Abstract:Porcine peripheral T‐cells bear CD4 and CD8 cell surface antigens that distinguish helper from cytotoxic T‐cells. In distinction from what has been found in other species, a large percentage of peripheral T‐cells simultaneously express both CD4 and CD8. Monoclonal antibody 76‐7‐4 was found to stain all cortical and 7 ± 3% of medullary thymocytes, Ia+epidermal cells (i.e. Langerhans cells), no peripheral T‐cells and 50% of peripheral B‐cells. The antigen detected appears analogous to human CD1. All cortical thymocytes were also stained with CD4 and CD8 mAb. Abstract:In view of numerous recent reports of T‐cell receptor (TCR) β‐chain/disease associations with HLA‐associated diseases, we tested the possibilities that associations might exist directly between these two gene complexes at the level of the germline DNA. We determined frequencies of five TCR‐β DNA polymorphisms in 33 HLA‐DR2/2 homozygotes, 29 HLA‐DR3/3 homozygotes and 42 HLA‐DR4/4 homozygotes. The control population (n = 74) was chosen without “bias toward” their HLA‐DR genes. We selected DR2, DR3 and DR4 homozygotes because they have been the most frequently involved in HLA‐DR associated diseases. Our results indicate that the recent reports in the literature of TCR‐β/ disease associations can not be explained by a significantly different distribution of TCR‐β genes in HLA‐DR2+, ‐DR3+, or ‐DR4+subpopulations. Our results also suggest that if co‐evolution between TCR‐β and MHC haplotypes does exist, the selective pressures in recent generations have not been strong enough to significantly alter the germline TCR‐β gene frequencies

Abstract:Our group recently developed oligonucleotide probes associated to the TA10 and 2B3 specificities (1). Unambiguous typings were observed in a panel of homozygous typing cells and a family, using 32P‐end labeled probes and PCR‐amplified DNA (DQB exon 2). To investigate whether these TA10 and 2B3 associated oligonucleotides could be used in routine HLA‐typing we extended the study to a panel of healthy, unrelated individuals. When TA10 and 2B3 typings by oligonucleotides were compared with those obtained in routine serology a complete correlation was observed for TA10. A discrepancy was seen for 2B3 typing in material obtained from DQw5‐ (and possibly DQw4)‐positive individuals which could be explained by the CYNAP (cytotoxicity‐negative, absorption‐positive) phenomenon, using the IIB3 monoclonal antibody in routine tissue typing. Abstract:The secondary sex ratio (male births of total births) variation was investigated as a function of MHC antigens. Previous studies have indicated that at conception male zygotes are more frequent than female ones and abortions of male fetuses prevail. The present investigation addresses the question whether the histocompatibility relationships either between parents or between mother and offsprings may favor or hinder the outcome of pregnancy: particular combinations may cause distortions in the sex ratio. The results of the analysis of a sample of fertile families, demonstrate that, in first panties, female births are favored when parents share antigens at both HLA‐A and ‐Bloci(S.R. = 0.389), and mothers and fetuses share antigens at the HLA‐Blocus(S.R. = 0.222). The lack of antigens shared by parents at the threelociHLA‐A, ‐B and ‐DR increases first male births (S.R. = 0.696). In the case of fetal maternal histocompatibility at DRlocusonly a slight, non‐significant, increase in male first births (S.R. = 0.667) was observed. Abstract:Full HLA‐A,B and DR typing was carried out on 92 women with proteinuric pre‐eclampsia, 80 of their husbands and 46 of their babies. The results were compared with corresponding data from 65 normotensive pregnancies involving primiparous women. The frequency of HLA‐DR4 was increased in the pre‐eclamptic women (RR 3.1; p<0.005) and in the babies of pre‐eclamptic pregnancies (RR 2.6; p<0.03). The strongest association, however, was with sharing of HLA‐DR4 between mother and fetus (RR 4.2; p = 0.01). There was no increase in HLA antigen sharing in general between spouses or maternal‐fetal pairs in pre‐eclampsia. Nor did pre‐eclamptic women exhibit increased homozygosity in general at any HLA locus. Abstract:In order to evaluate genetic linkage between Allergic Asthma (AA) and the HLA system, we studied 20 families having AA affected sib pairs and 8 families with Intrinsic Asthma (IA) affected sib pairs. All AA patients had a strong IgE immune response to the miteD. farinae.Serological HLA typing (A, B, C, DR, and DQ antigens) was performed by the standard microcytotoxicity assay. Genetic analysis was made by means of the “Affected sib pairs” method. Out of the 20 affected sib pairs, 14 shared two HLA haplotypes, five shared one HLA haplotype and one was HLA‐non‐identical. These results differed significantly from the random ratio 1:2:1 for sharing, 2, 1 or 0 haplotypes (p<0.0001), and was very close to that expected for a recessive mode of inheritance. In contrast, among the IA sib pairs there was not an important distortion in the pattern of haplotypes segregation. However, a significant association between any of the HLA alleles and the two types of Asthma studied was not found.
