摘要:
The incidence of cytotoxic antibodies in relation to histories of abortions was analyzed in sera from 474 nulliparous pregnant women. In patients with previous spontaneous abortions, there is a higher incidence of positive sera than in primigravidae. Furthermore, patients with two or more prior abortions show a greater incidence of cytotoxic activity in their sera than women who have had only one abortion.

The immunoglobulin class of circulating immune complexes (IC) was examined in 35 patients with Behcet's syndrome (BS) according to the HLA phenotype of these patients. Cold precipitable complexes were separated and assayed by laser nephelometry and polyethylene glycol precipitable complexes were assayed by rocket immuno‐electrophoresis for IgG, IgA and C3. The results suggest that in patients with BS, HLA‐B12 is associated with an increased proportion of IgG and a decreased proportion of IgA and C3 in their immune complexes, whilst the converse was found with HLA‐B5 or DRw2. The ratio of IgA:IgG in immune complexes was therefore low in patients with HLA‐B12 but high in those with HLA‐B5 or DRw2. Linkage disequilibrium has been described between HLA‐B antigens and allotypes of properdin factor B (Bf). The association between HLA‐B and DR antigens and the rare allotype BfF1 was investigated. In 27 HLA‐A, B and DR typed individuals only one person was found who was DR3 negative but five who were B18 negative. Family studies showed that in nine cases B18, BfF1 and DR3 segregated as a haplotype; in another family which was B18 negative, BfF1 and DR3 segregated together. Lymphocytotoxic antibodies reacting with husband's lymphocytes were demonstrated in 24 sera from women with trophoblastic neoplasia studied before any chemotherapy.These antibodies exhibited a maximal cytotoxic activity at +4°C. They reacted mainly with B lymphocytes as assessed by microlymphocytotoxicity assays using B‐or T‐cell enriched subpopulations and T‐or B‐cell lines as targets. They could not be absorbed out with platelets or erythrocytes, and did not react with autologous lymphocytes.In one double‐labelling immunofluorescence experiment, these antibodies could be stained by rhodamine‐conjugated Fab'2 anti‐μ fragments and thus appeared to belong to the IgM class. They were shown to react mainly with surface Ig (SIg) bearing lymphocytes, plus a minor SIg negative subset.Studies with panels of allogeneic normal B cells and of HLA homozygous B‐cell lines showed that the target antigen(s) recognized by these antibodies is clearly distinct from HLA‐D antigens. Corresponding antigens seem to be expressed on placenta, since the lymphocytotoxic antibodies could be absorbed out with trophoblastic homogenates and one serum reacted with JAR cells (cultured choriocarcinoma line).An enhancing role of such antibodies in the growth of trophoblastic malignanci

Lymphocytes from uremic patients showed statistically significantly lower (P<0.01) MLC reactivity against pooled stimulator cells as compared with lymphocytes from age matched controls. A consistently low MLC reactivity was recorded in lymphocytes from patients who had received more than 20 units of blood and the lowest responses were seen with cells from patients with multispecific anti‐HLA‐antibodies. Removal of B cells and removal of phagocytic cells from the responder cells resulted in a significant (P<0.05) increase in MLC reactivity in the uremic but not in the control group. Both addition of enriched B cells to autologous enriched T cells and addition of adherent cells to non‐adherent autologous cells caused a slight supression of MLC reactivity. PHA reactivity was lower (P<0.001) in lymphocytes from the multitransfused patients compared to cells from age matched controls. The anamnestic response by human bi‐directional (BD) mixed lymphocyte cultures (MLC) to restimulation by cells of the original stimulating type is generally strikingly reduced as compared to that of standard one‐way cultures. This difference was shown not to be related to a change in kinetics nor was it due to exhaustion of the media or soluble factors since fresh media did not ameliorate the effect nor were supernatants from BD cultures found to be suppressive. The relative inhibition was also not reversed by removal of the allogeneic cells by phenotype specific antiserum. Cytotoxic tests with donor and responder specific antisera revealed that the cells bearing that phenotype were dramatically reduced in BD as compared to one‐way cultures. Thus, the diminished secondary response appears to be due to cytotoxic elimination of the responder cells. This allogeneic cytotoxicity is dependent on non‐T, phagocytic, adherent cells. The diminished response to secondary stimulation of human lymphocytes primed in bi‐directional (BD) mixed lymphocyte culture (MLC) has been demonstrated to be due to cytotoxic destruction of the responder cells by the numerically superior allogeneic stimulating cell population. This phenomenon is called Functional Cell Mediated Lympholysis (F‐CML). Matching for HLA‐A and B, HLA‐DR or for MLC non‐responsiveness (HLA‐D) in unrelated pairs does not ablate F‐CML, indicating that none of these loci serve as the exclusive target for this activity. One locus appears to be centromeric from HLA‐A in an A/B recombinant family and a B/D recombinant family demonstrates a target centromeric from HLA‐B. A special family with homozygous for HLA‐A, B and D provided evidence for an additional locus other than HLA‐A, B or D. A panel of 79 individuals were typed for HLA‐D/DR associated Primed Lymphocyte Typing (PLT) defined “DP”‐antigens, HLA‐D and HLA‐DR antigens. Typing for DP‐antigens was carried out with local PLT‐cells. HLA‐D and‐DR typing was performed with all homozygous typing cells and all DR‐antisera included in the 8th International Histocompatibility Workshop. Assignments of DP‐, HLA‐D‐and HLA‐DR‐antigens were done independently and the correlations between DP/D/DR1–8 were analyzed. The panel included random unrelated individuals, and individuals previously found to have one or no identifiable HLA‐D antigen (B). In the random group, 80% of the individuals were assigned to possess the same antigen with the 3 techniques, while this was only the case in 46% of B‐group individuals. The overall correlation coefficients, r, for the antigens HLA‐Dw/‐DR/DP1–8 were 0.95 (DP/D), 0.94 (DP/DR), and 0.89 (D/DR). There is a remarkably strong correlation between HLA‐D and‐DR typing results concerning D/DR1–8, in particular in random individuals. It is possible to select PLT‐cells that give typing results which are almost identical to those of HLA‐D and‐DR typing. When discrepant results were seen, HLA‐DR was in general “broader” than DP which in turn was broader than HLA‐D, indicating that it may be
