The polymorphism of the HLA system has been defined by alloantisera, monoclonal antibodies, MLC reactivity, protein chemistry and RFLP patterns in DNA analysis.Typing for the alleles of HLA‐DR at the DNA level as an additional typing technique is useful since any nucleated cell can be used. Moreover, it is not known whether the additional polymorphism found at the DNA level in an unambiguous serotype is of functional importance and thus needs to be included in HLA‐DR typing.A main problem in DNA typing is the interpretation of the complex patterns in Southern blot analysis, especially in heterozygous individuals. Therefore we constructed subprobes from full length DRβ, DQα and DQβ cDNA probes to reduce the number of hybridizing fragments while retaining the discriminating capacity. The clearest differences among DR alleles have been found using the restriction enzyme PvuII and the subprobe containing the 3’untranslated region of the DRβ probe. Although further characterization is necessary to be able to type at the DNA level, the simplified patterns facilitate DNA typing in heterozygous individuals for a number of haplotypes. Interestingly, the number of fragments thus obtained corresponds with the number of genes described for DR1 to DRw8 haplotypes.Based upon the finding of common hybridizing patterns in DR3, DR5 and DRw6 it may be concluded that DR3, DR5 and DRw6 have been evolved from a common ancestor. Multiple polymorphisms on the DQ molecule(s) have been detected by monoclonal antibodies (MoAbs). Among these, TA10 and IIB3 have been described as two new alleles in the DQ region other than the conventional DQw1‐w3 allelism. The TA10 specificity is DQw3‐related and is in linkage disequilibrium with DR5 and weakly associated with DR4. The IIB3 specificity is DQw1‐related and is in linkage disequilibrium with DR2, DR4, DRw9 and DRw13. Thus, the DQw3‐carrying haplotypes are either positive with TA10 or IIB3. The molecular and topological analysis has revealed that both TA10 and IIB3 determinants were expressed on a single DQ molecule that also carried the DQw3 determinants on DR5 and DR4 cell lines, respectively. Thus, a single DQ molecule generated multiple epitopes detected by alloantisera and/or MoAbs at least on the DQw3‐carrying haplotypes. HLA‐A, B, C, DR antigen frequencies and Properdin factor B (Bf) allotypes were studied in a group of 44 patients with rapidly progressive periodontitis. HLA‐A9 (A24) was the only antigen with a frequency statistically significantly different from the control population. An increased frequency of HLA‐A9 was previously reported in periodontal diseases. We report the production of a lymphoblastoid human B cell line, which secretes monoclonal antibody with HLA‐DRw53‐like specificity. The immune lymphocytes were obtained from a patient who had been undergoing immunotherapy at Memorial Hospital with allogeneic cultured melanoma cells. The antibody producing cell line was established by transformation of immune B cells with Epstein‐Barr virus and stabilized by serial selection of antibody‐positive cell clusters. The cell line (R39.14) has been producing a cytotoxic antibody of IgM, kappa type during nearly two years of continuous culture. The specificity pattern was determined on a panel of HLA‐DR‐typed lymphoblastoid cell lines, and resembled the HLA‐DRw53 specificity closely. In studies of informative families the R39.14 specificity segregated with DRw53‐bearing haplotypes, which included DR‐4, 7 and DRw9 antigens. The R39.14 specificity was found to be expressed in similar quantities on all antibody positive homozygous cell lines tested. In this paper the first MHC data including HLA‐A, B, C, DR, DQ and complement BF, C4A, C4B determinants in German narcoleptics are presented together with the first family studies in European Caucasoids. 57 out of 58 unrelated patients (98.3%) were positive for DR2 and DQw1, respectively. In contrast to all other reports, one patient with typical signs of narcolelpsy was found to be DR2/DQw1 negative. Data showing significant increase in the frequency of B7, and normal frequencies of B35 were discordant with data from Japanese patients. Definition of the extended DR2 linked haplotypes, deduced from 6 families, revealed that 5 out of 12 were DQw1, DR2, BFS, C4B1, C4A3, B7 (Cw7), while 11/12 had DR2, DQw1, BFS, C4A3, C4B1 in common. In one multiple case family two genotypically different DR2 haplotypes were identified in affected siblings. The ability of mouse monoclonal antibodies (MoAbs) directed against HLA‐class II molecules to mediate in Antibody Dependent Cellular Cytotoxicity (ADCC) was investigated. The results indicate that both MoAbs to monomorphic and polymorphic HLA‐DR and DQ determinants are able to mediate ADCC in an antigen specific manner. However, not all antibodies mediate ADCC to a similar extent. Furthermore, antibodies were identified that appeared to mediate ADCC in an HLA‐DR haplotype dependent fashion. The occurrence of HLA‐A, ‐B, ‐C antigens and the HLA controlled complement factors (Bf, C2, C4) was investigated in an unselected group of 116 consecutively admitted patients with intracranial saccular aneurysms, and compared to that of healthy controls (blood donors). When multiplying the p‐values with the number of comparisons made, none remained significant. However, a rather high etiologic fraction of the BfS gene (0.59) was obtained. Moreover, for HLA ‐B7 a significant deviation from the Hardy‐Weinberg equilibrium with an increase of homozygotes was found. Due to linkage disequilibria this could indicate a strong association between HLA‐DR2 and saccular aneurysms. The presence of HLA‐DR2 was therefore investigated in a series of 15 aneurysm patients used as cadaver kidney donors and not included among the 116 consecutively admitted patients. In this group the HLA‐DR2 antigen frequency was significantly increased (66.7% vs. 29.7%, p<0.01). The present study thus demonstrates an association of saccular aneurysm and the major histocompatibility complex and shows the existence of a genetic predisposition to

In this report, a new HLA‐B locus antigen is described (tentatively called POT).
