 0.03 and 0.08, respectively. The associations between DR3‐DPw1, DR2‐DPw4, and DRw6‐DPw2 were confirmed. Class II histocompatibility antigens were assessed by complement‐dependent cytotoxicity on B enriched mononuclear cells and on purified monocytes isolated from the peripheral blood of renal patients before and after transplantation. After transplantation, a significant decrease of detection of DR alloantigens (cytotoxicity score before transplantation 3 × 0.1; after transplantation 0.95 × 0.1; p<0.001) was found. A similar defect was observed both on B enriched frozen cells and on purified monocytes isolated during the post‐transplant period. Nevertheless, antigens could be adequately detected by a sensitive microabsorption method and by a procedure involving preincubation of mononuclear cells at 37°C for 3 h followed by B cells enrichment by rosetting. The 37°C induced reexpression could be inhibited by 10 μg/ml cycloheximide and by 50 μM monensin. No difference was found in the magnitude either of decreased expression or of temperature‐induced reexpression between patients treated by CyS‐Pred and those receiving Aza‐Pred.
