The ADCC test has been miniaturized to use 0.05 ml of whole blood per test as ADCC effectors. A linear dose response was shown by dilutions of 0.05 ml of blood down to a 1:16 dilution. ADCC effector activities of 40 patients with lung cancer and four with leukemia were markedly less than the activity of 200 normal subjects. Melanoma patients did not have comparable statistically significant loss of ADCC activity.

Celiac disease (CD), which occurs in Europe with varying frequency, is known to be associated with HLA‐B8 and even more strongly with HLA‐Dw3 because of the high linkage disequilibrium between these two antigens. After typing for HLA‐A, ∼B and ‐DR antigens in adult patients with CD, we can confirm the high prevalence of B8 (72.7%) and of DRw3 (63.7%). Studies of the expression of HLA and DR alloantigens on cultured human melanoma cells in comparison with those expressed on peripheral lymphocytes and B‐cells derived from the same patients have shown that the HLA‐A B, and C locus antigens expressed on the cultured tumor cells were consistent with those expected from the typing of periperal blood lymphocytes. Our data have demonstrated, for the first time, expression of HLA‐DR antigens on heated T cells (45*C, 1 h) and have confirmed the occurrence of these antigens on mitogen‐stimulated T cells.HLA‐DR antigens were detected by microlymphocytotoxicity employing defined and non‐defined anti‐HLA‐DR sera. A PLT cell was raised between a responder cell which carried the HLA‐D and DR specificities Dw3, 8; DRw3, 8 and a stimulator cell which was most likely homozygous for HLA‐Dw3, DRw3. Blymphocytes were shown previously to be killed at 5*C by some autologous and allogeneic human sera. We show here that such cold cytotoxins are directed against immunoglobulins on the surface of B lymphocytes. The activity is partially removed by passing serum through IgG‐coupled Sepharose and usually completely removed by passing serum through IgM‐coupled Sepharose. Activity is regained from the columns by acid elution and IgM inhibits the cytotoxicity of these eluates. 125I‐labeled eluates bind to IgG‐ and IgM‐coupled Sepharose beads and binding is inhibited by IgM and to a lesser degree by IgG thus showing a primarily more avid binding to IgM as compared to IgG. The distribution of HLA antigens was studied in 40 patients suffering from systemic lupus erythematosus. For loci A and B, increased frequencies of Al, B8 and the presence of only one B antigen were found. Nevertheless, in relation to the number of antigens tested, this increase was not significant. For the DRw locus, DRw3 was significantly increased (P>0.001, after correction for the 39 antigens tested). However, the patients with DRw3 did not show any correlation with a specific clinical picture or the presence in their serum of lymphocytotoxic antibodies or autoantibodies against T and B lymphocytes.
