Abstract:TheDQA1*0104allele is known to differ fromDQA1*0101by a single nucleotide in the sequenced part of the first exon.DQA1*0104has a guanine in the second position of the second expressed codon, whereasDQA1*0101and all other sequencedDQA1alleles have an adenine in that position, changing aspartic acid to glycine. TheDQA1*0104allele was originally described in African Americans with theDRB1*12, DRB3*0101, DQA1*0104, DQB1*0501, DRB1*12, DRB3*0202, DQA1*0104, DQB1*0605 or DRB1*14, DQA1*0104, DQB1*0503haplotypes. When developingDQA1typing by PCR amplification with sequence‐specific primers (PCR‐SSP), we observed that all DR10‐ and DR14‐positive samples carried theDQA1*0104allele, wheres allDRB1*01‐positive DNAs carried the closely relatedDQA1*0101allele. In the present study, samples representing the major ethnic groups with DR‐DQ haplotypes known to carry theDQA1*0104allele or the very similarDQA1*0101allele were investigated byTaqI RFLP analysis, PCR‐SSP typing and nucleotide sequencing. TheDQA1*0104allele was found to differ fromDQA1*0101not only in the second expressed codon, but also by a productive mutation in the signal peptide. All investigatedDRB1*1001‐(n = 24) andDRB1*1401‐positive (n = 25) haplotypes, defined by homozygosity or association, of Caucasian, African or Oriental origin carried theDQA1*0104allele, whereas theDQA1*0101allele was found on allDRB1*01‐positive (n = 32) haplotypes. These findings demonstrate that in the assignment of HLA class II alleles, polymorphism outside the second exon sometimes must be considered. The maintenance of theDQA1*0104allele on a few distinct haplotypes indicates that the allele is old and might also be compatible with a functional difference between theDQA1*0101

Abstract:We describe here the generation and characterization of a human monoclonal IgG antibody (UL/F14) specific for HLA‐B12. The antibody is suitable for complement‐dependent lysis on lymphoblastoid cell lines and stimulated peripheral blood mononuclear cells. 摘要:
Abstract:We have developed monoclonal antibodies to most HLA specificities, making it possible for us to devise a simple, rapid, one‐step micro‐cytotoxicity test. The test is performed by adding 1 μl of cells to 1 μl of antibody‐complement mixture predotted on the microtest tray. The reactions are read following a 1‐hour incubation period (30 minutes in some instances). The analysis of reactions seen on testing 105 class I antibodies and 50 class II antibodies is shown. A comparison of typing by the standard NIH method and the new one‐step procedure showed a>96% concordance in the 500 T cells and 200 B cells we examined. Class I and class II typing could be performed using B cells, thus obviating the need to isolate both T and B cells for
