The effects on plasma lipids of daily intraperitoneal injections of 4mg of melatonin (N‐acetyl‐5‐methoxytrypt‐amine) for 10 27 day periods were examined biochemically and morphologically in rats fed regular and high‐cholesterol (1% cholesterol, 0.5% cholic acid) diets. Melatonin administration had no significant effect on plasma lipids and lipoproteins in the rats on a normal diet but blunted the effects of a high‐cholesterol diet on these parameters. No effects of melatonin on lipase activity were noted. Melatonin also diminished the fatty infiltration in the liver of animals on the high‐cholesterol diet. The high‐cholesterol diet produced major increases in VLDL and LDL cholesterol and protein content, and decreases in HDL cholesterol and protein. Melatonin decreased the extent of this plasma lipoprotein increase, although it did not completely prevent the phenomenon. In order to investigate the role of polymorphonuclear leukocytes (PMN) in the distribution of antigen in various organs and in the development of nephritis, chronic serum sickness type nephritis was induced in both anti rat PMN rabbit serum (APS) treated and normal rabbit serum (NRS) treated rats by preimmunization with bovine serum albumin (BSA) and subsequent daily intravenous administration of BSA for 4 weeks. Kinetic studies using radiolabeled BSA showed that accumulation of BSA after the first intravenous administration was reduced by APS treatment in the liver, lungs and spleen and increased in the circulation, but was not affected in the kidneys and glomeruli. Histological studies supported the above findings. After 4 weeks of BSA administration, the BSA accumulation in the kidneys and glomeruli was significantly less in APS treated rats than in NRS treated ones, while amounts of BSA in the circulation and other organs were not different between the two groups. Furthermore, APS treatment reduced proteinuria, PMN infiltration and IC deposition in the glomeruli. These observations indicate that PMN play a partial role in IC deposition in the glomerular capillary walls and subsequent destruction of glomerular permeability in chronic serum sickness‐type nephritis. The human gastric cancer cell lines, MKN1 and SCH, were biochemically and biologically characterized according to the monophenotypic expression of placental alkaline phosphatase (PLAP)‐ like enzymes. The MKN1 cell line, derived from adenosquamous carcinoma, showed the same enzyme properties as the Regan isoenzyme, whereas the SCH cell line, derived from primary gastric choriocarcinoma, had properties identical with the Nagao isoenzyme. Regan isoenzyme activity expressed by MKN1 cells was stimulated by glucocorticoid and suppressed by retinoic acid. Both agents had no significant effect on SCH cells. On the other hand, Nagao isoenzyme activity expressed by SCH cells was stimulated by sodium butyrate, which had no stimulatory effect on MKN1 cells. Moreover, the PLAP like activity of MKN1 cells showed no observable relationship to human chorionic gonadotropin (hCG) producibility. Whereas expression of the Nagao isoenzyme by the SCH cell line is presumably a result of functional differentiation in the trophoblastic direction, that of the Regan isoenzyme by the MKN1 cell line is probably not. In order to elucidate the relationship between the level of cellular Ki‐67‐reactive antigen and cell proliferation activity, the effects of serum deprivation on the antigen expression and cell proliferation of HeLa cells were investigated using flow cytometry. The antigen was constitutively expressed in almost all cells cultured in normal medium containing 10% calf serum. In the serum deprived cells, the antigen negative fraction increased with culture time, and the level of Ki 67 reactive antigen fell rapidly during the first 24 h. The reduction in the antigen caused by serum deprivation was independent of the cell cycle phase. Both BrdUrd incorporation and the S phase fraction were diminished during the experiment with evidence of cell growth retardation. However, after refeeding with fresh normal medium, the antigen level that had been reduced by serum deprivation recovered to the level of the control cells within 24 h. Immunohistochemical and immunoblot analyses were performed on 49 cases of surgically resected primary lung carcinoma to examine the expression ofrasoncogene product using monoclonal antibodies toras‐p21. Two different monoclonal antibodies, NCC RAS 001 and RAP 5 were used for immunohistochemical study. Cancer cells of 16 cases (33%) and 15 cases (31%) were strongly positive for NCC RAS 001 and RAP 5, respectively. The staining pattern of antibodies was heterogenous among cancer cells, even in the same case. Among various histologic type of lung cancers, squamous cell carcinomas and well‐differentiated adenocarcinomas had a tendency to react more intensively than other histologic types of carcinoma. Immunoblot analysis using monoclonal antibody NCC RAS 004 revealed the presence of ras p21 not only in cancer tissues bus also in non‐cancerous tissues in all cases analysed. In 13 cases (27%), cancer tissue expressed more than twise as much ras p21 as non cancerous tissues. Our study showed that the over expression ofras‐p21 in lung carcinomas compared with non cancerous tissues was a relatively common phenomenon, especially in squamous cell carcinomas and adenocarcinomas. Although intrahepatic bile duct injury following bone marrow transplantation is considered to be one feature of graft versus host disease, its developmental mechanism has not been clarified. In order to elucidate this aspect, an immunohistochemical study of the liver following human allogeneic bone marrow transplantation was made. Cytotoxic T lymphocytes (Tc) and natural killer cells (NK) were found in contact with intrahepatic bile duct epithelial cells showing degeneration and necrotic changes. These findings suggested a cytotoxic effect of these cells on bile duct epithelial cells. Abnormal expression of HLA class II (DR) antigen was recognized in intrahepatic bile duct epithelial cells following bone marrow transplantation. Cell injury was prominent in cells with weak DR antigen expression, whereas the cells demonstrating conspicuous expression appeared almost normal. There results suggest that abnormal expression of DR antigen plays an important role in the development of GVHD of the intrahepatic bile duct. Acta Pathol Jpn 39: 648 655, 1989.

Immunophenotypic features of human sarcomatous Wilms’tumor (SWT) were studied using a newly established mouse monoclonal antibody (5H10, IgG). 5H10 was produced against CR SW2, one of several transplanted SWT lines in nude mice, and defines a 200‐kDa cell surface protein. The antibody was found to react equally with all subtypes of SWT; clear cell sarcoma of the kidney, malignant rhabdoid tumor of the kidney, and unclassified sarcoma. Furthermore, it reacted equivalents with surgically resected tumors, transplanted tumors in nude mice, and cell linesin vitro.On the other hand, 5H10 was entirely negative for any of the components of nephroblastic Wilms’tumor (NBW). Considering these results, 5H10 appears to recognize the antigen expressed preferentially on SWT, and therefore the subtypes of SWT may be closely related to one another immunophenotypically. In normal human tissues, however, 5H10 only reacted with fetal kidneys, its reactivity being restricted to the lower limbs of S bodies in both the metanephros and mesonephros. No reactivity was identified in any other tissues including adult kidney. These results indicate that 5H10 detects an oncofetal antigen expressed preferentially in both SWTs and fetal kidney and that the histogenesis of SWT should be considered in connection with nephrogenesis. Autopsy was performed on a 52 year old man with a 20‐year history of neurological symptoms. At autopsy, both a brain stem tumor and a spinal cord tumor were found. These showed the features of pilocytic astrocytoma histologically. A pilocytic astrocytoma was also found in the optic chiasm upon microscopical examination. These three tumors were thought to be multicentric astrocytomas, because there was no continuity among them and no evidence of dissemination or metastasis by any pathway. A case of blastic transformation of essential thrombocythemia (ET) is reported. A 69 year old male was first admitted to hospital because of fever in February, 1982. He was diagnosed as having ET because of marked thrombocytosis (205.5 ± 1010/1), absence of erythrocytosis, absence of splenomegaly, normal karyotype and no increment of blasts in the bone marrow, and normal levels of neutrophil alkaline phosphatase, vitamin B12and folate. He was treated with busulfan, and subsequently his platelet count was well controlled for about five years. At the second admission, blasts were present in the peripheral blood, and later accounted for 49% of the total leukocyte count. Histological examination of a bone biopsy specimen showed homogeneous proliferation of blastic cells and slight reticulin fibrosis. At autopsy, the degree of bone marrow fibrosis had increased. An extrarenal malignant rhabdoid tumor arising from soft parts in the prepubic region of a 37 year‐old man is described. This case appears to be unusual with regard to its location and age at onset. To our knowledge, the patient is the oldest recorded in whom such a lesion has arisen. Histologically, the tumor consisted of an admixture of polygonal and elongated cells with abundant eosinophilic cytoplasm frequently containing hyaline‐like globules. Ultrastructurally, these cytoplasmic inclusions were compatible with intermediate filaments. Immunohisto‐chemical staining disclosed keratin (non squamous epithelial type) and epithelial membrane antigen positivity. These characteristic features were identical to malignant rhabdoid tumor of the kidney seen in infants and young children.
