Abstract:Extended MHC haplotypes comprising HLA‐A, ‐B, ‐DR, C2, BF and GLOl loci observed in the parents of 497 Danish normal families are presented, with particular regard to the haplotypes that include BF variants or the C2*2 allele. The known association of HLA‐B35, ‐DR1 with both ‐A3 and ‐A11 appeared to depend upon the BF type: HLA‐B35, BF*S, ‐DR1 is strongly associated with ‐All, whereas ‐B35, BF*F,‐DRl is strongly associated with ‐A3.Further, in the present material DZ twins of the same sex shared HLA‐haplotypes more often than did

Abstract:A plan for DRB typing at the sequence level is detailed. Only one polymerase chain amplification reaction is needed and the application of a limited number of short oligonucleotide probes allows an almost complete definition of DRB alleles. The scheme was tested on 40 homozygous cell‐lines selected to cover a wide range of specificities, and 40 RFLP‐typed controls. The results are presented and discussed. Abstract:HLA typing was performed in 49 families, each containing at least one woman with a history of unexplained recurrent spontaneous abortion (RSA), and reproductive histories were recorded for the siblings. Abortion rate in sisters sharing two HLA haplotypes with the proband was significantly (p<0.01) increased, whereas abortion rates of brothers’wives seemed independent of the brothers’degree of HLA sharing with the proband. Birthweights of offspring of both the sisters and the brothers decreased with increasing HLA haplotype identity between the sibling and the proband (p = 0.05). The mean birthweight of infants of siblings sharing both parental haplotypes with the proband was 3158 g which was significantly (p<0.02) less than the average birthweight in Denmark (3417 g). Significantly (p<0.05) decreased mean birthweight in infants of brothers who were HLA‐identical with the proband suggested that the disposition to growth retardation could be inherited with HLA also through the male gametes. It has been shown that, in their few successful pregnancies, women with RSA bear infants with a birthweight which is approximately 300 g less than normal. The present study demonstrated that the two obstetrical conditions, RSA and retarded fetal growth, share common genetic markers: HLA. Abstract:We examined the polymorphism of DR2,DQw1 haplotypes in Epstein‐Barr virus‐transformed B‐lymphoblastoid cell lines (HTCs) and unrelated (32 Canadian Caucasians and 24 Asian Indians) individuals by restriction fragment length polymorphism (RFLP) and oligonucleotide typing. The data demonstrate that three subtypes of DR2,DQwl haplotypes, DRwl5(B1.1501).DQw6a(A1.0102,B1.0602), DRwl5(B1.1502). DQw6b(Al.0l03,Bl.0601), DRwl6(B1.1601).DQw5(A1.0102,B1.0502) are present in HTCs and Canadian Caucasians. Of these, DRwl5(Bl.l501).DQw6a (A 1.0102,B 1.0602) haplotype was present in majority (81.3%) of Caucasians. Among Asian Indians, this haplotype was present only in one DR2,DQw1‐positive individual. In addition, three new haplotypes representing different combinations of DRB1, DQAl and DQBl genes were demonstrable in Asian Indians. These new haplotypes are DRwl5(BI.1501).DQw6b(AI.0103,B1.0601), DRwl5(B1.1501). DQw5(A1.0102,B1.0502), and DRwl5(B1.1501).DQw6c(A1.0102, B1.0601). The most frequent haplotypes among Asian Indians were DRwl5(Bl.1502).DQw6b(A1.0103,B1.0601) and DRwl5(B1.1501). DQw6b(A1.0103,B1.0601). The distribution of subtypes of DR2,DQwl haplotypes in Asian Indians was significantly different from that in Canadian Caucasians. Abstract:In the 11th International Histocompatibility Workshop pre‐screening study, HLA Class I and Class I1 monoclonal antibodies (mAb) were tested on panels of peripheral T and B cells and homozygous typing cell (HTC) lymphoblastoid lines. The 102 Class I mAb, of which 64 were new, were screened on 63 HTC lines and 20 peripheral T cells. The 144 Class II mAb were tested on 63 HTC lines, 20 peripheral B and 10 peripheral T cells. For Class I, 45 of the 102 mAbs tested proved to have clear and identifiable reaction patterns on both PBL and HTC lines and included several interesting new HLA‐A and ‐C locus antibodies. Of the 144 Class II mAb, of which 99 were new, 18 DR, 22 DQ and 7 DP antibodies were judged excellent. The DR antibodies included some which were antigen‐specific but the majority were complex antibodies, while the DQ antibodies, as previously, identified the majority of DQ antigens both singly and in combinations.
