The F antigens, which are serologically defined Class I gene products of the major histocompatibility complex in chickens (the B complex), were analysed in outbred birds. Private specificities of the F13 antigen from the inbred CC strain were detected in 20 outbred chickens by a haemagglutination technique. In the GVH‐in‐hibition‐release test F13 antigens from outbred and inbred chickens were identical. The L antigens, which are the Class II antigens of theBcomplex, were detected with specific anti L13 alloantisera by indirect immunofluorescence. Antisera defining the L13 antigen(s) of the inbred CC line reacted with all F13 positive out‐breds. As a test of one‐way direct compatibility of the inbred and outbred animals typing F13, graft versus host reactions were performed, injecting blood of F13 positive outbreds into inbred B13/13eggs. No GVH stimulation attributable to MHC determinants was found. In MLR, responder cells from outbred MHC heterozygous chickens, which typed F13/x, were stimulated by inbred F13/13 homozygous typing cells, and weak, but significant, reactions were found. Further analysis in the accompanying paper, however, revealed no difference in the lymphocyte activating determinants (Lads) of inbred and outbred birds typing F13 and L13. MLR phenotypes of outbred and inbred chickens typingB13of the major histocompatibility complex (MHC) of the chicken were compared. F1hybrids of outbred and inbredB13positive chickens were analysed in mixed lymphocyte culture (MLC). The intermediate strength responses of cells fromB13heterozygous outbred chickens stimulated by inbred B13homozygous chicken cells were not due to minor variations of B encoded lymphocyte activating determinants (Lads). Nor were Lads encoded by genes unlinked to the B complex responsible for these reactions. In contrast, F1anti‐parental type reactions were observed, and these alone are probably responsible for the intermediate strength reactions so often seen in typing of heterozygous outbreds with homozygous typing cell

摘要:
A new B‐cell alloantigen, designated as TB21, was serologically defined by 2 monospecific sera, T383 and T2200. Both T383 and T2200 had cytotoxic titers of 1:8 against TB21 positive B cells and were highly correlated to each other (r = 0.876). The phenotype frequency of TB21 was approximately 55% in the Japanese population. TB21 was strongly associated with DRw9 and weakly with DR5, but was definitely distinct from the known supertypic specificities such as MT or MB antigens. Family studies showed that TB21 segregated with different DR antigens on different haplotypes, indicating that TB21 is coded for by a gene at a closely linked but distinct locus from HLA‐DR. Furthermore, TB21 was preferentially detected on B cells but not on monocytes by complement‐dependent cytotoxicity. Failure of detection of TB21 on monocytes further distinguished the TB21 antigen from DR and MT antigens. This report presents the antigen frequencies for HLA‐A, B, C, DR, immunoglobulin, and red blood cell systems for the Navajo Indians of North America. HLA‐A‐B, B‐C, and B‐DR haplotype frequencies and significant delta values are given. These data are compared with similar data for other American Indians and major non‐Indian ethnic groups. Neoplastic cells from patients with a variety of B‐or non‐B/non‐T‐lymphoid malignancies, including chronic lymphocytic leukemia, various histologic types of non‐Hodgkin lymphoma and acute lymphocytic leukemia, showed strong variation with respect to stimulatory capacity in mixed lymphocyte culture (MLC). MLC stimulatory capacity was either normal or strongly decreased in comparison with that of normal lymphocytes despite the expression of HLA‐DR antigens and p23,30 (“la‐like”) determinants on the tumor cells of all patients. Strongly decreased stimulatory capacity of HLA‐DR‐positive tumor cells could not be ascribed to suppressive activity of the tumor cells. Tumor cells from three patients with a T‐cell malignancy failed to stimulate in MLC and did not react with anti‐p23,30 serum. Over 200 cases of non‐Hodgkin's lymphoma were typed for HLA‐A, B, C and DR antigens, and typing was correlated with the morphologic diagnosis according to the Lukes‐Collins classification system of non‐Hodgkin's lymphomas. Three major racial groups were represented: Caucasoids, Mexican‐Americans and Negroids. Significant associations were observed between HLA‐AW33 and Caucasoid B‐cell lymphomas, and for HLA AW24, HLA‐B37 and HLA‐B40 and B cell lymphomas in Negroids. The lymphocytes of 52 patients with the clinical diagnosis of idiopathic thrombocytopenic purpura (ITP) were typed for the HLA‐A,‐B and‐C antigens, and of 27 of those patients also for the DR antigens. ITP proved not to be significantly associated with any of the HLA‐A,‐B,‐C or‐DR antigens tested for. On the platelets of 35 of these 52 patients, autoantibodies were detected in the direct immunofluorescence test. In these 35 patients with autoimmune thrombocytopenia (AITP), an increased frequency of HLA‐Bw38 was found (14.3% versus 2.6% in controls). The frequency of none of the HLA antigens was significantly increased in the group of 17 ITP patients without demonstrable autoantibodies. Antigen‐specific T cell blasts may be separated from alloreactive cells on Percoll gradients. The absence of alloreactivity allowed us to study HLA‐D/DR restriction of proliferative T cell responses towards PPD and HSV antigens using allogeneic antigen‐presenting cells. An overriding impact of self HLA‐D/DR determinants as restriction elements forin vivosensitized T cells was demonstrated.
