We have studied 25 patients with polyglandular autoimmune disease with respect to HLA antigens. Whereas the combination of insulin dependent diabetes with Graves’disease or atrophic thyroiditis was associated with an increase in HLA–B8, this was not found to be the case for patients with I.D.D.M. and goitrous thyroiditis. However 4/7 of these patients were DRw3 positive in contrast to previously established normal distribution of HLA–B8 in patients with goitrous thyroiditis alone. These data suggest that patients with polyglandular failure may be highly selected for HLA‐B8/DRw3 positivity.We also report on two families with polyglandular autoimmune disease; the results suggest that these disorders are not necessarily transmitted with B8/DRw3 bearing haplotypes. In one family both the affected mother and non‐affected father were B8 positive. The mother's B8, which was associated with DRw7, BfF and Rgadid not appear to be involved in the transmission of disease susceptibility to two affected offspring. The search for complete haplotypic arrangement should be pursued to see whether this uncommon haplotype arrangement is peculiar to autoimmune

Two monoclonal antibodies recognizing HLA–DRw (human Ia) antigens were produced. The DA2 antibody binds a monomorphic determinant, common to all specificities and Genox3.53 antibody binds to a cross‐reacting site on the HLA–DRw1, 2 and 6 specificities. Both antibodies are IgGl and show complement dependent cytotoxicity only in the presence of rabbit anti‐mouse IgG serum. Specificity of both antibodies for the HLA—DRw molecule was shown by inhibition of antibody binding by preincubation of antibody with detergent solubilized Ia from JY (HLA–DRw4, 6) cells and by preincubation of target cells with F(ab')2fragments of a rabbit anti‐Ia serum. DA2 antibody reacted with all cells of human B cell origin tested and with peripheral blood lymphocytes of several primate species tested. Genox3.53 antibody bound only to human cells expressing HLA–DRw1, 2 or 6 antigens, giving a negative reaction with all primates tested. Genox3.53 antibody detected a split in the HLA–DRw6 specificity, showing reduced binding to the Daudi cell (HLA–DRw6) in comparison with binding to several other cell lines typed as HLA–DRw6, under saturating conditions. This low reactivity with Daudi was confirmed by absorption experiments. The ratio of DA2 binding to Genox3.53 binding to homozygous and heterozygous cell lines under saturation conditions was compared. Results suggested that, on some cell lines, DA2 might be reacting with a second population of human Ia antigens in addition to the HLA–DRw antigens. When a mixture of saturating concentrations of DA2 and Genox3.53 antibodies was tested for binding to cells under saturating conditions, the number of counts bound suggested the antibodies could bind simultaneously. H‐2 alloimmunization evokes antibodies of differing cross‐reactivity patterns among individual mice of one inbred strain. We investigated whether this is due to unknown variations at the time of bleeding or whether it remains constant for an individual mouse during long‐term immunization. At various times during immunization, serum was collected from individual mice and it was tested on a panel of mouse and human lymphocytes. Sera from three donor‐recipient combinations were examined. In some sera, a broadening of the cross‐reactivity pattern occurred after prolonged immunization, but no major change in specificity was observed. We conclude that the differences in cross‐reactivity pattern among sera from individual mice remain constant during the immunization period and do not reflect variations in the condition of the animals at the time

Treatment of porcine valvesin vitrowith glutaraldehyde significantly decreases their hetero‐antigenicity, as judged by immune reaction in mice heterografted with glutaraldehyde‐treated valves or injected intraperitoneally with valvular homogenates.Conditioned porcine valves, grafted in a cutaneous pouch onto the backs of two different inbred strains (C57B1 and DBA) behave like inert material and show minimal cellular infiltration at histological examination. Little or no antibody response, demonstrable with the cytotoxity test is elicited by glutaraldehyde‐treated valvular tissue, either grafted or injected intraperitoneally as homogenate. HLA antigens of the A and B loci were determined on the lymphocytes of 30 patients with acute lymphocytic leukemia (A.L.L.), as well as all of their mothers and 26 of the fathers. Seven of the 26 parents shared a common haplotype. This incidence of 269 per 1,000 contrasts with an expected incidence of 90.7 per 1,000, calculated from haplotype frequencies in a North American population (x2= 7.61,P<0.01) and a frequency of one in 27 in parents of patients with renal failure in the local population (x2= 3.91,P0.10). This suggests that the genetic background of a large proportion of patients with A.L.L. has restricted heterogeneity, presumably leading to the increased expression of leukemia associated recessive genes.

We have recently described a “new” private HLA–DR antigen, DR“LTM”, which has a frequency of approximately 0.6% in Danes. Primed Lymphocyte Typing (PLT) cells directed towards DR“LTM”‐associated determinants were generatedin vitroby haplotype primings in two unrelated families with DR“LTM” positive individuals. Both PLT‐cells reacted in parallel and gave positive reactions with (i) two unrelated and (ii) eight related individuals, all of whom were DR“LTM”‐positive. Both PLT‐cells gave negative reactions with (i) 38 unrelated and (ii) 15 related DR“LTM”‐negative individuals. Thus, there was total agreement between the results obtained by HLA–DR typing with the antiserum “LTM” and by PLT‐typing with these two haplotype primed PLT‐cells. None of the DP“LTM”‐positive individuals carried more than one of the antigens HLA–Dw/‐DRw/DP1‐8 and the local specificity D/DP“H”.
