Glutathione peroxidase (GSH‐PO) and glutathione S‐trans‐ferase (GST), class α, showing GSH PO like enzymatic activity, were localized immunohistochemically in frozen sections of rat intestine in order to elucidate changes in the expression of these antilipoperoxidative enzymes during normal development. The direct immunoperoxidase method was performed using specific rabbit antibodies (Fab fragments) against the enzymes purified from rat liver. Immunoreactive GSH PO and GST ‐α were demonstrated in the intestinal villous epithelial cells. In the duodenum, GSH‐PO was positive during the period from 19 days of gestation to 1 week after birth, while GST α was negative during this period. Two weeks after birth, positivity for GST α appeared, and GSH‐PO became undetectable. In the ileum, both of the enzymes were observed until 2 weeks of age, but after weaning their expression disappeared. These immunohistochemical findings were confirmed by immunoblot analysis using intestinal tissue extracts. To evaluate environmental effects on the expression of these enzymes, germ free animals, common bile duct ligated rats, and Hank's solution‐fed infant rats were prepared. No remarkable alterations in the immunohistochemical localization pattern were observed. Since the switching of enzyme expression around the time of weaning was not influenced by these experimentally induced environmental conditions, it appears that these enzymatic changes are genetically predetermined. After60Co irradiation of rat spleen, the most severe damage was found initially in the lymph follicles (LFs); lymphocytes were decreased in number in the periarteriolar lymphoid sheath (PALS), and almost completely depleted in the marginal zone (MZ). Recovery was observed first in the PALS, followed by the LFs, and finally the MZ. From the results of immunohistochemistry, the process of recovery was assumed to involve migration of T and B lymphocytes along the arterioles from the area surrounding the terminal arteriole at the end of the PALS into the central part of the PALS and into the LF. Peripheral blood lgM+, IgD−B lymphocytes, known to be MZ precursor cells, flowed from the follicular artery to settle in the MZ. The authors' previous report concluded that increased mesangial IgA deposition seen in ddY mice pretreated with mesangiotropic anti‐type IV collagen serum might be due to a dysfunction of mesangial transport of macromolecules such as polymeric autologous IgA. In the present study we examined the effect of macromolecules upon mesangial transport in similarly treated ddY mice, using intravenously administered FITC labeled dextrans of various molecular weights as tracers. The intensity of each resulting mesangial dextran deposit inspected directly by immunofluorescence was measured periodically and compared with the deposition of autologous mouse IgA examined using rhodamine labeled rabbit anti‐mouse IgA. High‐molecular‐weight dextran of 2,000 kDa showed prolonged mesangial deposition which paralleled the intensity and distribution of the autologous mouse IgA deposition. In contrast, medium‐ and low‐molecular‐weight dextrans of 500 and 150 kDa, respectively, disappeared earlier from the mesan‐gium, unlike the autologous IgA deposition which persisted. Based on these results, it was concluded that the mac‐romolecularity of certain substances and a transport dysfunction of the mesangium may both be major factors in prolonged mesangial deposition. These findings may provide some clues to help clarify the pathogenesis of human IgA nephritis. Acta Pathol Jpn

The frequency of neurofibrillary tangles (NFTs) in relation to aging was examined in five regions of the hippocampus of 139 normal controls (60 to 106 years) and 14 patients with senile dementia of the Alzheimer type (SDAT). The regions were CA4, CA3, CA2 and CA1, the latter being subdivided into CA1a (the region near CA2) and CAlb (the region near the subiculum). Only CA2 showed no correlation of NFTs with normal aging. CA2 in SDAT was the site most vulnerable to NFT formation. Therefore, it is considered that CA2 is a special region in both normal aging and SDAT. CA1 was characterized by the early appearance of large amounts of NFT, and was the most reliable marker of senile change related to aging. Like CA1, both CA3 and CA4 showed an increase of NFTs with normal aging, but their aging process was far less marked. In addition, statistical analysis based on a “normal” distribution was considered to be unsuitable for quantitative study of senile change, because no histogram of NFTs in any subdivisions for any age showed such a distribution. In order to obtain objective histologic criteria for distinguishing intraductal carcinomas of the breast from benign intraductal hyperplasia, a morphometric study was carried out on 15 cases of breast carcinoma (5 cases of comedo carcinoma, S cribriform carcinoma and 5 papillotubular carcinoma) and 10 benign control lesions (5 papilloma and 5 fibrocystic disease). Morphometric analysis was performed for each group with regard to 1) mean nuclear area, 2) mean nucleolar size, 3) mitotic index, and 4) nuclear count within predefined area (nuclear density) in histologic specimens. The results obtained were as follows: 1) Mean nuclear area for comedo carcinoma was distinctively large. This, together with its coefficiency variance, may be a good indicator for identifying this type of carcinoma. 2) Mean nucleolar size was significantly larger in each carcinoma group than in their benign counterparts. Nucleolar size was therefore considered to be the most reliable parameter for discriminating malignant cells from benign hyperplasia. 3) Mitotic index was relatively high in comedo and papillotubular carcinomas. 4) No difference was seen in nuclear density between the malignant and benign groups. Bone tumors were categorized into alkaline phosphatase (ALPase)‐positive (2 ossifying fibromas, 1 benign osteoblastoma and 16 osteosarcomas) and negative (2 chondromas, 2 chondrosarcomas, 3 non ossifying fibromas, 2 malignant fibrous histiocytomas and 6 giant cell tumors of bone) groups. Production and distribution of matrix vesicles (MVs) in the tumor tissues were examined to clarify their role in neoplastic bone formation. Four distinct types of MV were isolated primarily in ALPase‐positive bone tumors: empty, amorphous, crystalline and ruptured MVs. They were formed by budding off from the cytoplasmic projections of the osteoblastic tumor cells. The significance of differences in the production rate of MVs between ALPase positive and negative bone tumors was investigated in view of the predominantly high production of MVs in ALPase‐positive bone tumors. Many more mature MVs (crystalline and ruptured) were observed in the osteoblastic lesions of osteosarcoma than in the fibroblastic and MFH‐like lesions, suggesting an intimate relationship with maturation and differentiation of the osteoblastic tumor cells. The above findings indicate that production of MVs is one of the diagnostic parameters for osteoblast‐ derived bone tumors, as well as ALPase activity, and that vesicle‐induced mineralization is a major mineralization mechanism in neoplastic bone formation. A 38‐year old man presented with a cavernous hemangioma in the liver. Transarterial embolization (TAE) using a gelatin sponge was carried out 14 days prior to surgical resection of the tumor. Granulomatous arteritis with massive infiltration by eosinophilic leukocytes and histiocytes was present at the periphery of the hemangioma, and transient eosinophilia in the peripheral blood occurred six days after resection. Granulomatous arteritis was evident in medium‐sized arteries and there was narrowing or occlusion of the vascular lumen. In the granulomatous cellular infiltrates in the arteries, giant cells of the foreign body type were numerous. An eosinophilic substance differing from fibrin was present in some of the vascular lumina. As this showed staining for collagen, it was considered likely to be fragments of the gelatin sponge. The patient had no symptoms of fever, chills or general fatigue. The clinical course and pathologic findings suggest a causative role of the gelatin sponge in this case of granulomatous arteritis. Vascular change, a rare complication of TAE therapy, may be induced by a hypersensitivity reaction against the intraarterial gelatin sponge. Acta Pathol Jpn 41: 618‐622,

We present two similar cases of rectal B‐cell lymphoma with diagnostic problems. Grossly, both tumors appeared as a well demarcated polypoid mass with eroded mucosa. In spite of their histologic resemblance to reactive lymphoid hyperplasia, Including the presence of lymph follicles, fibrosis and polyclonal plasma cells, both cases were diagnosed as malignant lymphoma of low‐grade malignancy because of the full‐thickness involvement of the rectum with diffusely infiltrated small or medium‐sized lymphoid cells showing rare mitoses. In one case, clonal proliferation of differentiated B cells was demonstrated by analysis of gene rearrangement. Therapeutic problems related to low‐grade malignant lymphoma in the rectum are also discussed. Acta Pathol Jpn 41: 623

An unusual pancreatic tumor in a middle‐aged man is reported. The tumor was a large encapsulated mass with necrosis and hemorrhage, and microscopically, it showed a predominant microcystic pattern with solid pattern areas. The clinical and histological features of this tumor were similar to those of solid and cystic tumor. Immunohisto‐chemical studies showed that the tumor was positive for ax‐AT, NSE, CA 19 9 and somatostatin. Electron microscopy studies showed neurosecretory granules, intercellular junctions, annulate lamellae and aberrant crystalloid structures. These findings indicated that the tumor had differentiated into acinar, ductal and endocrine cells, suggesting that solid and cystic tumor originates from multipotential immature cells. A rare duct‐islet cell tumor of the pancreas was studied using immunohistochemical, cytofluorometric and histo‐chemical methods. Histology and immunohistochemistry revealed that the tumor contained two distinct cell types; islet cell‐like neuroendocrine cells and exocrine duct cell components, suggesting an endodermal origin for both types. The cells showed marked pleomorphism and vascular and perineural invasion at the tumor periphery. Cytofluorometric study of the tumor cell DNA revealed an increased mean nuclear DNA content, without any aneuploidy. Histochemically, the tumor cells contained an increased number of argyrophilic nucleolar organizer regions (AgNORs) in their nuclei.
