Fab'‐fragments derived from antibodies against β2‐microglobulin will completely abolish the cytotoxic effect of antibodies against HL‐A serological determinants. This is the result of the Fab'‐fragments competing with the antibodies against the HL‐A antigens for binding to the lymphocyte surface. Reciprocally, it is shown that Fab'‐fragments against the HL‐A alloantigenic structures impeded the binding of Fab'‐fragments to cell surface bound β2‐microglobulin.Fab'‐fragments against β2‐microglobulin inhibit considerably the mixed leucocyte culture reaction (MLR). The main effect is on the responding cells, whereas when the stimulating cells are treated with Fab'‐fragments against β2‐microglobulin they do not (or, at most, only slightly) change their characteristics in the MLR. The presence of HL—A antigens on various blood cells was determined by absorption techniques with the absorbed antisera being tested in the lymphocyte cytotoxicity test for loss of antibody activity.To overcome the risk of contamination by lymphocytes, methods for the preparation of pure cell suspensions of platelets, granulocytes and erythrocytes were evolved, giving high yields of cells.The following antigens were detected on platelets and granulocytes, but not on erythrocytes either fresh or papainized: HL—A1, HL—A2, HL—A3, HL—A9, HL—A10 and HL—A11 of the first series, and HL—A5, HL—A7, HL—A8, HL—A12, HL—A13, W5, W14, W15, W17, W18, W22, W27 and possibly W10 of the second series.The distribution of antigens on the various blood cells using cells from the same donor was also investigated, and it was found that the same HL—A antigens were present on platelets, gra

Antisera specific for human lymphoid cells in tissue culture were prepared. Rabbits were immunized with a lymphoid cell membrane fraction solubilized by papain and purified by physicochemical means. After appropriate absorption (with peripheral blood leukocytes of the same HL—A type) the antisera reacted with all established lymphoid cell cultures tested, but not with peripheral blood leukocytes.The antigen(s) reacting with these antisera seem to be unique for cells in culture and appear at the time the cell becomes an established cell line. Using the principles of Lightbody et al. (1971) the influence of SD differences in Cell Mediated Lympholysis (CML) between unrelated individuals was studied. These experiments showed that effector‐lymphocytes sensitized against a number of SD determinants showed declining killing efficiency confronted with targets having four, three, two or one of these determinants. Normally maximum killing efficiency was observed using stimulator and target lymphocytes from the same individual, while stimulator and target lymphocytes being unrelated but SD identical showed high, but not maximum killing. In SD identical effector/target combinations killing was not seen at a degree comparable to SD unidentical combinations. Typing for MLC determinants was done in 82 beagles of five different colonies using reference cells which could be shown by family analysis to be homozygous for both the serologically defined (SD) antigens and for the lymphocyte defined (LD) MLC determinants. One‐way nonstimulation with the homozygous reference cells was taken as evidence for the presence of the respective MLC determinant on the tested cells.Four LD specificities (DL—50, 51, 52, 53) are defined by homozygous cells and at least three additional LD types can be recognised by their nonidentity with the known types. AS and Lewis strain rats (both Ag‐B1) were immunised with each others’ thymus cells in an attempt to produce antisera analogous to the anti‐theta sera of the mouse. An AS anti Lewis thymus serum gave strong and equal reactions against thymus lymphocytes of Lewis, DA and AS2 strains, but was only weakly reactive with peripheral blood lymphocytes of those strains. The Lewis anti‐AS thymus serum was non‐reactive against both thymus and blood lymphocytes of all strains. The AS anti Lewis thymus serum appeared to be monospecific as determined by absorption studies and Fl hybrid to parental backross studies. The alloantigen system defined by this serum had the following characteristics: (1) the antigen was present on both thymus and lymph node lymphocytes, but not on brain, red cells, kidney, liver or heart, (2) the antigen segregates independently of Ag‐B, (3) the antigen was present in equal quantities on thymus and lymph node lymphocytes, (4) the antigen could not be modulated, (5) the antibody could not be produced in response to skin grafting. Immunogenetic studies were carried out on an HL‐A2/HL‐A3 heterozygous lymphoid cell line, 8866, and on two HL‐A2 negative variant sublines of 8866 which had arisen independently of one another. The parent 8866 line reacted positively with some W28 sera: these cross reactions were absent in both HL‐A2 “loss” variant sublines. Other serum reactions, apparently characteristic of lymphoid cell lines, were present in 8866 and were retained in the variants.
