On the basis of studies with 6 HTCs in four different families the new cluster DB3 identified in the 8th Workshop can be considered an established specificity of the HLA‐D series. In 112 healthy Caucasoids resident in South East England the frequency of this antigen was 2.6% similar to the frequency found in 142 Caucasoid patients with rheumatoid arthritis. The frequency in 54 unrelated Chinese individuals from Shanghai and of 120 Nigerians was 7.5% and 3%, respectively.All Caucasoid individuals who were DB3 were also DR4 whereas no such association was found in the two other population group

Cross‐reactivity between antigens of the HLA‐B7 cross‐reactive group (B7 CREG) was investigated by the serological analysis of 60 “broad” cytotoxic HLA antisera produced by pregnancy alone, the HLA typing of the antiserum donors and the identification of their immunizing antigens. Thirty‐five sera, made in response to B7, covered (as a group) HLA‐B7, B27, Bw42, Bw48, Bw54, Bw55, Bw56, Bw60 and Bw61 — the B7 CREG antigens. Bidirectional cross‐reactivity occurred between the B7 antigen and B27, Bw55, Bw56 and Bw60 antigens but not between the major B7 CREG antigens B27, Bw22 and B40. HLA‐B27 stimulated antisera included B7, B13 and Bw47 within their reaction range and Bw55/Bw56 cross‐reacted with Bw42, Bw54, Bw57, Bw58, Bw62 and Bw63. Bidirectional cross‐reactivity was observed between Bw55 and Bw57. Fourteen responders possessed an antigen cross‐reactive with their immunizing antigen. Seventy‐six North African patients (most from Algeria) affected with nasopharyngeal carcinoma (NPC) have been studied for their HLA‐A, ‐B and ‐DR phenotypes and compared with a control North African population.Antigens HLA‐A3, HLA‐B5 and HLA‐Bw15 were found more frequently in the NPC group than in the control group (30.3% vs 17.6%, 38.2% vs 24.4% and 9.2% vs 0.8%, respectively). HLA‐Aw33, HLA‐B14 and HLA‐DR4 were less frequent in the patients than in the controls (3.9% vs 16.8%, 1.3% vs 16% and 13.2% vs 29.1%, respectively). After correction for the number of specificities tested, these differences were not statistically significant. They were, however, more striking when compared to normal Kabyles (Algerian Berbers), a major ethnical population in Algeria, with lower incidences of the HLA‐B5 antigen and of the HLA‐Aw33‐B14 haplotype. This could suggest, in North Africa, either the existence of MHC‐linked genes of resistance or susceptibility to NPC, in Berbers especially, or a preferential occurrence of NPC in non‐Berbers.Antibody titers against the Epstein‐Barr virus (EBV) associated early antigen (EA) and viral capsid antigen (VCA) have been measured. Forty‐one patients with Hashimoto's thyroiditis (HT) and 82 with Pernicious anaemia (PA) were investigated. All 123 patients were HLA‐D typed and results correlated to thyroglobulin antibodies (TgAb), microsomal antibodies (MAb), parietal cell antibodies (PCA), circulating immune complexes (IC), and intrinsic factor antibodies (IFA).In PA, TgAb was found less frequently in Dw2 positive patients than in Dw2 negative patients. IFA was rarely found in Dw5 positive PA patients. In HT, patients positive for Dw5 had lower levels of TgAb. IC were present in 67% of patients with HT, but only in 2.5% of patients with PA (p<0.01). Dw5 was associated with low levels of IC in HT.In conclusion, HT and PA seem to be related by their association with HLA‐D types, but a heterogeneity in the pattern of antibodies and IC could be seen. Linkage was investigated between a dominant gene determining familial polyposis coli (FPC) and HLA in a large pedigree. A tight linkage was excluded at a value of the recombination fraction between 0 and 5%. Linkage studies with various markers should be pursued to permit detection of high risk individuals and to better understand the phenotypic variability observed in certain polyposis families.

The HLA‐D/DR region in man encodes major determinants which stimulate T lymphocytes to proliferation. The genetic organization of this region is apparently complex and is at present largely unknown. One obstacle is the scarcity and quality of available typing reagents. In an attempt to obtain high quality anti‐DR sera, a series of active immunizations was performed between highly selected, healthy unrelated donors and recipients.One recipient (AR8) was immunized using cells incompatible for HLA‐A2, B40 (w60), Cw3 and DIDRw6 and readily developed anti‐A2 and B40 antibodies but no anti‐C, DR, or other antibodies. When tested against his HLA genotypically fully identical brother using the cellular MLC, PLT, or CML techniques before immunization, results were mutually negative as expected. Following immunization, however, AR8 was able to mount MLC, PLT, and possibly CML responses against lymphocytes from the brother while the reverse combinations remained negative. When tested in the family the trait(s) thus identified seems to be maternally inherited.These results suggest the existence of minor histocompatibility determinants encoded from regions not closely linked to HLA. Reactivity of two Cw5 and three Cw7 antisera was compared in the standard NIH and the Two Colour Fluorescence (TCF) techniques. Contrary to the NIH, the TCF method yielded strong positive reactions which enabled clear assignment of the Cw5 and Cw7 specificities. The TCF technique combines three factors that increase the sensitivity of serum reactions:1. typing on B lymphocytes, 2. prolonged incubation and 3. staining of the lysed cells with ethidium bromide.
