Serum levels of the protein factor of fertility (PFF) were measured by immunoenzymatic assay with a sensitivity of 4 ng/ml. The normal concentration of PFF was within 8–20 ng/ml in males and 20–40 ng/ml in females. 摘要:
An apparent activation of the malate dehydrogenase activity is observed in the double-reciprocal plot at high oxaloacetate concentrations when human hepatoma extracts are analyzed. This phenomenon does not occur in healthy liver samples. In hepatoma extracts, the ratio of lactate dehydrogenase to malate dehydrogenase activities becomes fivefold higher than that of normal liver. Experiments performed with mixtures of both purified enzymes and, conversely, by using oxamate, a specific inhibitor of lactate dehydrogenase, reveal that the deviation in Michaelis-Menten behavior observed is due to the oxaloacetate reductase activity of lactate dehydrogenase instead of the presence of a novel malate dehydrogenase isoenzyme.

This study investigated the influence of two formula diets containing 20 g/100 g diet of either whey protein concentrate or casein, or Purina mouse chow on 1,2-dimethylhy-drazine (DMH)-induced colon carcinoma in A/J mice. Four weeks after the 24th DMH treatment the incidence of tumour and tumour area in the whey protein-fed mice was substantially less in comparison to either the casein or Purina groups. The Purina group exhibited the greatest tumour burden. At the end of the experiment all animals continuously fed the whey protein diet were found to be alive, whereas 33% of those on the casein or Purina diet had died. Animals fed Purina diet for 20 weeks and then switched to either milk protein diet for a further 8 weeks exhibited a decrease in tumour burden as compared to those animals fed the Purina diet continuously. Body weights were similar in all dietary groups. In conclusion, a whey protein diet appears to significantly influence the development of chemically induced colon tumours and the short-term survival of mice.

Oncoprecipitins isolated from a number of marine invertebrates have been shown to be glycoproteins possessing a higher specificity to carcinoembryonic antigen (CEA) – similar to the interaction of antigen-antibody. The oncoprecipitin-CEA interaction was found to be substantially more specific than that of carbohydrate-containing polymers with lectins. Two oncoprecipitins, crustacin and cyprein, isolated from Pagurus prideauxii and Cyprea caputserpentis, respectively, have been characterized. The binding reaction of CEA with crustacin and cyprein failed to be inhibited with glycoproteins having sugar chain patterns closely related to those of CEA. Inhibition of the oncoprecipitin-CEA interaction with sugars was shown to be nonspecific and was observed only at sufficiently high concentrations of sugars used as inhibitors. Immunoenzymic assay of CEA content in normal and tumor tissues using CEA crustacin and CEA antiserum against CEA test systems revealed highly consistent results for both system

The mouse monoclonal antibody MA5, generated versus a membrane-enriched extract of breast cancer metastatic to liver, detects one or two high molecular weight species ( > 200kD) in breast tumor membranes, human milk fat globule membranes, and various breast tumor cell lines. From comparative studies of five breast carcinoma lines (BT20, BT549, MCF-7, T47D, and ZR75–1), as well as an epithelial line established from milk (HBL-100), we report the stimulation of expression of MA5-reactive antigen in a mucinous breast tumor cell line (BT549) through the use of a culture medium supplemented with charcoal-absorbed fetal calf serum, insulin, and hydrocortisone. Large amounts of aggregated MA5-reactive antigen are secreted into the culture medium and can be recovered from the media for further purification by centrifugation. These findings suggest that BT549 cells, grown in the special nutritive medium, may be useful in providing an ample source of epithelial membrane antigen (also termed polymorphic epithelial mucin) for standardization of clinical assay protocols, as well as provide a model system for studies of the regulation of expression for this class of antigens in breast carcinom

Four monoclonal antibodies reactive with the rod portion of human cytokeratin No. 8 were evaluated using experimental tumour radio-immunolocalization. Nude mice carrying human HeLa cell tumours were injected with 125I-labelled F(ab′)2 fragments or intact IgG antibodies. Positive immunolocalization was observed with all the specific antibodies, the intact antibodies being superior to their analogous F(ab′)2 600 h for some of the antibodies compared to a half-life of 40–80 h in non-tumour tissues. Optimal imaging was obtained as late as 10–14 days after administration of the antibody. A sensitive enzyme immunoassay (IMx) for the squamous-cell carcinoma (SCC) antigen was used to evaluate low concentrations of this marker in the blood circulation. The assay can detect 0.05 ng/ml of serum SCC antigen, with intra- and interassay variabilities of 4.8 and 8.9%, respectively. Daily changes in serum SCC antigen were determined in healthy volunteers and in patients with cervical squamous-cell carcinoma after complete resection of the primary tumor, which revealed that, although the absolute values were variable from one case to another, the daily changes were relatively constant in each case, with a mean daily variation of 24.1 ± 3.7% (mean ± SE). The determination of the cutoff value for each individual patient may be effective for the early detection of an abnormal rise of circulating tumor marker, and will be useful in detecting small tumor foci during follow-up after treatmen
