The fate of split thickness skin allografts between and among major histocompatibility complex (MSLA) homozygous herds of miniature swine has been examined. Among animals matched for all known blood groups, MSLA‐identical skin grafts survived 11.8 ± 0.89 days, while skin grafts from animals differing for one or two haplotypes survived 7.0 ± 0.36 days. Cytotoxic antibodies invariably appeared in the serum of animals rejecting MSLA‐nonidentical grafts, and were never detected among animals rejecting MSLA‐identical grafts.Among the homozygous DD herd, it was possible to assess the effect on skin allograft survival of a defined difference at the A‐O blood group locus. Skin grafts between DD animals identical at the A‐O blood group locus. Skin grafts between DD animals differing at this locus survived 7.66 ± 0.54 days. Blood group O animals rejecting skin from blood group A animals produced a low liter of antibodies which were cytotoxic for donor lymphocytes. These antibodies were at least partially absorbable by donor red cells. An absorption analysis using other blood group‐defined animals of these herds showed that ability to absorb activity segregated with the A blood group.These studies confirm the expectation that MSLA is a major histocompatibility complex of miniature swine, having important implications for the outcome of allografts. Using a microcytotoxicity assay, the serological reactivity of human granulocytes, namely neutrophils and eosinophils, and chronic myeloid leukemia (CML) cells and cultured CML cell lines (K562, NALM‐1) were examined. Mature granulocyte forms and cord granulocytes are readily lysed by specific granulocyte cytotoxins that do not react with random T and B lymphocytes, monocytes, red blood cells, or platelets. Furthermore, certain antisera were preferentially cytotoxic for eosinophil‐enriched populations. Granulocytotoxins detected antigens on one of three CML blast cell populations tested and K562, but failed to react with NALM‐1. By cytotoxicity, mature granulocytes were poor targets for B2‐microglobulin and the appropriate HLA antisera although both sera types are absorbed with granulocytes. Furthermore, granulocytes did not possess B‐lymphocytes (la‐like) or blood group A, B, and Rh (D) antigens. The predictability of MLC non‐reactivity by HTC typing was tested in a single checkerboard experiment which involved 39 unrelated individuals belonging to 12 different groups of HLA‐D identical phenotypes. The strength of one‐way MLC reactions in all possible responder‐stimulator combinations (39 × 39) was quantitated by the Linear Clustering Analysis Program. Individuals who, by HTC typing, were identical for two HLA‐D antigens gave 51% negative, 36% intermediate and 13% strong positive MLC responses. Identity for only one HLA‐D antigen resulted in 3% negative, 40% intermediate and 57% strong MLC reactions. When no HLA‐D antigen was shared, 88% of the reactions were strongly positive. HLA‐D antigens behaved as equipotent stimulators in MLC between half‐identical pairs. The only exception consisted of the higher frequency of weak responses displayed by the Dw5 positive individuals against stimulators differing by LD107. The same pattern was observed when LD107 homozygous cells were used as stimulators, suggesting that determinants of this specificity might be partial

HLA locus A and B typing was performed on 86 Chinese thyrotoxicosis (Graves' Disease) patients and 238 normal Chinese subjects. The frequency of HLA‐Bw46 (Sin 2) was found to be significantly higher among the patients than controls (X2= 26.15, correctedP<.003, relative risk = 3.74). Ten antisera containing antibodies directed against specificities controlled by the Ia, locus of rhesus monkeys were tested for their ability to inhibit the MLC reaction. In cultures with responding cells from the serum producer, all sera with anti‐Ia1activity depressed the stimulatory capacity of cells carrying the Ia1antigen in question. In other responder/stimulator combinations, the stimulation‐inhibition was Ia1‐related for the majority of the antisera. These data provide further evidence for a possible identity (or close association on the cell membrane) between Ia1and D locus determinants of rhesus monkeys. In addition, some sera showed an inhibitory effect also on responder cells (with cells from the serum producer as stimulators).
