Forty mothers who had given birth to children with a clinically probable diagnosis of allo‐immune neonatal thrombocytopenia were typed for ABO, HLA—ABC and Zwaantigens and their sera were investigated with the platelet complement fixation microtest (CFT), the lymphocytotoxic test (LCT), and the platelet suspension immunofluorescence test (PSIFT). Seventeen (43%) of the 40 mothers were Zwa‐negative, which is significantly (p>10‐10) much more frequent than in controls (2.5%). Anti‐Zwaantibodies could be demonstrated by PSIFT in most of the Zwa‐negative mothers investigated, but in no cases by CFT. The only serum positive by this technique also contained HLA‐antibodies demonstrable by LCT. Lymphocytotoxic antibodies were found in 33% of the cases and this is not different from the frequency observed in normal pregnancies. In ten cases, antibodies of unknown specificity were only detectable by PSIFT. Nine of these mothers were Zwa‐positive and these antibodies may be directed towards platelet specific antigens different from Zwa.Of the Zwa‐negative mothers, 65% were HLA—B8 positive which is significantly (corrected p = 0.02) different from the frequency (25%) of this antigen in controls. This observation definitely confirms a preliminary observation in a small material. In the 23 Zwa‐positive mothers, a borderline increased frequency (43%) of HLA—B8 was found.ABO compatibility between mother and child was not found more often than expected.It is concluded in this study that anti‐Zwaantibodies may account for about half of all cases of AINT, and that PSIFT is a useful method to detect both these antibodies and perhaps other platelet specific antibodies causing AINT. The increased frequency of HLA—B8 in the mothers may reflect the operation of an HLA—B8 linked immune response determinant in the mothers, but may also be explained by the action of an HLA controlled complement variant facilitating destruction of antibody‐coated platelet in the infant.Studies to investigate thes

The frequencies of 12 DLA‐D alleles in a random canine population were determined in one‐way mixed lymphocyte cultures using a panel of homozygous typing cells established in this laboratory. The homozygous typing cells served as stimulators for responder lymphocytes obtained from 160 random dogs. The results of these studies were compared to those with lymphocytes from 75 dogs in our research laboratory. DLA‐D allelic frequencies were estimated by maximum likelihood techniques. The use of a relative response (RR) ≫5% as a definition of a typing response resulted in the recognition of a total allele frequency of 59% in dogs from the research laboratory. Three of the 12 DLA‐D alleles were not detected. Typing responses of cells from random dogs to the 12 DLA‐D alleles were determined using RRs °5%, °10%, °15%, and °20%. With RRs of °5%, °10%, and °15%, the total allele frequencies recognized were 39%, 47%, and 55%, respectively. Within each of these %RR ranges all but one of the DLA‐D alleles were detected. With an RR °20% the total allele frequency recognized was 58% and all 12 alleles were detected. Our results indicate that an RR of °10% could be used to define a phenotypic DLA‐D typing response in the dog. The level of allelic frequencies detected in both the research and random canine populations indicates the need to identify additional DLA‐D alleles through expanded family studies using mixed lymphocyte culture

52 symptomatic (SPB) and 64 asymptomatic (APB) pigeon breeders were investigated for the HLA‐A,‐B,‐C,‐DR; C2,C3,C4 and Bf systems, and C3, C4 and factor B serum concentrations. HLA‐DR3 and Bf S frequencies were significantly higher in the SPB than the APB group. Mean factor B concentrations were lower in the SPB than in the APB group. A positive two‐way association between HLA‐DR3 and the disease was found. Investigations for the HLA‐A, ‐B, ‐C and ‐DR antigens were conducted on 124 random North Indian patients with confirmed diagnosis of pulmonary tuberculosis by the demonstration of acid fast bacilli in the sputum. 109 appropriately matched controls from the same ethnic background were also tissue typed. No significant deviation was observed in the HLA‐A, ‐B, and ‐C locus antigens. With the HLA‐DR typing, there was a marginal increase in DR2 and a concurrent significant decrease in DRw6 in the patient group. These deviations were, however, insignificant when correction for thePvalue was made. ABO blood group typing results indicate that blood group ‘O’ may afford protection against TB. An increased prevalence of ankylosing spondylitis and other HLA B27‐associated rheumatic diseases has recently been demonstrated in a group of men with permanent pacemaker‐treatment. The purpose of the present study was to find out if HLA B27 was associated with severe bradyarrhythmias also in the absence of rheumatic disease.The frequency of B27 was determined in 83 permanently paced men with complete heart block, in whom presence of radiological or clinical signs of a B27‐associated rheumatic disease had been excluded. Eighty‐four healthy subjects were HLA typed for comparison.HLA B27 was found in 17% of the patients and in 6% of the controls, a significant difference withP= 0.017 (Fisher's exact test).
