Rhesus monkeys were immunized with partially purified HLA‐A, ‐B, ‐C and DR antigens. The resulting sera were shown to have activity against species‐specific determinants on both HLA‐A, ‐B, ‐C chains and β32 microglobulin by the use of somatic cell hybrids. When this was removed by absorption, the sera showed activity against three of the four HLA‐A and ‐B antigens in the immunogen when tested on a panel of peripheral blood lymphocytes and T cells. Antibodies recognizing HLA‐DR antigens were detected by testing platelet absorbed sera on a panel of typed lymphoblastoid cell lines. After absorption to remove activity against species‐specific determinants on the HLA‐DR antigens, two cross reacting specificities were defined. One consisted of a determinant in common between HLA‐DRwl, 2 and 6 and the other a putative determinant in common between HLA‐ DRw4, and 5. Red blood cell Mg was measured in 351 blood donors of known HLA antigens. The level of red cell Mg is significantly lower in subjects bearing an HLA‐Bw35 allele (P>0.001). It also varies according to the HLA‐Bw4 and Bw6 antigens of the subjects tested. These two effects seem to be independent of one another. Since HLA‐DR antigens are closely related to HLA‐D antigens, we studied the effect of low‐dose ultraviolet irradiation on the ability of human lymphocytes to induce a proliferative response in mixed lymphocyte culture (HLA‐D) and on the serologic reactivity of B lymphocyte HLA‐DR antigens. We found (1) no alteration in the serologic definition of DR antigens despite the abrogation of the induction of proliferative responses in mixed lymphocyte culture, (2) no evidence that ultraviolet light (UVL) stimulated suppressor cells contributed to the loss of allostimulation, and (3) no evidence that the abrogation of the induction of proliferative responses could be modified by increasing the number of UVL exposed lymphocytes in mixed lymphocyte culture. These findings suggest that HLA‐D and HLA‐DR antigens are different molecularly, but are also consistent with the concept that certain active metabolic processes are required of stimulator cells for allostimulation and that these processes are

Seventy‐two families, selected for having at least two children affected with leprosy, were HLA typed for 57 A, B and C locus antigens recognized by the WHO Nomenclature Committee. In addition, 20 possible new “splits” were investigated. The distribution of A, B and C locus antigens in affected and unaffected family members was similar, irrespective of the type of leprosy in the family. Gene frequencies (derived by direct gene counting from 253 haplotypes), haplotype frequencies and delta values were calculated. There is evidence for heterogeneity of B5, B15, B17, Bw16 and Bw35 and for the existence of at least one A locus and one B locus antigen not previously detected. An algorithm is presented for clustering antisera by computer. It has two novel features: the leading serum to which all other sera in the cluster are compared is chosen as the most centrally located serum in the cluster; the similarity between two sera is defined from the 2X2 table of serum reactions as s = 2a/(2a + b + c). This similarity index is abetter measure of the similarity between two sera than conventional measures of similarity such as the correlation coefficient. Finally, the identification of cluster and serum subsets provides a more complete analysis of cross‐reactivity and multispecificity, and suggests which absorptions might yield monospecific typing sera. HLA typing was done on 2,956 healthy, unrelated individuals living in four areas in Bulgaria. Of these, 163 had G6PD deficiency. The persons examined lived in villages situated at different altitudes. A low degree of correlation was found only for HLA‐Al, ‐Aw23, ‐Bw35 and ‐B40 in the selected group with G6PD deficiency. A total of 74 healthy unrelated random individuals and 36 patients with juvenile rheumatoid arthritis (JRA) were typed for HLA‐D antigens with the homozygous typing cell technique and typed for HLA‐D/DR associated DP‐antigens with the primed lymphocyte typing (PLT) technique. All patients and some of the controls were also HLA‐DR typed with a limited battery of anti‐DR sera. Selected PLT‐cells, specific for the HLA‐D/DR antigens D/DRwl‐8 and the local specificity D “H” were used. The results of the PLT‐experiments were evaluated with the Normalized Median Response (NMR) method and the further procedure of DP‐antigen assignment was analyzed. The DP‐antigen assignments could be done solely according the NMR‐values in approximately two thirds of the individuals. In the remaining individuals, further interpretation of the experimental data had to be done for the assignment of DP‐antigens. The correlation coefficients were estimated between the HLA‐D assignments and (i) the individual PLT‐cell NMR‐values with a fixed cut‐off for positive reactions and (ii) the DP‐antigen assignments. These coefficients were 0.79 and 0.92, respectively. The correlations between HLA‐D, ‐DR and DP‐antigen assignments of the specificities HLA‐D, ‐DR and DPI, 2, 3, 4, 7 and 8 were analyzed in 42 controls and 36 JRA patients. The total correlation coefficients were: (i) HLA‐D/DR: r = 0.78; HLA‐DR/DP: 0.77; and HLA‐D/DP: 0.96. Antibody secreting B cells were measured as plaque forming cells (PFC) in a hemolysis‐in‐gel assay using fluorescein isothiocyanate (FITC) coupled SRBC as targets or protein A coupled SRBC as targets and developing antisera. Peak antibody secretion occurred on day 5 and the highest number of PFC was seen when mitomycin treated stimulator cells: responder cells were used in a ratio of 1 A. The number of PFC in MLC was not correlated to the DNA synthetic response. Antibody secretion in MLC was found to be of IgM, IgG and IgA classes. Significant numbers of PFC in MLC from blood lymphocytes were detected with the protein A technique, but not using FITC‐SRBC targets. Compared to spleen cells, fewer PFC were stimulated in blood lymphocytes. B cells alone, enriched by rosetting of T cells, did not respond by antibody secretion or DNA synthesis in MLC.
