A total of 135 patients with a definite diagnosis of Multiple Sclerosis (MS) has been HL–A typed. Increased frequencies of HL–A3 and 7 were found. Combining our material with other data revealed significant increases in HL–A3, HL–A7 and W18 and significant decreases in HL–A2 and HL–A12. The relative incidence ratios of Woolf (1955) were used in these comparisons and depicted graphically.Titres of anti‐measles virus antibody were determined in sera of 132 of the patients. Increased titres were significantly more frequent in patients carrying HL–A3, 7 and/or W18 as compared to patients lacking these antigens. Rhesus monkeys have a major system of serologically defined histocompatibility antigens, called RhL–A. Like the analogous human system (HL–A), RhL–A is highly polymorphic, and its antigens are controlled by two closely linked segregant series of alleles. About 50% of the gene products of both allelic series can now be identified serologically. Recombination frequency within the RhL–A region is approximately 1%.The relevance of RhL–A to histocompatibility was proved by intersib skin grafting experiments. The influence of matching for RhL–A on graft survival in unrelated host/donor combinations is equivocal.A genetic locus exerting a major influence on mixed lymphocyte reactivity has been identified and found to be closely linked to the RhL–A region3. Rhesus monkeys have a genetic locus which has a major influence on reactivity in mixed lymphocyte cultures (MLR). This MLR locus is closely linked to the RhL–A system2, which controls the serologically defined histocompatibility antigens of this species. Three cases of recombination between RhL–A and the MLR region are described. Consequently, the MLR locus is located outside the RhL–A region proper, possibly adjacent to the locus defining the antigens of the 1st series. The recombination frequency between the MLR and RhL–A regions of rhesus monkeys is similar to that found for the analogous systems of man.Inhibition of MLR can be achieved by pre‐incubation of responder or stimulator cells with cytotoxic anti‐RhL–A sera. Such inhibition is highly specific and probably caused by an interaction of the sera with RhL–A surface antigens, not with MLR determinants. Yet, there is circumstantial evidence that non‐cytotoxic sera can be produced which inhibit MLR, presumably by affecting MLR determinants on the cell surface.The relevance of MLR to histocompatibility and the genetic control of the immune response of rhesus monkeys to synthetic antigens, are the subjects of current studies which are

Cattle alloimmune sera produced against whole blood and isolated leucocytes were cytotoxic to human lymphocytes. After absorption with human erythrocytes, some antisera exhibited differential cytotoxicity on human lymphocytes. In addition, normal sera from parous cows also exhibited differential human lymphocytotoxicity, whereas those from heifers and bulls did not, indicating that antibodies were probably produced by foetal‐maternal alloimmunization as in multi‐parous women. Most of the activity of the cattle sera on human cells was associated with the HL–A system, though family studies indicated the possible existence of some non HL–A activity. The majority of the cattle serum reactions seemed to correspond to broadly cross‐reacting HL–A activities, associated especially with 4a and 4b. Some sera, however, showed considerable specificity. A system is described which rapidly detects antibody‐mediated cell‐dependent immunity in multiparous women; their purified peripheral lymphocytes act as effector cells against51Cr lymphocytes as targets, in the presence of specific antibody. A Danish family with twelve members and with two first cousin marriages was studied. Two inbred siblings (designated J) were found to be homozygous for HL–A2,27 (AJ) and therefore also presumably homozygous for all or most of the major histocompatibility complex. When used as stimulators in one‐way MLC, these two individuals elicited a very low response in five members of the family possessing this same HL–A haplotype, which was clearly disjunct from the response by family members not possessing the HL–A2,27(AJ). When J cells were used as stimulators against cells from 27 unrelated individuals, the responses against J – expressed relative to the median response of a given responder against several unrelated persons – fell into two clearly disjunct groups. One group of seven gave low responses, from 13% to 41%. The individuals of this group are designated J‐like. The rest gave responses from 83 % to 132 %. In one family with three children where the father was J‐like and the mother was not J‐like, the trait J‐like segregated in coupling with the HL–A determinants. It is postulated that these specifically low responding, or J‐like, individuals possess the MLC allele j, for which the individuals J are homozygous. Among the 27 unrelated individuals tested, there was no obvious association between the MLC allele j and any particular HL–A antigen. Another inbred HL–A homozygous individual did not stimulate the family members at a low level in compatible combinations and is thus presumably not

A study of seven pig families enabled the identification of individuals who were homozygous for the gene products identified by the mixed lymphocyte reaction. Homozygotes were engaged as markers to elucidate histocompatibility phenotypes. Thus a system evolved in which histocompatibility phenotypes were assessed independently of serologically dependent techniques.
