Aided in part by a grant from the Jane Coffin Childs Memorial Fund for Medical Research. A case is reported of a 34‐year‐old female with chronic thyroiditis, multiple upper respiratory ailments, and a monospecific anti‐HL–A8 lymphocytotoxic isoantibody occurring in her serum in the absence of prior pregnancy or transfusion. The antibody showed a rise in titer over several years of observation and was contained wholly within the IgM serum fraction, which fraction was mildly increased above normal values. Whether the antibody reflects cross‐reaction to exogenous antigens or a spontaneous monoclonal proliferative phenomenon, the case is of interest in relation to Jerne's theory of the generation of antibody

HL–A phenotype and gene frequencies are presented for a population sample of Nahua Indians from Mexico. These Indians showed restricted variability in the first and second segregant HL–A series as compared to Caucasian, Black and Mongoloid populations, thus resembling other American Indian populations. Determinants for HL–A2, HL–A9, W28 and W31 accounted for 95% of first series alleles. For the second series W16, W5 and W10 comprised 75% of all genetic determinants. This population sample is the only American Indian group so far studied with W16 as the most frequently occuring second series antigen. Disturbances in the Hardy‐Weinberg equilibrium for the second series may reflect racial admisture and an antigen which cross reacts with W5 and W15. When adjustments were made for sample size and selection of the extreme deviate delta value, no evidence of linkage disequilibrium was noted in this population. Thirty‐nine leprosy patients (20 tuberculoid and 19 lepromatous) have been HL–A typed and compared to 36 non‐leprosy individuals of the same ethnic group (Amharas). Similarity between human and chimpanzee alloantigens was investigated by absorbing human alloantisera with chimpanzee lymphocytes and testing the absorbed antisera with appropriate HL–A‐phenotyped human lymphocytes. Chimpanzee lymphocytes were consistently negative in cytotoxicity assays with monospecific human HL–A2, 3, 9, and 13 antisera. Chimpanzee lymphocytes failed to remove reactivity for these specificities in absorption studies. Positive cytotoxic reactions were observed with chimpanzee lymphocytes and anti‐HL–A1, 11, 12, and W27 human alloantisera. Absorption with chimpanzee lymphocytes removed reactivity removed reactivity for these specificities as shown by tests with human lymphocytes of the appropriate HL–A phenotype. Experiments were performed to establish optimal conditions for specific and non‐specific lymphocyte transformationin vitro, in cultures containing 1–3 × 105human lymphocytes. The stimulation as measured by14C‐thymidine incorporation was evaluated when using various methods of lymphocyte purification and different numbers of contaminating erythrocytes, different kinds of tubes, serum from different sources, and varying PHA concentrations.Furthermore, the dependence of the14C‐thymidine incorporation on the thymidine concentration in the medium and on the total number of lymphocytes present per culture was determined.Two methods of harvesting the cells to measure the14C‐thymidine incorporation were compared.On the basis of this analysis, conditions for maximal sensitivity and reproducibility could be defined in measuring lymphocyte reactivity to PHA, allogeneic lymphocytes and antigens in a miniaturized cult

The serum of a 12‐year‐old boy with CID, reconstituted by a bone marrow graft from an HL–A identical sibling, was studied for the presence of blocking and cytotoxic antibodies. After grafting, the patient's serum inhibited the response of autologous and of normal lymphocytes in the MLC. A non‐HL–A lymphocytotoxic antibody was detected at the time severe GVHR. It reacted with lymphocytes from the patient's father, mother, sisters, and with 70 % of unrelated allogeneic lymphocytes. It was not reactive with the donor lymphocytes, suggesting that it was formed against a non‐HL–A antigen present on the recipient tissues but absent on the donor cells. A population of 224 unrelated dogs was tested with 54 selected canine lymphocytotoxic sera in order to define histocompatibility antigens of the dog. Mickey's factor analysis program, which is able to determine different specificities contained in one serum, was used since most of the sera were multispecific. Groups of sera containing the same specificities were formed and used for the determination of transplantation antigens. Twenty‐three preliminary groups could be defined by statistical methods. Thirty‐five families and 33 litters were tested with the same sera to validate the established groups.
