Using Southern-Western and zinc-blotting techniques, the interactions of single-stranded DNA and zinc with structural proteins of type B, C, and D retroviruses were examined. Besides nucleic capsid proteins of retroviruses known to form complexes with nucleic acids, some other core proteins of type C retroviruses were shown to bind nucleic acids. All nucleic capsid proteins of the examined retroviruses appeared to be also zinc binding. In the present report, we propose two protein-blotting techniques that could be performed on a single nitrocellulose membrane. A first technique allows to detect zinc- and DNA-binding proteins immobilized on the membrane; a second (a modification of Southern-Western blotting) makes it possible to detect DNA-binding proteins followed by immunological re-probing.

摘要:
Bovine adenovirus type 3 (BAV3) is a DNA virus that causes respiratory and gastrointestinal disorders in cattle. The viral genome consists of a linear double-stranded DNA molecule (35,000 base pairs) with inverted terminal repeats at each of its 5’ molecular ends. We have subcloned 10 Hindlll fragments spanning 4.9–96.0%, 5 EcoRl fragments spanning 3.4–89.5% and 2 Xbal fragments spanning 35.7–82.9% of the BAV3 (strain WBR-1) genome into the bacterial cloning vector pUC19. The subcloning of the viral genome facilitated the construction of linear restriction enzyme maps for BamHI, Clal, EcoRl, Hindlll, Kpnl, Notl, NspV, Pstl, Pvul, Sall, Xbal and Xhol. 摘要:
Data on 537 Arboviruses and 180 other viruses have been collected and coded in two different formats. These data include information not only regarding the taxonomy and history of isolation, but also regarding the properties of biomacromolecules, proteins and nucleic acids. Information on antigenic relationships, histopathology and experimental viremia is also included. This information is stored in formats which allow the manipulation and analysis of data by dBASE III PLUS and MICRO-IS. A set of programs was written for interconversion and editing purposes. Transmission electron micrographs are scanned and stored. This stored information can be used in viral classification as shown by carrying out analysis of data on the Bunyaviridae family.

摘要:
Two closely related viruses were isolated from the blood of bovines near Darwin, Northern Territory, Australia. When studies of virus morphology indicated that these were rhabdo-viruses, serologic studies were done. These isolates are closely related or identical and are related to, but distinct from, the rabies-related kotonkan virus. Other serologic studies showed that these are two isolates of a newly recognized virus, for which the name Koolpinyah virus is proposed.

A 980-bp subfragment of human cytomegalovirus (HCMV) strain Towne has been previously identified as morphologic transforming region II (mtrll) because of its ability to induce focal transformation of NIH 3T3 cells. Transcripts from this region, which could encode the three open reading frames (ORFs), 79, 83, and 34 amino acids (aa), detected by DNA sequence analysis, are expressed early during HCMV infection. In this report, the mRNA start sites for promoters (PI and P2) were mapped within Towne mtrll by primer extension using RNAs isolated from transformed NIH 3T3 cells. The Towne mtrll promoters exhibited similar activities to the SV40 enhancerless early promoter. Equivalent promoter activities were detected within the mtrll colinear nontransforming region from HCMV strain Tanaka. Two subclones of Towne mtrll (5’ 440-bp and 3’ 540-bp), each containing one promoter, were generated utilizing a unique BgII site which also interrupted the 79-aa ORF. In transfection assays, neither the 5’ 440-bp promoter subclone containing a truncated 79-aa ORF nor the 3’ 540-bp subclone containing intact 83- and 34-aa ORFs exhibited transforming activity. These data indicated that transformation by HCMV mtrll did not occur by promoter insertion. The protective efficacy of Candid No. 1, a live-attenuated vaccine against Argentine hemorrhagic fever (AHF), was evaluated in non-human primates. Twenty rhesus macaques immunized 3 months previously with graded doses of Candid No. 1 (16–127, 000 PFU), as well as 4 placebo-inoculated controls, were challenged with 4.41 log10 PFU of virulent P3790 strain Junin virus. All controls developed severe clinical disease; 3 of 4 died. In contrast, all vaccinated animals were fully protected; none developed any signs of AHF during a 105-day follow-up period. Viremia and virus shedding were readily detected in all placebo-vaccinated controls, while virus could be recovered only once (by amplification) from throat swabs of 2 Candid No. 1 vaccinees on day 21. Vigorous secondary-type neutralizing and immunofluorescent antibody responses were seen in most vaccinees that had received 3 logι0 PFU Candid No. 1 or fewer; all others, including those receiving 127,200 PFU, maintained relatively stable titers during follow-up. 摘要:
To determine whether antigenic differences occur among rubella virus strains, five wild-type strains of rubella virus isolated in the UK, the USA, and in Japan between 1964 and 1987 and four attenuated vaccine strains were compared employing a panel of 28 monoclonal antibodies in neutralization, haemag-glutination-inhibition, enzyme immunoassay, and indirect immunofluorescence assays. No antigenic differences were detected which confirms that rubella vaccines will protect against circulating strains and that rubella antigens used in serological tests for screening and diagnosis will detect antibodies induced by all strains.
