Abstract:Binding of immunoglobulins to major histocompatibility complex (MHC) molecules was demonstrated by two different assays: the binding of IgG to B cells by flow cytometry, and purified MHC antigens with an Elisa assay. Fc fragment from immune‐complex binds to the Fc receptor on B lymphocytes. Here, Fab was also shown to bind to B cells. This binding was inhibited by specific human alio anti‐HLA Class I and II sera directed at the polymorphic sites. Thus, in addition to the Fc receptor, MHC can also serve as a binding site for IgG. In an Elisa assay using purified antigens, IgG was shown to bind to HLA Class I and II molecules. Other proteins such as transferrin, human serum albumin, gelatin, etc., did not bind to the MHC proteins. Immunoglobulins bound to MHC molecules by sites on the Fab fragment independent of the hypervariable region. This was demonstrated by the retention of antibody activity even after binding of antibody (anti‐lactoferrin) to MHC. The relative avidity between Fab and HLA Class I and II was 4‐8

Abstract:To obtain a better estimate of major histocompatibility complex (MHC) polymorphism in the Old World macaque,Macaca fascicularis, class I and class II MHC proteins from 42 animals were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS‐PAGE) and one‐dimensional isoelectric focusing (1D‐IEF). The panel represented both related and unrelated animals with a total of at least 30 serologically distinct haplotypes. Cells were sequentially immunoprecipitated with monoclonal antibody (mAb) W6/32 for class I and with mAb L243 for class II molecules, followed by neuraminidase treatment. Both sets of immunoprecipitates yielded 5‐7 major bands on IEF. All bands present in offspring were present in at least 1 parent. Siblings which were serologically identical for class I and which were non‐stimulatory in mixed lymphocyte culture (MLC) yielded identical IEF patterns for both class I and class II; in other sibling pairs which were serologically identical for class I antigens (Ag), IEF produced convincing evidence that the siblings were indeed nonidentical, or helped to verify that recombination had occurred within the MHC. Specific bands were found which correlated with class I specificities A8, A24, and B25 previously defined by serology. Abstract:The analysis of HLA‐DR5 haplotypes unravelled a new DRB3 polymorphism and permitted the identification of various associations between alleles of the DRB1 and DRB3 loci. This new polymorphism consists of a 10.5 kb Taql restriction fragment which was encountered in an African‐American family (JS). In Caucasoids, the DRwll allele has been previously observed only in association with the DRw52b allele. RFLP and oligonucleotide typing of HLA‐DRw52 alleles associated with DRwll showed, however, that 4 Caucasoid individuals from our panel carried the DRw52a allele and 1 the DRw52c allele. Similarly, DRwl2, which is usually associated with DRw52b, was encounterd with DRw52a in 1 Chinese and with DRw52c in an African‐American and a Chinese panel member. The study of DRB3 alleles associated with DRwll and DRwl2 indicates that, similar to serology, RFLP studies become particularly informative when individuals of different races and ethnic origins are

Abstract:The DR7‐associated Dw specificities, Dw11, Dw17 and DB1 were investigated with regard to DRB‐ and DQB‐gene polymorphism, as revealed by RFLP analysis using the restriction enzyme Taql. In the 22 DR7 homozygous cell lines investigated, each of these Dw specificities was found to correlate to one specific RFLP defined DR‐DQ haplotype. In addition, a clear linkage disequilibrium to a specific HLA‐B locus allele for each Dw specificity was noted, indicating that the Dw subtypes of DR7 often are associated with a conserved HLA‐B‐DR‐DQ haplotype. Only one genetically homozygous cell line, PLH, deviated from these correlations. This cell line, notably derived from an individual with a deletion of the 21‐hydroxylase B‐gene (21‐OHB), carries the HLA haplotype Bw47, DR7, DQw2, DB1, but displayed a DRB RFLP otherwise found in

Abstract:HLA antigens of black North Americans are not as well‐characterized as those of other ethnic groups. We present results from HLA typing of black Americans using the 10th International Histocompatibility Workshop (IHW) sera. 1502 black Americans are typed for HLA‐A, ‐B antigens. 128 black Americans are typed for HLA‐DR, ‐DQ antigens. New specificities and splits established by the 10th IHW are identified in our data. Antigen frequency, gene frequency and haplotype frequency are computed for each phenotype observed. Haplotype A30 ‐Bw42 ‐ DRwl8 ‐ DQw4 and haplotype Aw36 ‐ Bw53 in strong linkage disequilibrium is characteristic of this black population. Abstract:The human trophoblast‐derived JAR cell line, that does not express polymorphic HLA class I antigens even after IFN induction, can be stably transfected by genomic clones encoding the entire HLA‐A2, ‐A3 and ‐B7 α‐chain genes. The transfected genes were expressed at the cell surface in association with endogenous β2‐microglobulin (shown by FCM analysis) as a single allelic product without reexpression of any endogenous class I gene (shown by 1D.IEF analysis). Furthermore, TNF‐α and IFN‐γ, alone and synergistically, increase cell surface expression of transfected MHC class I/endogenous β2m heterodimers without induction of endogenous class I α‐chain genes. These data show that the MHC class I‐negative JAR human cell line might be used for transfections with the aim of establishing human cells expressing just one defined MHC class I allele for functional and regulatory studies. These findings are discussed in relation to the methylated status solely of endogenous class I α‐chain genes in JAR cells and suggest that transfected class I genes are not regulated in the same fashion and, in particular, that constitutive and TNF/IFN inducibletrans‐actingregulatory factors able to bind tocis‐promoter/enhancer sequences of clas

Abstract:Thirty myasthenia gravis (MG) patients (9 with thymoma, 12 with thymus hyperplasia and 9 with thymic atrophy) and 181 Norwegian healthy controls were serologically typed for HLA‐A, ‐B and ‐DR antigens and genomically typed for HLA‐DQA1 and HLA‐DQB1 alleles by probingin vitroamplified DNA with sequence‐specific oligonucleotides. In patients with thymus hyperplasia the frequency of the DQB 1*0201 allele was increased compared to controls (RR = 3.5, p<0.05), whereas among the patients with thymoma this allele was not observed (RR = 0.06, p<0.01). The frequencies of HLA‐B8, ‐DR3 and ‐DQA1*0501, which are in strong linkage disequilibrium with DQB 1*0201, were increased in patients with hyperplasia and reduced in patients with thymoma.
