The distribution of the HL–A antigens was studied in 76 patients who have had rheumatic fever or have rheumatic heart disease and in 177 disease‐free Caucasian individuals. There was a decreased incidence of A–3 in the disease group as compared to the control. No significant increase in the frequency of any HL–A antigen was detectable in the disease group as compared to normal controls. The results were not affected by the exclusion of non Caucasians from the disease group. The parents of patients with rheumatic fever and heart disease showed a significant increase in shared antigens as compared with the parents of disease‐free individuals. Transplantation antigens were extracted from various tissues of A and CBA mice with hypotonic solutions or with distilled water and were solubilized either by autolysis or by digestion with papain. The origin of the tissue (lymph node, spleen, liver, SaI cells) and the nature of the starting material (fresh or frozen, whole cells or homogenates) were also varied. The extraction procedures were evaluated in terms of the yield and specific activity of the antigen preparations and the various procedures were compared also with respect to their efficiency in solubilizing transplantation antigens. The antigenic activity of the preparations was established by their capacity to inhibit the cytotoxic reaction using different combinations of alloantisera and target cells. The best antigen preparations were obtained with an aqueous‐autolytic procedure employing extraction with distilled water, followed by limited autolysis. With all antigen preparations the bulk of the antigenic activity was eluted from Sephadex G‐100 after the void volume, indicating a molecular weight of less than 100,000 for the active components. The results also indicated that all H–2 specificities were not solubilized with the same efficiency and that the concentration and the immunogenicity of certain of the specificities are different for A lymphocytes and SaI

A series of 23 platelet absorptions and elutions were performed with a broadly reactive HL–A antiserum, Tuckerman. This multispecific reagent could be separated into three components: anti‐HL–A1 (10,11); anti‐W23 (W24) and anti‐HL–A12.2 (12.1,13). Each component was examined in detail, revealing some of the intricacies of the specific and cross‐reacting antibodies.A possible subdivision of HL–A12 followed the observation that platelets from certain HL–A12 donors failed to absorb anti‐HL–A12 cytotoxins, even against their own lymphocytes. Certain anti‐HL–A sera react more frequently with lymphocytes of chronic lymphatic leukmia than with normal lymphocytes. Selected antisera for most but not necessarily all HL–A specificities may show this hyperreactivity. Antisera to the W19 and possibly the HL–A27 complexes seem to be involved more often than are others. CLL lymphocytes can be typed accurately only if a sufficiently large number of antisera of each specificity are employed. Cross‐absorption analysis of sera EV, Thorsby‐FJH, and THA, using normal and CLL lymphocytes, yields evidence for six or more additional antigenic factors on CLL cells which are not obviously related to the HL–A system. Possible interpretations of these findings involve cross reactions within the HL–A system, reactivity with postulated non‐HL–A receptors on cell surfaces, with antigens specific for different varieties or subdivisions of chronic lymphatic leukemia (leukemia type‐specific antigens), or with T‐cell or B‐cell antigen systems that might be more abundantly expressed on leukemic than on normal cells. While a few anti‐HL–A sera are shown to have anti‐immunoglobulin antibodies, such antibodies do not suffice to
