Human neuroblastoma (NB) cell lines have been suggested to represent a model of neural crest differentiation. The expression of several Schwann-cell-associated antigens was examined by flow cytometry and Northern blot analysis. Variable reactivity of the human NB cell lines was found in both the level and pattern of reactivity. Retinoic acid treatment of cell line SMS-KAN resulted in a neuron-like morphological differentiation and a decrease in several of the glial markers under study. Similarly, Northern blot analysis illustrated myelin-associated glycoprotein expression, and decreased expression of this message with retinoic acid treatment was consistent with the neuron-like morphological changes. Overall, human NB in vitro was found to be multipotential, but we have shown that it is capable of expressing several Schwann cell markers which are modulated during induced differentiation.

The serum levels of tissue polypeptide antigen were determined using the M3 monoclonal antibody (TPS) and compared with the serum levels of carcinoembryonic antigen (CEA) and breast carcinoma antigen 15–3 (CA 15–3) in 96 patients with benign breast tumors, in 25 breast cancer patients with no evidence of disease, in 139 preoperative breast cancer patients and in 298 samples of 25 breast cancer patients during therapy monitoring (4–22 samples per patient). The 95th percentile of TPS in 89 apparently healthy females was 51 U/l. The 95th percentile of TPS in patients with benign breast tumors was 55 U/l. The maximum TPS level in breast cancer patients with no evidence of disease was 56 U/l. In preoperative breast cancer the number of patients with TPS levels above the 95th percentile of TPS in benign breast tumors was significantly higher in stage III breast cancer as compared with stage I + II. This was not established for CEA and CA 15–3. During therapy monitoring TPS followed the course of the disease faster than CEA and CA 15–3 in patients with bone metastases, liver metastases, lung metas-tases and pleural effusion, with one exception. In the search for a method to facilitate the preoperative discrimination of ovarian carcinomas from colorectal carcinomas serum levels of 6 tumor markers were measured in 47 patients presenting with ovarian cancer and compared to levels found in 24 female patients with advanced, untreated colorectal cancer. The markers studied were CA 125, CA 15.3, CA 19.9, CEA and two recently developed mucin markers, CA M29 and CA M26. Levels of CA 125, CA 15.3, CEA and CA M29 showed significant differences between both groups. In predicting ovarian cancer, sensitivity was highest for CA 125 at 94% (35 U/ml cutoff level). However, the specificity of CA 125 was at 71% low. Specificity increased significantly by using a combination of a CA 125-positive score ( > 35 U/ml) and a simultaneous negative CEA score (≤ 5 ng/ml) (specificity 100%, sensitivity 81 %). A CA 125/CEA serum ratio > 25 resulted in the highest discriminative power with a specificity of 100% and a sensitivity of 91 % resulting in an overall test accuracy of 94%. The expression of a proliferating antigen by Ki-67 immunohistochemistry was evaluated in 32 gastrointestinal carcinoids and in 5 pancreatic islet cell tumors. In the tissue sections the number of labelled nuclei was calculated per tumor area. The tumors were classified as low proliferating ( < 0.3 labelled cells/mm2), medium proliferating (0.3–1 labelled cells/mm2 1 labelled cell/mm2). In 26 tumors obtained from patients receiving antitumor therapy (α-inter-feron) the proliferative activity was decreased. In treated mid-gut carcinoids the proliferative activity in metastatic tissue was significantly reduced (p < 0.05). Though not statistically significant, primary midgut carcinoids collected from untreated patients displayed a lower proliferative activity than liver metastases. A survival analysis revealed that patients with tumors displaying low proliferative activity had a better survival than those with high proliferative activity (p < 0.05). Single cell cytofluorometric DNA analyses showed regular diploid stem cell lines in the majority of tumors from untreated patients (9/11 cases). No correlation was found between the calculated proliferative activity and the DNA profile. A neural origin of Ewing’s sarcoma (ES) has often been suggested and we have demonstrated neurofilament protein expression in ES cells. However, only the 200-kD subunit has been revealed in all of the ES cells analyzed. The 160- and 68-kD subunits were always absent. For these reasons, we have attempted to induce neural differentiation in 3 ES cell lines with different types of inducers: tetradecanoylphorbol-13-acetate (TPA) retinoic acid and nerve growth factor. When the cell lines were cultured for 7 days with TPA (10-9M) or retinoic acid (10-7M), only the 68-kD neurofilament subunit was slightly induced. No inducation was obtained when nerve growth factor was used, even at a 21-day culture. These results are in agreement with the putative neural origin of ES and may indicate an abnormal expression of neurofilament proteins in this tumo

A novel chicken gonadotropin-releasing hormone (GnRH) analogue (Folligen), which has a different mechanism of action than superactive mammalian GnRH analogues, has been developed. In this study we report that in 9,10-dimetyl-1,2-bezanthracene-indced mammary carcinomas in rats, Folligen caused an almost 100% tumour remission during a 3-week daily treatment, without blocking ovarian functions. It decreased oestradiol but not to castration levels, while progesterone was not decreased at all; instead, it was slightly stimulated. The histological picture of the ovaries showed no signs of the inhibition of ovarian functions, but the presence of developing follicles and corpora lutea. In comparison, super-active mammalian GnRH analogues caused similar tumour remission but with hormonal castration and a complete block of ovarian functions.
