Cells from the human immature monocytoid cell line U‐937 were induced with 12‐0‐Tetradecanoyl‐phorbol‐13‐acetate (TPA) to differentiate towards macrophage‐like cells. The expression of HLA‐antigens during differentiation was examined with a panel of monoclonal antibodies directed against monomorphic and polymorphic determinants. Class II antigens could be detected neither on uninduced nor on TPA‐induced U‐937 cells. While the expression of HLA‐A3 did not change significantly during differentiation, the “supertypic” specificities HLA‐Bw4 and Bw6 as well as the “private” specificity HLA‐B18 could be detected only on a drastically decreased number of cells after 4 days of exposure to TPA. Patients with Sjögren's syndrome (SS) in association with either Rheumatoid Arthritis (RA), Systemic Lupus Erythematosus (SLE) or Sclerodactyly were tested for HLA‐A, B and Dr antigens and the phenotypes of the 4th component of complement (C4). A panel of crossreactive anti‐human, ‐mouse and ‐rat MHC class II monoclonal antibodies (Mabs) was used to examine MHC class II antigen expression on canine T lymphocytes by cytofluorometry. The presence of MHC class II antigens was demonstrated on activated T lymphoblasts as well as on non‐stimulated peripheral blood T lymphocytes. A number of anti‐MHC class II Mabs reacted only with activated T lymphoblasts. Immunoprecipitation studies confirmed the Ia‐like or MHC class II molecular character of the antigens on canine T cells.The expression of MHC class II antigens on peripheral blood T lymphocytes appeared to be not induced by stimulation of the T cells, as purified T lymphocytes of specific pathogen free dogs reacted with anti‐MHC class II Mabs. Moreover, the study indicates that MHC class II antigen expression is present in the neonatal thymus. We previously reported that the expression of Ia antigens on cultured monolayers of murine thyroid epithelial cells (TEC) occurred with a particular distribution exclusively on the basal part of the cultured thyroid cells, while class I antigens of the major histocompatibility complex (MHC) and thyroglobulin (Tg) are only detected on the apical surface. It appears that, 24 to 48 h after the deposition of syngeneic lymphocytes, Ia antigens are expressed on the apical side of cultured TEC, i.e. the surface that bears Tg and is in direct contact with the responder lymphocytes during syngeneic sensitization of T lymphocytes. We hypothesized that this phenomenon could represent, in syngeneic situations, the restriction process in antigen recognition by T cells, as demonstrated by I‐A restricted primary syngeneic sensitization (PSS) on murine TEC.Moreover, we found that the addition of either PSS supernatants or γ interferon (γ IFN) to monolayers of TEC induced the expression of Ia antigens on the apical cell surface of these TEC cultures 24 to 48 h later.Taken together our data demonstrate that during the first hours of PSS, class II antigen and Tg are expressed on murine TEC cultures. In an attempt to further map the chicken MHC (the B complex), a systematic search for genetic recombinants within the B complex was performed by serotyping the progeny from F2 crosses of chickens by means of specific anti‐class I, anti‐class II, and anti‐class IV alloantisera.Two recombinant B‐haplotypes (B21r and B15r) were found by analysing 2,656 F2 chickens representing 5,312 informative typings. In either case, the B‐G (class IV) allele was recombined with both the B‐F and B‐L alleles of the opposite haplotype. MLC typings, tests for direct compatibility by GVH reactions, and absorption analyses confirmed the original serological typing of the two recombinant B haplotypes. 摘要:
We have used the serological and cellular (PLT) techniques to investigate the heterogeneity of HLA‐DR2. The serologic studies demonstrate that DR2 can be subdivided into two specificities based on the patterns of reactivity with the Ninth Workshop serum, 9w555. These two DR2 subtypes were also recognized by cellular assay utilizing PLTs generated between HLA‐DR2, DQ‐identical, DP‐identical or incompatible individuals. The data, therefore, suggest the existence of two alleles of DR2, DR2.1 (long) and DR2.2
