Ten recognized members of the tombusvirus group, including two strains of tomato bushy stunt virus, were compared for their ultrastructural effects in Nicotiana clevelandii, Gomphrena globosa, and Chenopodium quinoa. The cytopathic inclusions known as multivesicular bodies and tonoplast evaginations containing virus particles were induced by all the viruses in all three hosts. Some of the viruses could be differentiated from each other (especially in C. quinoa) by other characters, such as the accumulation of membranes in cell nuclei, or the type of organelle (chloroplasts, mitochondria or peroxisomes) from which multivesicular bodies developed. The usefulness of these characters for virus identification and grouping is discussed.

Three double-stranded (ds) RNA-containing viruses, named hop trefoil cryptic virus (HTCV) 1, 2 and 3, were found in apparently healthy seedlings of hop trefoil (Medicago lupulina). Only 1–2% of plants tested were virus-free. The particles occurred in the cytoplasm, nuclei and nucleoli of parenchyma cells. They were purified as a mixture (HTCV-M) with a mean yield of 50–100 µg/kg fresh tissue, and an antiserum was obtained. HTCVl was serologically related to one component (ACV1) of a mixture of alfalfa cryptic viruses (ACV-M) present in alfalfa (M. sativa), and was separated by immunoprecipitation from HTCV-M; its particles, about 30 nm in diameter, contained two dsRNAs (Mr 1.27 × 106 and 1.15 × 106) comigrating in PAGE with two RNAs of ACV-M. Particles of HTCV2, about 38 nm in diameter and with distinct morphological subunits, were serologically closely related to particles of similar morphology (RCCV2) present in a mixture of cryptic viruses (RCCV-M) from red clover (Trifoliumpratense), and were distantly related to white clover cryptic virus 2, also of similar morphology. HTCV3 had particles 30 nm in diameter, morphologically similar to those of HTCVl, and reacted with an antiserum to vicia cryptic virus. HTCV2 and 3 could not be individually separated. The RNAs of HTCV-M, in addition to the two bands attributed to HTCVl, gave, upon electrophoresis, three bands of estimated Mr 1.34, 1.08 and 0.95, all × 106, and up to five minor bands of Mr ranging from 1.45 to 0.98, all × 10

摘要:
The packaging proteins, gp2 and gp3, of Salmonella phage P22 were separated by DEAE-sephacel column chromatography. The activity of both proteins was measured by complementation in an in vitro phage packaging assay. For the first time gp3 activity was demonstrated in vitro. The majority of the gp3 activity could be separated from gp2. While gp2 was stable during chromatography and storage, gp3 was very unstable. However, it was stable when stored as a crude extract. The ratio and interaction of these two proteins critical for phage maturation are still uncertain.

摘要:
This paper describes the derivation and characterization of monoclonal antibodies reactive with the group-specific epitopes of the VP1 peptide shared by enteroviruses. When tested against a wide range of the prototype strains of Coxsackie viruses A and B, polioviruses, and echoviruses, the monoclonal antibodies reacted with all of them in two or more assay systems. The virus of hepatitis A was the only enterovirus tested which was not recognized by these monoclonal antibodies. This finding is consistent with recent reports demonstrating genomic dissimilarity between this virus and the other enteroviruses. Although monoclonal antibodies have frequently been found to be assay specific, these particular antibodies were found to be equally effective in different immunochemical and biological assay systems. Biochemical characterization of one of these monoclonal antibodies is described and its potential usefulness for diagnosis is discussed.

Following establishment of chronic guinea pig cytomegalovirus (GPCMV) infection, animals received by gavage either cyclosporine (CS), 20–33 mg/kg, or drug vehicle daily for 28 days. At sacrifice the frequency of CMV isolation from tissues of GPCMV-infected animals was 20% for the immunosuppressed group versus 12% for controls (difference, NS). GPCMV isolation rates from lymphoid tissues (thymus, spleen, cervical lymph nodes) of experimental animals was 7 of 69 (10%) vs. 0 of 42 (0%) from controls (p < 0.05). Although CS immunosuppression was associated with increased rates of GPCMV isolation from lymphoid tissues of chronically infected animals, it was not accompanied by detectable systemic diseas

The genome of Herpesvirus sylvilagus, a gamma herpesvirus capable of inducing a lymphoproliferative disease in cottontail rabbits, was analyzed with respect to its infectivity, density in cesium chloride isopynic gradients, and molecular weight. The buoyant density of virus DNA in cesium chloride was 1.710 g/cm3. Sucrose gradient centrifugation showed virus DNA to be 86.08 megadaltons (129.25 kilobase pairs). The restriction endonucleases EcoRl, Xhol, Sstll, and BglI cleaved the virus DNA into 8,7,23, and 13 major fragments ranging from 38.32 to 0.53 kilobase pairs, respectively. The size of the virus DNA as determined from the restriction endonuclease analysis was 72.59 megadaltons (108.89 kilobase pairs). The average size of H. sylvilagus DNA, determined from electron microscopic observations, was 78.06 megadaltons (117.08 kilobase pairs).
