Cohen Institute for Radiology and Radiation Protection, the Whitehall Foundation, and the Dutch Organization for Pure Research (FUNGO).The results of testing erythrocytes from eight mouse strains with 20 haemagglutinating antisera and five lymphocytotoxic antisera, using a Technicon Haemagglutinating Autoanalyser, are presented. The suitability of the autoanalyser to regular H–2 typing is proved and the adaptability of this methodology to an investigation of the haemagglutinating capacity of lymphocytotoxic antisera is demonstrated. Twenty‐one HL–A antigens of the first and the second HL–A series have been studied in a total of 122 German children with retinoblastoma and in 64 families. The frequency of antigen W5 was significantly increased (P<0.001) and that of HL–A12 was significantly decreased (P<0.001) in the patients than in healthy controls. The presence of W5 and the absence of HL–A12 may be among the factors influencing the penetrance of the retinoblastoma allele (hereditary form) or could in some way produce, together with several other unknown factors, an increased susceptibility to the disease, possibly through a defect in the immune response towards the tumor. Such correlations, however, must be very complex, since segregation analysis and family studies of patients with the hereditary and the nonhereditary form of the tumor failed to demonstrate clear‐cut correlations between HL–A antigens and the incidence

Forty‐nine of 127 normal sera from New Zealand white rabbits were found to contain cytotoxic antibodies against human lymphocytes. Reactions with these sera did not correlate with those of known HL–A or ABO specificities. However, four distinct specificities were found in 18 sera. These antibodies are labile upon prolonged storage at −20°C and heating at 56°C for 30 min and have low avidity. Since rabbit serum is the most efficient source of complement in the cytotoxic reaction with human lymphocytes, the quantity as well as the specificity of these natural antibodies in rabbit serum have to be considered when selecting this reagent for the lymphocytotox

We have studied the antibody binding capacity of mouse H–2 antigens after fixation with paraformaldehyde. Spleen cells and erythrocyte stroma fixed for one hour in 0.9% paraformaldehyde were found to bind anti‐H–2 antibodies to essentially the same extent as fresh material. This study concerns the persistence of antibodies in 42 sera. Twenty‐seven of these, sampled from 12 to 175 weeks after the first sample, remained positive. In general, the titre of the antibodies had decreased. A reproducible, well‐defined technique is described for measurement of lytic rate of lymphocytes in the presence of HL–A antisera. Using this technique, it has been possible to determine HL–A2 zygosity in related and unrelated people using both fresh and stored cells.
