Two well characterized Eighth International Workshop sera, 8w1090 and 8w112, described to have anti‐DR3 and anti‐DR5 activity, respectively, were tested by standard and antiglobulin augmented (AHG) complement dependent cytotoxicity (CDC) assays with 21 target cells. We demonstrate that both sera, in fact, have only one antibody (most likely anti‐MT2), but have different CYNAP patterns. The HLA‐B17 cross‐reactive group and the participation of the subdivisions of B17 (Bw57 and Bw58) in cross‐reactivity were investigated by the serological analysis of 81 cytotoxic HLA antisera (produced by pregnancy alone), the HLA typing of the antiserum donors and the identification of their immunizing antigens. The sera, all of which contained B17 activity, were produced in response to one of 10 HLA antigens (A2, Bw44, Bw49, Bw51, Bw55, Bw56, Bw57, Bw58, Bw62 and Bw63). Antisera stimulated by Bw57 and Bw58 cross‐reacted with Bw49 and both subdivisions of B5 and B15, with bidirectional cross‐reactivity occurring in many instances. Bidirectional cross‐reactivity was also observed between Bw57 and A2 (an A2 stimulated antiserum also reacted with Bw58), and Bw57 and Bw55. Immunization by Bw62 produced some antisera which showed strong cross‐reactivity with Bw57 but no reactivity with Bw58. Significant HLA‐B antigen frequency disturbances were found in the responders to the B17 cross‐reactive group antigens. Plasma samples from 129 dogs have been typed using an immunofixation technique to define the polymorphism of canine C4. The mode of inheritance was inferred by study of four large pedigrees. In contrast to the situation in man, canine C4 may be encoded by a single locus. At least five different co‐dominant structural alleles have been detected. Antibodies against Epstein‐Barr virus (EBV) antigens, i.e. the viral capsid antigen (VCA) and the Epstein‐Barr nuclear antigen (EBNA), were determined in two independent populations with known HLA‐DR phenotypes. The first population consisted of 151 patients with rheumatoid arthritis; the second one of 88 healthy parents of leukemic children. Although the group of patients with rheumatoid arthritis differed significantly in the frequency of 4 DR antigens from the second group, both groups had the same correlation between HLA‐DR antigens and the antibody response to EBV antigens. There was a significant correlation between HLA‐DR1 and reduced titers of antibodies to VCA, whereas the persons with only one identifiable DR antigen had higher anti‐VCA titers. Using a hybridoma technique, a monoclonal antibody (TA10) reactive with a new polymorphic B‐cell antigen was obtained. The TA10 antibody reacted with 12 (24%) of B cells from 50 healthy individuals, but not with T cells. In contrast with the HLA‐DR antigen, the TA10 antigen was preferentially detected on B cells, but not on monocytes. The reactive pattern of TA10 was closely associated with DR5 and its related DR antigen NJ2, and also found with DR4 or DRw9. The linkages of TA10 to these DR antigens were confirmed in the family study, indicating that the TA10 antigen is controlled by a closely linked but distinct locus from HLA‐DR. Furthermore, two‐dimensional gel analysis demonstrated that the TA10 determinant was expressed on a class II molecule whose heavy and light chains were different from those carrying the DR determinant. Human Purified Protein Derivative of tuberculin‐ (PPD‐) specific cytotoxic cells have been detected in cultures of peripheral blood mononuclear cells stimulated for 6 days with PPD. These cytotoxic cells are demonstrated by their ability to lyse PPD‐pulsed autologous monocyte target cells, but not unpulsed targets.In a series of checkerboard experiments each involving 3–5 randomly combined donors, effector cells from 35 donors have been tested in autologous and 130 allogeneic combinations. Analysis of results from the pooled allogeneic combinations reveals that HLA‐B — and even more pronounced HLA‐DR — antigen sharing correlates positively to high lysis. No effect of HLA‐A antigen sharing is found. A more detailed analysis shows that the effect of HLA‐B sharing may be fully accounted for byHLA‐B‐DRlinkage disequilibrium.
